Abstract

A micro-organism, particularly yeast, capable of growth in the presence of a hydrocarbon feedstock consisting wholly or in part of straight chain hydrocarbons in a growth stage is cultivated in the presence of the feedstock, an aqueous nutrient medium and a gas containing free oxygen and subsequently, if necessary, the micro-organism is separated from the bulk of the unconsumed hydrocarbon. The content of straight-chain hydrocarbon in the feedstock is thereby reduced with an improvement in the pour point.ALSO:A micro-organism, particularly yeast, capable of growth in the presence of a hydrocarbon feedstock consisting wholly or in part of straight chain hydrocarbons, in a growth stage is cultivated in the presence of the feedstock, an aqueous nutrient medium and a gas containing free oxygen and subsequently, if necessary, the micro-organism is separated from the bulk of the uncomsumed hydrocarbon whereby there is obtained a micro-organism free of a substantial amount of hydrocarbon but containing a small amount of hydrocarbon as contaminant, and thereafter in a purification stage the micro-organism is maintained with an aqueous nutrient medium and a gas containing free oxygen, whereby the hydrocarbon contaminating the micro-organism is reduced in quantity or is eliminated. The growth stage and purification stage may be carried out continuously in separate fermenters or in different zones of the same fermenter. After the growth stage, the major part of the continuous aqueous phase is first separated by decanting and centrifuging. About 80% of the aqueous phase recovered is returned to the fermenter together with make-up quantities of the necessary nutrients. The micro-organism "cream" also obtained by the separation is then mixed with an aqeuous nutrient medium, which may be the same as or different from the aqueous nutrient medium employed in the growth stage, and is maintained in admixture with a gas containing free oxygen whereby the hydrocarbon contaminating the micro-organism is reduced in quantity or is eliminated. Again, the major part of the continuous aqueous phase is separated with recycle of a proportion of the recovered aqueous phase either to the growth stage or to the purification stage. The micro-organism "cream" obtained by this second separation may be further treated for the removal of traces of oil held to the cells, as by washing with an aqueous solution of a surfactant and thereafter with water. Examples are directed to the growth on heavy-gas oil of Candida lipolytica from an inoculum on mixed C9-C16 normal hydrocarbons, and to a subsequent purification of the yeast under the same culture conditions except for the lack of the heavy-gas oil. The purified yeast was recovered by centrifuging and washing at +5 DEG C, and was then freeze-dried. The dried yeast exhibited a substantially reduced lipid content.
