Abstract

Novel hexahydroindenopyridine derivatives of the formula <FORM:1117806/C2/1> wherein R is a C1- 6 alkyl or C3- 9 cycloalkyl radical and wherein the benzene ring of the indenopyridine nucleus and/or the phenyl radical at the 4a-position may bear one or more substituents and acid addition salts or quaternary ammonium salts thereof are prepared by (a) refluxing the appropriate 3-benzoyl-4-phenyl - 1,2,3,6 - tetrahydropyridines with anhydrous hydrazine, removing excess hydrazine and decomposing the hydrazine derivative produced and cyclising the 3-benzyl-4-phenyl-1,2,5,6-tetrahydropyridines produced with polyphosphoric acid at an elevated temperature not exceeding 150 DEG C. or (b) by reducing the corresponding 9-oxo-hexahydroindenopyridines. The compounds may be separated into two racemates, e.g. by means of their picrate salts. Therapeutic compositions useful as central-nervous-system depressants and as analgesic, anticholinergic and antihistaminic agents contain the above hexahydroindenopyridine derivatives or pharmaceutically acceptable acid addition salts or quaternary ammonium salts thereof as active ingredients. The composition may be in the form of tablets, suspensions, solutions or suppositories.
