Abstract
Aggregations of viruses in suspension are separated into single viruses and also purified by subjecting an aqueous suspension of the virus to chromatography at a speed of flow of 0.001 to 0.400 cc per minute per cm2 in a calcium phosphate adsorption column, which has been filled at a pressure of 500-600 m.m. of Hg with a suspension of baHP04 2H2O in a phosphate buffer having a pH value of 6.5 to 7.5 and a molar concentration of about 0.001 m, the ratio of the diameter of the column to its length being 1 : 10 to 1 : 20 and the concentration of the virus suspension being about 4000-6000 gamma N. per cc, washing the column with a phosphate buffer having a pH of 6.5 to 7.5 and a molar concentration about 0.001 m. and electing the adsorbed viruses from the column at a speed of flow of 0.001-0.400 cc per minute per cm2 with a phosphate buffer having a pH value of 6.5 to 7.5 and a molar concentration of 0.1 to 1.0 m. The virus may be further purified by treatment with an anion exchange cellulose derivative. Typical viruses are poliomyelitis and influenza virus.
