Abstract
1,131,847. Cyclic and non-cyclic amidines. CHAS. PFIZER & CO. Inc. 18 Nov., 1966 [30 Dec., 1965; 1 Aug., 1966], No. 51854/66. Heading C2C. [Also in Division A5] Novel compounds of formulµ and non-toxic acid addition salts thereof, wherein R 1 is H, methyl or halogen, Z is ethylene or trimethylene, R 2 is H or methyl, R 3 is H and R 4 is methyl or R 3 and R 4 taken together are ethylene or trimethylene and X is ethylene or vinyline are prepared by reacting a compound of Formula III wherein Y is the residue of a phenyl ring in which case R is R 1 and X is vinyline, or Y is oxygen in which case R is R 2 and X is ethylene or vinyline, and W is cyano or where R<SP>1</SP> is alkyl or if X is vinyline W may be with an amine of formula (CH 3 ) 2 NH or H 2 N-Z-NH-CH 3 or a salt thereof or by reacting a compound of Formula III above in which -X-W is -CHO with an amine of formula and when required forming an acid addition salt of any product. The vinylene compounds may be isolated in the cis or trans forms. Ethyl 2 - furanpropionimidate hydrochloride and the 3-methyl-2-furan compound are prepared by alcoholysis of the corresponding propionitriles which have been obtained by hydrogenation of the corresponding acrylonitriles. 3 - Methyl - 2 - furanacrylonitrile is prepared by action of cyanoacetic acid and ammonium acetate on 3-methyl-2-furaldehyde. 3 - (3 - Methyl - 2 - furanacrylimidoyloxy) - propanesulohonic acid and the 3-normethyl compound are prepared by action of 1,3-propane sultone on 3-methyl-2-furanacrylamide or 2- furanacrylamide; the amides being prepared from the corresponding carboxylic acids which are themselves prepared by condensation of malonic acid and the appropriate furaldehyde.
