Abstract

A disaggregated human gamma globulin, freed from substances affecting the complement system of human serum and having a sedimentation constant S within the range of from 3.0 to 5.5, is prepared by treating a gamma globulin fraction of human serum, in an aqueous medium at a pH of from 1.5 to 5.5 and at a temperature of from 0 DEG to 50 DEG C., with pepsin until no anti-complement activity (in respect of human serum) is detectable, removing low molecular weight impurities from the disaggregated gamma globulin so obtained, filtering the product under sterile conditions, and, if desired, lyophilising the product so obtained. The removal of the low molecular weight impurities may be achieved, for example, by fractional precipitation with salts or organic solvents or by ultrafiltration. The initial gamma globulin fraction may be one obtained by precipitation from human serum with ammonium sulphate or alcohol.
