Abstract

Novel compounds of the general formula <FORM:1123413/C2/1> in which Z1 represents a primary amino group or an amino group substituted with one or two alkyl groups or with one acyl group, an hydroxyl group, an esterified hydroxyl group, or an etherified hydroxyl group, the ether group comprising an aliphatic radical containing from 2-6 carbon atoms or a halogen atom of atomic weight greater than 19, salts thereof are obtained by the cylization of an ester of the acid of the Formula III or a compound of Formula VII <FORM:1123413/C2/2> reduction of the olefinic bond, the nitro group and where present the cyclic amide group of the resulting cyclized product followed, if desired, by the conversion of the resulting 10-amino group into another group. Cyclization may be effected with phosphorus oxychloride and reduction with lithium aluminium hydride. Methylation of 2,3-dimethoxy-10-aminoberbine may be effected with formaldehyde and conversion to a 10-hydroxy compound may be effected by standard procedures. A 10-halogen atom may be introduced by treating the 10-amino compound with cuprous chloride. Pharmaceutical compositions having tranquillizing and analgesic properties containing the above compounds are administered orally, rectally or parenterally.
