Abstract

Compressed tablets of pharmaceutical materials which are low melting or adhesive in nature and are not readily compressed by standard procedures, may be moulded by compression under a low compression force e.g. of 2 to 8 tons per sq. inch, by a punch within a die cavity which is subjected to a cleaning operation by the alternating sequential production therein of a tablet from an abrasive material under a high compression force e.g. 15 to 25 tons per sq. in. Preferably the low compression force is achieved with a punch penetration into the cavity of up to 1/8 in., and the high compression force with a penetration of at least 3/8 in. Preferably the process is operated on a multiple cavity rotary tablet press provided with a hopper for each of the materials and whereby one tablet of each material is produced from each die for each complete revolution of the head. The abrasive material is preferably one or more of lactose, wood, cellulose, magnesium hydroxide, tricalcium phosphate, and dicalcium phosphate. The low melting material may be anhydrose lactose with stated combinations and proportions of wood cellulose, magnesium stearate and magnesium hydroxide or may be tricalcium phosphate with lactose, starch, acacia and magnesium stearate. A suitable granulation comprises, in weight proportions, at least 50% of low melting material, 10-30% filler, 5 to 20% disintegrating agent, 2-15% binding agent and 1-5% lubricant. In a preferred tybamate granulation formulation, 67 parts thereof were coated with 3.5 parts gelatin followed by admixture with 13 parts wood cellulose and further coating with 1.5% of starch from a 5% concentration paste; the granules thus obtained were dried and admixed with 3 parts wood cellulose, 10.5 parts starch and 1.5 parts stearic acid. Suitable suppository granulations may contain a bismuth salt, benzocain and meprobamate with polyethylene glycol and a filler; sodium bicarbonate and sodium biophosphate may be contained in efferescent laxative suppository compositions.
