Abstract
1,154,505. Derivatives of morphine. RECKITT & SONS Ltd. 4 Dec., 1967 [15 Dec., 1966], No. 56284/66. Heading C2C. Novel morphine derivatives of formula and acid addition salts thereof, wherein Y is ethano or etheno, R is H or methyl, R<SP>1</SP> is H, CN, C 1-8 alkyl, C 2-8 alkenyl, C 2-8 alkynyl or methyl substituted by a C 3-6 alicyclic ring, R<SP>2</SP> is methyl or aryl optionally substituted and R<SP>3</SP> is H, aryl optionally substituted or is C 1-6 alkyl optionally substituted at carbon atom 1, 2 or 3 by an alicyclic or aryl group, are prepared by dehydration of a compound of formula followed in the cases where R is H and/or R<SP>1</SP> is CN by mild alkaline hydrolysis, or for compounds where R<SP>1</SP> is H either by treatment of the corresponding compound R<SP>1</SP> = CN with alkali at 140-170‹ C. or by treatment of a compound of Formula III with hot formic acid, or for compounds where R<SP>1</SP> is CN by heating the corresponding compound R<SP>1</SP> = methyl with cyanogen bromide and for compounds where R<SP>1</SP> is other than H or CN treating the compound R<SP>1</SP> = H with the appropriate halide or in special cases by reaction of a ketone of formula with an ylide of formula Ph 3 -P = CH 2 . Compounds of Formula III above are prepared by reacting compounds of the second general formula above with dialkyl azodicarboxylate. Pharmaceutical compositions having action on the central nervous system and in conventional forms comprise an above novel compound and a carrier therefor.
