Abstract

A composition comprising a standard amount of cholesterol and cholesterol esters of fatty acids ("total cholesterol"), lyophilised human blood plasma and a solubilizing compound RO(CH2CH2O)nH where R is a straight or branched chain alkyl group having 12-18 carbon atoms, an alkyl phenoxy group having 1-9 carbon atoms in the alkyl group or an acyl group having 12-22 carbon atoms and n=10-20 may be used as a standard in determining total cholesterol in blood. The composition has an optical density of less than 0.450 at 620 mm when reconstituted. The only cholesterol ester specified is the acetate: specified solubilizing compounds and compositions are (a) oleyl alcohol/ethylene oxide condensate (if desired together with a polyoxyethylene-polyoxypropylene condensate), (b) a mixture of nonylphenoxy/ and stearyl/ethylene oxide condensates, and (c) a mixture of nonylphenoxy/and oleyl/ethylene oxide condensates. The composition may be prepared, if desired, immediately before use by adding the total cholesterol components to processed blood plasma together with the solubilizing agent mixed with hot water, or the cholesterol may be first solubilized. The Specification describes a test for cholesterol in blood plasma comprising adding alcoholic KOH; extracting with petroleum ether and determining the optical density after adding Liebermann-Burchard reagent.
