Abstract
1461408 Lipase BRITISH PETROLEUM CO Ltd 10 May 1974 [25 May 1973] 25132/73 Heading C3H A fermentation process for the production of lipase involves cultivating a lipase-producing micro-organism in a broth comprising a microbial lipid as the carbon source typically in a proportion of 1 to 25 g./l., an aqueous nutrient medium and a gas containing free oxygen and thereafter recovering a broth fraction containing lipase. Lipase in solid form may be isolated from the broth by employing conventional techniques. Lipase-producting micro-organisms: species of bacteria, yeast and fungi (specified) but especially Candida lipolytica CBS No. 6331. The microbial lipid is preferably that obtained by extracting with isopropanol the biomass produced by the cultivation of the hydrocarbonutilizing yeast Candida tropicalis CBS No. 6373 on a gas oil boiling range hydrocarbon as the carbon substrate. Phospholipids are removed from the microbial lipid by solvent treatment before use. The yield of lipase is enhanced by including in the broth (1) a dispersant for the lipid, e.g. 0À3-3À0 parts by weight per part by weight of the microbial lipid, of a petroleum fraction which contains n-alkanes and has a boiling range of 250-380‹ C., and (2) non-ionic surface active agent, e.g. 0À05-1À0 part by weight (based on microbial lipid) of either a fatty acid ester of a polyalkylene oxide sorbitol condensate or sorbitan mono fatty acid ester. The fermentation is carried out under mild conditions of agitation with aeration rates in the range 30 to 60 vol./vol./hr. An antifoaming agent (specified) may be included in the broth.
