Dans le follicule pileux de cheveu blanc, certains ARNm sont sur-représentés (gènes sur- exprimés) et d'autres sont sous-représentés (gènes sous-exprimés). Ces gènes sont des bio marqueurs de la canitie et leurs changements d'expression sont associés au développement de la canitie. Le niveau d'expression de ces bio marqueurs de canitie est analysé par la technique classique de RT-PCR quantitative (Q-RT-PCR) dans des follicules pileux humains. 10 Pour la réalisation de l'essai, les follicules pileux humains de cheveux blancs sont microdisséqués et placés en culture in vitro dans le milieu de culture William's E (Gibco-BRL) supplémenté notamment en glutamine, insuline, hydrocortisone et antibiotiques. Pour le groupe de follicules pileux « essai » (30 follicules), le milieu de culture est 15 complété avec l'extrait de Rubus fructicosus. Pour le groupe « contrôle » (30 follicules), le milieu de culture est complété avec le véhicule (en même quantité que pour le groupe « essai »). Après le temps nécessaire à la révélation de l'activité de l'extrait, en général de 24h à 5 jours, les ARNm des follicules pileux sont extraits. Les ARNm de la même condition 20 expérimentale sont regroupés. Le niveau d'expression des ARNm d'intérêts est déterminé par une méthode classique de Q-RT-PCR, par exemple à l'aide du kit QuantiTect SYBR® Green RT-PCR référence 204243 de Qiagen, la mesure pouvant être réalisée sur l'appareil MyIQ de BIORAD. 25 L'extrait de Rubus fructicosus corrige les changements d'expression de certains bio marqueurs de la canitie dans les follicules pileux des cheveux blancs.
