An antibiotic effective against pneumonialike organisms in poultry, methobottromycin, has the following properties: m.pt. 166- 167‹ C.; soluble in alcohol, esters, ethers, chlorinated solvents and benzene; sparingly soluble in water and insoluble in hexane and petroleum ether; [α]<SP>20</SP>D=- 15 (95% alcohol); infra-red absorption spectrum as in the accompanying drawing; elementary analysis C= 59À50%, H= 7À52%, N= 13À50%, S= 3.90% and O=15À58% (possibly C 41 H 62 N 8 O 7 S); Rf is 0À19 in capryl alcohol saturated with 0À1 M phosphate buffer of pH 6À0; hydrolysis with acid yields substances giving a positive ninhydrin test but no proline; forms salts with acids, e.g. hydrochloric, tartaric and salicylic acids. Methobottromycin is produced by cultivating Streptomyces canadensis ATCC 17776, or a methobottromycin-producing mutant thereof, in an aqueous nutrient medium containing assimilable sources of C and N. Methobottromycin is recovered from a clarified fermentation broth by treating the broth with a strongly acidic cationic ion-exchange resin, eluting the resin with aqueous methanolic ammonia, neutralizing the eluate, extracting it with chloroform, performing partition chromatography on the chloroform extracts using chloroform and acetone as eluant, precipitating antibiotics of the methobottromycin class by acidification and isolating methobottromycin itself by partition chromatography using 0À1 1 M phosphate buffer as a developing solution and diatomaceous earth impregnated with capryl alcohol as stationary phase. Methobottromycin is coproduced with amethobottromycin, Specification 1,150,476. Methobottromycin, or a salt thereof, is administered orally or subcutaneously to poultry infected with pleuropneumonia-like organisms.
