Abstract

The invention comprises substituted butenones of the formula <FORM:1009909/C2/1> wherein R1 and R2, the same or different, represent carbocyclic or heterocyclic radicals and Am represents the residue of a secondary amine and is linked through the amino nitrogen atom to the remainder of the molecule, or a quaternary derivative or a pharmaceutically acceptable salt thereof, and the preparation thereof by isomerizing, in acid medium, the corresponding tertiary acetylenic alcohol having the general formula <FORM:1009909/C2/2> Tertiary acetylenic alcohols of the Formula II are prepared by reaction of 2,3-dibromo-1-propene with a secondary amine of the formula AmH, to give the corresponding 3-tertiary amino-1-propene which is converted to the corresponding 3 - tertiary amino - 1 - propyne which is reacted with a ketone of the formula <FORM:1009909/C2/3> or by reaction of acetylene with a ketone of the Formula III to give the corresponding acetylenic carbinol which is reacted with formaldehyde or other methylenic group donor and a secondary amine of the formula AmH. 4 - Morpholino - 1,1 - diphenyl - 1 - buten - 3 - one gives 4 - morpholino - 1,1 - diphenyl - 3 - butanone, 4 - morpholino - 1,1 - diphenyl - 1 - buten - 3 - ol and 4 - morpholino - 1,1 - diphenyl-3-butanol on reduction with hydrogen and Raney nickel, lithium, aluminium hydride, and hydrogen and Raney nickel with sodium hydroxide respectively.ALSO:Therapeutic compositions with anaesthetic properties; in the form of pomades, ointments and lotions for topical application and solutions for parenteral administration, comprise compounds of formula <FORM:1009909/A5-A6/1> wherein R1 and R2, the same or different, represent carbocyclic or heterocyclic radicals and Am represent the residue of a secondary amine and is linked through the amino nitrogen atom to the remainder of the molecule, and quaternary derivatives and pharmaceutically acceptable salts thereof, together with suitable pharmaceutical excipients. The compounds of the formula may be optionally used in conjunction with, for example, noradrenalin.
