Abstract
Novel 3,8,9 - trihydro- and 1,2,3,8,9 - pentahydro - 3aH - benzo[6,7]cyclohepta[1,2,3-de]-isoquinolines of the general formulae <FORM:1113905/C2/1> and <FORM:1113905/C2/2> wherein R1 is hydrogen or C1-C3 alkyl, an pharmaceutically acceptable acid addition salts thereof are prepared by reduction of 10,11-dihydro - 5H - dibenzo [a,d]cyclohepten - 5 - yl nitrile to 10,11 - dihydro - 5H - dibenzo[a,d] cyclohepten-5-yl-methylamine, reaction of the latter compound with an acylating agent of the formula R1COX, wherein X is the hydroxyl group, a group -OCOR2 in which R2 is C1-C3 alkyl, a halogen atom or a group -OR2, and cyclization of the resulting acylamino compound of the formula <FORM:1113905/C2/3> followed by optional reduction of the resulting 3,8,9 - trihydro - 3aH - benzo[6,7]cyclohepta [1,2,3-de]-isoquinoline to give the corresponding 1,2,3,8,9 - pentahydro - 3aH - benzo[6,7]cyclohepta [1,2,3-de]-isoquinoline and optional conversion to pharmaceutically acceptable acid addition salts of the isoquinolines of the first or second formulae above. Pharmaceutical compositions, which are useful as antibacterial and trichomonicidal agents and as central nervous system depressants, comprise as active ingredient an isoquinoline of the first or second formula above or a pharmaceutically acceptable acid addition salt thereof, in association with a pharmaceutical carrier.
