COMPOSITIONS COSMETIQUES COMPRENANT UNE ASSOCIATION D’INGRÉDIENTS ET LEURS UTILISATIONSCOSMETIC COMPOSITIONS COMPRISING A COMBINATION OF INGREDIENTS AND THEIR USES
The present invention relates to the field of cosmetics and more particularly to a new composition comprising a combination of ingredients making it possible to maintain and stimulate the hydration of the skin.
The skin isolates the interior of the body from the environment and plays an important role in the maintenance of homeostasis. Its protective function depends on the integrity and hydration state of the stratum corneum.
The presence of water in the stratum corneum is of paramount importance to preserve its physical properties of flexibility and elasticity. Water acts as a plasticizer in association with proteins and soluble materials (BLANK, 1952, 1976).
The hydration state of the stratum corneum varies according to the following factors: quantity of water, water transported from the lower layers, speed of evaporation, speed of keratinization, quantity and composition of the epicutaneous emulsion.
La capacité de rétention d'eau du stratum comeum dépend principalement de la présence du Natural Moisturizing Factor (facteur hydratant naturel) - NMF, d'un ensemble de substances hygroscopiques et de la présence de lipides qui rendent la couche cornée imperméable à l'eau (SPENCER, 1988).The water retention capacity of the stratum comeum mainly depends on the presence of the Natural Moisturizing Factor (NMF), a set of hygroscopic substances and the presence of lipids which make the stratum corneum waterproof. (SPENCER, 1988).
An increase in hydration can be obtained with the use of topical products, which act by two main mechanisms:
The contribution of hydrating active ingredients and the limitation of insensitive water losses are the two main axes to maintain and reinforce this cutaneous barrier and consequently the good general state of the skin. New moisturizing and / or active ingredients capable of reducing insensitive water losses are therefore of great cosmetic interest.
To date, there are many moisturizing active ingredients on the market. Mention may in particular be made of hydrating active agents which reinforce the barrier function and reduce insensitive water losses and / or those which increase the water content of the skin and / or stimulate the synthesis of aquaporin to improve the circulation of water in cells. By way of nonlimiting example, mention may be made of moisturizing active agents which are classified into several categories such as:
- les agents humectants qui absorbent l’eau ou aident à retenir l’humidité au niveau cutané par leur hygroscopicité dont notamment, les acides aminés libres et certains peptides, l’acide pyrrolidone carboxylique ou acide pyroglutamique et ses sels (zinc et sodium PCA), le lactate de sodium, l'urée, les sucres simples notamment les pentoses et hexoses, les polyols dont notamment le glycérol et ses dérivés, le sorbitol, les glycols et les diols, ainsi que les acides alpha hydroxylés AHA ;- humectants that absorb water or help retain moisture in the skin through their hygroscopicity including, in particular, free amino acids and certain peptides, pyrrolidone carboxylic acid or pyroglutamic acid and its salts (zinc and sodium PCA) , sodium lactate, urea, simple sugars, in particular pentoses and hexoses, polyols, in particular glycerol and its derivatives, sorbitol, glycols and diols, as well as alpha hydroxy acids AHA;
- les agents filmogènes qui forment avec l’eau des hydrogels semi perméables dont notamment des protéines comme le collagène, l’élastine, des polysaccharides tels que la pectine, le chitosan, ou des sucres complexes comme les glycosaminoglycanes notamment l’acide hyaluronique ;- film-forming agents which form semi-permeable hydrogels with water, including in particular proteins such as collagen, elastin, polysaccharides such as pectin, chitosan, or complex sugars such as glycosaminoglycans, in particular hyaluronic acid;
However, the existing compositions currently on the market only have a moderate hydrating power and often of short duration. There is therefore a need to be able to obtain high hydration of the skin and preferably over a long period, that is to say for at least 6 hours, even 12 hours.
However, surprisingly, the inventors discovered that there was a synergy on the hydration of the skin by associating an effective amount of a hydro-alcoholic extract of the envelope of the seed of Chenopodium Quinoa Willd, also called pericarp Quinoa seed, and a skin moisturizing ingredient.
Quinoa (Chenopodium Quinoa) is a species of annual herb from the Chenopodiaceae family. It is a pseudo-cereal, rather than a real cereal, because it is not a herb. This traditional plant has been cultivated for over 5000 years on the highlands of South America. Like the bean, the potato, the corn, the Quinoa was at the base of the food of the pre-Columbian civilizations, but, contrary to these last, it did not hold the attention of the Spanish conquerors because of the content in saponin from the husk of its unpeeled seeds, and from the fact that the flour obtained from it is not breadable due to the absence of gluten.
In the 1970s, industrialized countries in search of healthier food discovered the nutritional qualities of Quinoa, which is now distributed in most supermarkets, especially in stores selling organic and fair-trade products.
There are mainly two main families of Quinoa: bitter Quinoa and sweet Quinoa. The first, whose grain is yellow, traditionally cultivated in the Andes for more than 5000 years requires washing and scarification of the grains because of the saponin content of the pericarp (bitter and having a certain level of toxicity). It is the variety mainly exported to the West through fair trade. Mild Quinoa, whose grain is beige in color, from more recent varietal selections, contains little or no saponins, about 0.2% saponins. The Quinoa seed is a dry, indehiscent, single-seeded fruit whose pericarp, more or less sclerified, is not welded to the seed.
The plaintiff has filed a request for access to the genetic resource Chenopodium Quinoa with the Peruvian government (MINAM: Peruvian Ministry of the Environment) in accordance with the Andean law, Decision No. 391, on the common regime for access to genetic resources.
In the context of the present invention, the Quinoa (Chenopodium Quinoa Willd) used is chosen from the varieties of yellow Quinoa Sacaca and Marangani, coming from Peru and in particular from the regions of Cusco, Ayacucho and Arequipa. These varieties are considered the most bitter on the market by their high saponin content, up to 7% in the pericarp of the seed. In order to be used in human food, it is necessary to scarify the seeds and then wash them. During this scarification by external abrasion of the seed, the Quinoa pericarp is eliminated. The pericarp, very rich in saponins, is considered to be a waste from the food industry, hence the possibility of co-recycling this waste by eco-extraction.
On connaît du document de Yawadio Nsimba et al. (Antioxidant activity of various extracts and fractions of Chenopodium quiona and Amaranthus spp. Seeds, ScienceDirect, Food Chemstry 106 (2008) 760-766) que de nombreuses saponines provenant notamment du Quinoa (Japon (JQ) et Bolivie (BQ)) présentent une potentielle activité anti-oxydante.We know from the document by Yawadio Nsimba et al. (Antioxidant activity of various extracts and fractions of Chenopodium quiona and Amaranthus spp. Seeds, ScienceDirect, Food Chemstry 106 (2008) 760-766) that many saponins, notably from Quinoa (Japan (JQ) and Bolivia (BQ))) potential antioxidant activity.
On connaît également du document de Ribaud et al.( L’Oréal, A new desquamating agent respectful of epidermal homeostasis as a complementary appoach to traditional peeling procédures, 2016) que des saponines extraites du péricarpe de la graine du Quinoa Real présentent une activité cosmétique desquamante sur la peau.It is also known from the document by Ribaud et al. (L'Oréal, A new desquamating agent respectful of epidermal homeostasis as a complementary appoach to traditional peeling procedures, 2016) that saponins extracted from the pericarp of the Quinoa Real seed have cosmetic activity peeling on the skin.
We also know from another L'Oréal document (QHE-IFFCC, November 2016) that an extract of the pericarp from the Quinoa Real seed contains three categories of compounds (sugars, flavonoids and saponins) which each have a well-defined biological activity on the skin. Thus, the saponins are shown to exhibit a desquamating activity on the skin, the polyphenols and flavonoids an activity for protecting the skin and the oligosaccharides, minerals and amino acids a hydrating activity. The process described for extracting the extract consists in eco-extracting the Quinoa pericarp with a suitable solvent.
However, the Applicant has demonstrated that a new hydro-alcoholic extract of the pericarp of the seed of Chenopodium Quinoa Willd, in particular of the varieties of Quinoa yellow Sacaca and Quinoa yellow Marangani, stimulates the biological effect of other moisturizing ingredients. the skin. The extract rich in saponins, without presenting any hydrating activity by itself, stimulates the activity of moisturizing ingredients on the skin.
Thus, the Applicant has surprisingly discovered that the combination of an effective amount of a hydro-alcoholic extract of the pericarp of the seed of Chenopodium Quinoa Willd and a skin moisturizing ingredient synergistically increases the water content. skin. Therefore, the combination of these ingredients has the advantage of providing a high moisturizing effect by increasing the water content.
The present invention therefore has for first object a hydro-alcoholic extract of the pericarp of the seed of Chenopodium Quinoa Willd mainly comprising saponin-type compounds to stimulate the biological effect of a skin moisturizing ingredient.
A second object according to the invention is a cosmetic composition comprising in a physiologically acceptable excipient, as active agent, the combination of an effective amount of a hydro-alcoholic extract of the pericarp of the seed of Chenopodium Quinoa Willd and of a skin moisturizing ingredient.
A third object according to the invention consists in the use of a cosmetic composition according to the invention to maintain and stimulate the hydration of the skin.
Finally, a fourth object according to the invention is a cosmetic treatment method for maintaining and stimulating hydration of the skin, comprising the topical application of a composition according to the invention.
The invention and the advantages which ensue therefrom will be better understood on reading the description and the nonlimiting embodiments which follow, written with reference to the appended figures in which:

The invention relates to a hydro-alcoholic extract of the pericarp of the seed of Chenopodium Quinoa Willd mainly comprising saponin-type compounds to stimulate the biological effect of a skin moisturizing ingredient.
The hydro-alcoholic extract of the pericarp from the seed of Chenopodium Quinoa Willd is preferably produced by solid-liquid extraction of the previously dried pericarp from Quinoa seeds in a solvent chosen from the group consisting of water in the presence of mono, di or tri alcohols such as ethanol, and diols including glycols, or glycerol, all agro-sourced solvents.
In a preferred embodiment, the extraction solvent is a water / 1,3-propanediol mixture. More particularly, within the framework of the solid-liquid extraction of the dry quinoa pericarp, between 0.1 and 20% by weight (dry matter) of epicarp is introduced into the extraction solvent, and preferably between 1 and 15% by weight, typically between 5 and 10% by weight (the% being expressed by weight of the dry matter relative to the total weight used). The pericarps are ground or cryo-ground using a grinder such as a knife crusher. In particular, in the presence of diol and more particularly of 1,3-propanediol, a proportion of between 0 and 90% of 1,3-propanediol in water is chosen, preferably between 10 and 80% of 1,3-propanediol , and advantageously between 35 and 45% (the% are expressed by weight of propanediol relative to the total weight of water + propanediol).
The extraction temperature is advantageously between 4 ° C and 100 ° C, and preferably between 10 ° C and 80 ° C, and more particularly between 20 ° C and 50 ° C. Heating can be provided by circulation of a thermal fluid in a double jacket or by microwave.
The extraction time advantageously varies from 10 minutes to 5 hours, and preferably from 30 minutes to 4 hours, and more advantageously from 2 to 3 hours.
The extraction can advantageously be carried out under vacuum, at atmospheric pressure or under pressure, preferably at atmospheric pressure or under pressure, and advantageously at atmospheric pressure.
The extraction is always carried out with stirring, thus allowing dispersion and homogenization of the solid in the liquid, effectively improving the diffusion of the solute in the solvent.
After the extraction, the plant material, residual and exhausted in compounds of interest, is advantageously separated from the liquid phase, for example by decantation, by centrifugation or by clarifying filtration. The liquid phase obtained can be microfiltered using filter media of suitable porosity in order to obtain a clear and bloodless solution of solid plant particles.
These first liquid-solid separation steps may be followed by purification steps, for example by ultrafiltration and / or nanofiltration, making it possible to concentrate the compounds of interest at the expense of other compounds such as traces of proteins, of sugars and minerals, as well as the solvent or mixture of solvents.
The extract obtained may be in liquid form but also in solid form after a drying step according to methods known to those skilled in the art such as atomization, lyophilization or zeodratation for example, with or without support such as maltodextrin.
Typiquement, le procédé de préparation de l’extrait hydro-alcoolique du péricarpe de la graine de Chenopodium Quinoa Willd comprend les étapes successives suivantes :Typically, the process for preparing the hydro-alcoholic extract of the pericarp of the seed of Chenopodium Quinoa Willd comprises the following successive stages:
(a) dispersion et extraction dans un solvant approprié d’épicarpes de graines de Quinoa et avantageusement des épicarpes cryo-broyés, le solvant étant avantageusement un solvant eau/l,3-propanediol, (b) décantation et/ou centrifugation et/ou filtration clarifiante de l’extrait obtenu suite à l’étape (a) ;(a) dispersing and extracting, in an appropriate solvent, Quinoa seed epicarp and advantageously cryo-ground epicarp, the solvent advantageously being a water / l, 3-propanediol solvent, (b) decantation and / or centrifugation and / or clarifying filtration of the extract obtained following step (a);
(c) microfiltration de l’extrait obtenu à l’étape (b) ;(c) microfiltration of the extract obtained in step (b);
(d) récupération de l’extrait de péricarpes de Quinoa ;(d) recovery of Quinoa pericarp extract;
(e) le cas échéant, ultrafiltration et/ou nanofiltration de l’extrait obtenu à l’étape (c) ; puis (f) séchage éventuel de l’extrait obtenu à l’étape (e) avec ou sans support de séchage.(e) where appropriate, ultrafiltration and / or nanofiltration of the extract obtained in step (c); then (f) optional drying of the extract obtained in step (e) with or without a drying medium.
The extract thus obtained turns out to be different from that described in the document of L’ORÉAL (QHE-IFFCC, November 2016). It indeed appears in FIG. 2 that the Sacaca and Marangani varieties differ from the Real variety by the presence of a type of saponin at significantly higher levels in the extract, that is to say oleanolic acid 3 -Q- [3-D-glucopyranosyl] -28-Q- β-D-glucopyranoside. The latter can therefore be considered as the key marker of the Sacaca and Marangani species.
In a preferred embodiment, the hydro-alcoholic extract of the pericarp of the seed Chenopodium Quinoa Willd is obtained from the varieties of yellow Quinoa Sacaca and Quinoa y aune Marangani.
In a preferred embodiment, the saponin type compounds are present at more than 50% in the extract. The varieties of Sacino Yellow Quinoa and Marangani Yellow Quinoa have a higher saponin content in the pericarp than the Real variety in particular. In addition, the varieties of Sacaca yellow quinoa and Marangani yellow quinoa are different from those of the Real variety by the different relative ratios of saponins.
Ainsi, dans un mode de réalisation très préférentiel selon l’invention, l’extrait hydro-alcoolique selon l’invention comprend majoritairement des saponines du type :Thus, in a very preferred embodiment according to the invention, the hydro-alcoholic extract according to the invention mainly comprises saponins of the type:
Acide phytolaccagénique 3-O-[ 3-D-glucopyranosyl-(l->3)-O-a-Farabinopyranosyl]-28-Q-D-glucopyranoside.Phytolaccagenic acid 3-O- [3-D-glucopyranosyl- (l-> 3) -O-a-Farabinopyranosyl] -28-Q-D-glucopyranoside.
As demonstrated in Example 4 below, the hydroalcoholic extract of the pericarp of the Chenopodium Quinoa Willd seed according to the invention, without itself having hydrating activity on the skin, makes it possible to stimulate synergistically the activity of a skin moisturizing ingredient such as glycerin.
A second object according to the invention relates to a cosmetic composition comprising in a physiologically acceptable excipient, as active agent, the combination of an effective amount of a hydro-alcoholic extract according to the invention and of a moisturizing ingredient skin.
The Applicant has discovered that an effective amount of a hydroalcoholic extract of the pericarp of the seed Chenopodium Quinoa Willd obtained from varieties of the yellow Quinoa Sacaca and of the yellow Quinoa Marangani present in association with a skin moisturizing ingredient a synergistic activity on the hydration of the skin.
Thus, the combination of the two ingredients of the composition turns out to allow a synergy of hydrating activity at the level of the skin.
These results are presented in Example 4 below, more specifically in the demonstration of a synergy of hydrating activity between the extract according to the invention and the glycerin.
In a very preferred embodiment according to the invention, the hydroalcoholic extract of the pericarp of the seed of Chenopodium Quinoa Willd and the skin moisturizing ingredient are each present in the composition according to the invention at a concentration of 0.001 to 10% by weight relative to the total weight of the composition, preferably from 0.01 to 4%.
In a particularly advantageous embodiment, the effective amount of the hydro-alcoholic extract of the pericarp of the seed of Chenopodium Quinoa Sacaca and / or Marangani is between 0.2 and 2% by weight relative to the total weight of the composition , preferably between 0.5 and 1% by weight relative to the total weight of the composition, and the effective amount of the skin moisturizing ingredient is between 2 and 4% by weight relative to the total weight of the composition, preferably is 3% by weight relative to the total weight of the composition.
Dans un autre mode de réalisation selon l’invention, l’ingrédient hydratant cutané dans la composition cosmétique est choisi parmi :In another embodiment according to the invention, the skin moisturizing ingredient in the cosmetic composition is chosen from:

The pericarps of the seeds of a mixture of yellow Quinoa of the Sacaca and Marangani varieties, previously dried, are cryo-crushed in a knife mill with a 1 mm grid, which allows to obtain a powder whose particle size is mostly located around 400 to 500 pm.
The powder (150 g) thus obtained is then placed in a glass reactor, provided with stirring and with a jacket heating using a heat transfer fluid.

Exemple 2 : Analyse analytique d’un extrait du péricarpe de la graine de Quinoa selon l’inventionExample 2 Analytical Analysis of an Extract of the Pericarp from the Quinoa Seed According to the Invention
Isolation of the compounds by preparative chromatography and identification of the saponins by NMR (nuclear magnetic resonance) and LC / MS (liquid chromatography / mass spectroscopy) from a hydro-alcoholic extract of Quinoa Sacaca.
Quinoa extracts contain two types of compounds, very polar minority compounds and majority compounds of medium polarity. These represent the active ingredients according to the invention.
A first stage of sample preparation was made by extraction on solid phase (SPE) in order to remove the most polar compounds and to keep only a fraction enriched in compounds of interest. To achieve this, the sample was placed on a cartridge filled with a specific material (divinylbenzene-polystyrene modified with functional groups), then the successive passage of water, methanol and dichloromethane made it possible to recover the different chemical families. in different fractions. In a second step, the different compounds of the methanolic fraction were separated by preparative chromatography. To do this, a solid deposit was made and the extract was injected onto a Cl8 column of suitable size. A gradient of elution solvent ranging from a water / acetonitrile mixture (ACN) (70:30 v / v) to a water / ACN mixture (25:85 v / v) allowed the collection of six compounds of interest .
The study of these compounds by nuclear magnetic resonance (1 H NMR, 13 C NMR and COZY (Correlation SpectroscopY) and by mass spectrometry (MS) allowed the identification of two saponins among the 6 purified (Table 7).

La structure générale des saponines de Quinoa sont illustrées ci-dessous :The saponins of Quinoa consist on the one hand, of a triterpene unit called aglycon which can vary by the nature of the substituents R2 and R3, and of glycosylated chains on the other hand. R2 and R3 may be methyl groups, alkyl chains oxidized or not. Ri and R4 may be glycosylated chains consisting of one or more sugars. The general structure of Quinoa saponins are illustrated below:
J. Am. Soc. Mass Spectrom. 2006, 77(6), 795-806.).By comparing the fragmentation patterns of saponins 1 and 2 with those of the other compounds of interest, it was possible to ensure that the other constituents of the fraction were indeed saponins due to the loss (s) of neutral observed characteristic of the loss of sugar grouping and of the mass observed for the residual aglycon. These fragments, as well as the masses observed in LC / MS support the hypothesis that the other compounds are indeed saponins by comparison with the data in the literature (Madl, T. et al., Tandem Mass Spectrometry Analysis of a Complex Triterpene Saponin Mixture of Chenopodium Quinoa. J. Am. Soc. Mass Spectrom. 2006, 77 (6), 795-806.).
1. Analyse des extraits par chromatographie liquide1. Analysis of the extracts by liquid chromatography
Le gradient d’élution est réalisé à l’aide d’un mélange eau/ACN 95:5 (v/v) + 0.1% acide formique (HCOOH) (voie A) et isopropanol (IPA)/ACN 80:20 (v/v) + 0.1% HCOOH (voie B) allant de 100 % A à 100 % B.The analysis of Quinoa extracts is performed on reverse phase (column Cl8) by high performance liquid chromatography (HPLC) coupled to an evaporative light detection detector (DEDL). The elution gradient is carried out using a water / ACN 95: 5 (v / v) + 0.1% formic acid (HCOOH) (route A) and isopropanol (IPA) / ACN 80:20 (v / v) + 0.1% HCOOH (channel B) ranging from 100% A to 100% B.
Under these analytical conditions, the saponins elute between 16 and 19 minutes as presented in FIG. 1 (DEDL profile of an extract 1,3-propanediol / Quinoa water with diosgenin as internal standard).
Figure 2 presents the comparative profiles of the Quinoa Real varieties on the one hand and the Quinoa Sacaca and Quinoa Marangani varieties on the other hand.
Quinoa saponins are quantified by internal calibration in diosgenin equivalent. This standard was chosen because this compound belongs to the same chemical family of triterpenes as the saponins of Quinoa.
Under these conditions, the saponin content is greater than 0.5% in diosgenin equivalent.
Estimation of the saponin content by column fractionation
Pour obtenir un ordre de grandeur de la teneur en saponines dans un extrait deTo obtain an order of magnitude for the content of saponins in an extract of
Quinoa, the latter was fractionated on reverse phase Cl8 in order to separate the sugars from the saponins. An elution gradient ranging from 100% water to 100% ACN followed by rinsing the phase using an IPA / ACN 80:20 (v / v) mixture was used. The proportion of saponins relative to the sugars was evaluated by establishing the mass yield of each fraction relative to the initial mass of extract used in this fractionation. The proportion of saponins is the majority in the extract and corresponds to 57%, that of the sugars is 27%. The remaining percentage corresponds to an intermediate fraction which contains in particular a mixture of saponins and sugars.

The purpose of this study is to evaluate the hydration of the superficial layers of the skin by application of the combination of an extract of Chenopodium Quinoa according to the invention and of glycerin. It is based on the relationship between the electrical properties of tissues and their free water content (capacitance). The coméometer measures this electrical capacity using a probe placed on the surface of the skin at a constant pressure. The results obtained are expressed in arbitrary units and designate a Hydration Index (HI) ranging from 0 to 130, from very dry to hydrated skin.
A variation in hydration of the cutaneous corneal layer results in a modification of the total capacity of the system.
1. Introduction1. Introduction
Cutaneous bioengineering, or cutaneous biometrics, is the study of the biological, mechanical and functional characteristics of the skin by means of a strict measurement of certain variables with a non-invasive scientific method.
Skin care products for normal, dry and greasy skin. In: BARAN & MAIBACH (eds). Cosmetic Dermatology. Williams & Wilkins, Baltimore, p.89-115, 1994).The main parameters that can be used to assess the effectiveness of a product on the skin are morphological changes on the surface of the skin, stratum corneum, hydration and sebaceous secretion. Since these parameters vary between different anatomical zones of the same individual and between individuals, these techniques are used to measure comparatively the variations of the same parameter, in the same zone, before and after the use of a product. (GALL et al., J.P. Skin care products for normal, dry and greasy skin. In: BARAN & MAIBACH (eds). Cosmetic Dermatology. Williams & Wilkins, Baltimore, p.89-115, 1994).
In the analysis of skin hydration, the most commonly used methods are corneometry and transepidermal water loss. The methodology described in this protocol is the measurement of skin electrical capacity (corneometry), based on the principle of electrical conductivity.
Cosm. Toiletries. 99:43-50, 1980).The passage of low frequency alternating current through the seed coat depends on the water content of the stratum corneum and its integrity. Thanks to this electrical property of the skin, skin hydration can be measured indirectly (LEVEQUE, J.L. physical methods to measure the efficiency of cosmetics in humans. Cosm. Toiletries. 99: 43-50, 1980).
