Abstract
The invention comprises compounds of formula <FORM:1036278/C2/1> wherein R1 is a phenyl group, R2 is a group -COR4, wherein R4 is a hydrogen atom or an alkyl, aralkyl, alkoxy, dialkylaminoalkoxy, amino or hydroxyalkyl group, and R3 is a hydrogen atom or an alkanoyl group, or R3 is a R3 in combination form a group <FORM:1036278/C2/2> wherein R5 and R6 are alkyl radicals and Y is a carbonyl or methylene group, and acid addition salts of compounds wherein R3 is a hydrogen atom, and their preparation by (a) reaction of a compound of the above formula, wherein R2 is a hydrogen atom with a reactive functional derivative of a carboxylic acid or with an alkali metal cyanate, (b) reduction of a carbonyl group Y to a methylene group, (c) tranesterifying an alkyl - N - (4 - hydroxy - 4 - phenyl - 1 - piperidine) carbamate with a dialkylaminoalcohol, or (d) treatment of a succinimide with lithium aluminium hydride to form a 4-hydroxy-butyramide. The compounds of the invention have antidepressant activity and may be used in pharmaceutical compositions in combination with a pharmaceutical carrier, for example in forms suitable for oral, parenteral or rectal administration, such as tablets, capsules, solutions and suppositories.
