Abstract
Description
<Desc/Clms Page number 1> <Desc / Clms Page number 1>
The present invention relates to a cosmetic or pharmaceutical product that contains an active ingredient to fight against adiposity and thus refine the silhouette.
Such products are already known and it is known that the active principles used exhibit lipolytic activity by which they stimulate the degradation (or lysis) of the fats stored in human adipocytes, especially in the form of triglycerides, and promote the elimination of this degradation, in particular fatty acids and glycerols.
For example, in French Patent No. 98 02152, the active ingredient used is a pyrone group which has the effect of activating the phosphorylation of a lipase enzyme for the degradation of triglycerides to fatty acids. For example, this pyrone group is mangostine extracted from a tree of the species Garcinia Mangostana.
It is also known that, in adipocytes, lipolysis is in competition with lipogenesis, which consists of the formation of fats, especially triglycerides, and which involves a series of biochemical reactions involving proteins with enzymatic activity such as Acyl-CoA synthetase and fatty acid synthetase
also known as FAS (fatty acicsynthase).
The purpose of the present invention is to provide a cosmetic product that contains an active ingredient to fight against adiposity and is new, especially compared to products whose active ingredient promotes lipolysis.
For this purpose, a cosmetic product according to the present invention contains as active principle at least one fatty acid used to exhibit an inhibitory activity on the lipogenesis of human adipocytes, said at least one fatty acid being derived from a lipid extract of plants or algae.
As shown below, such a cosmetic product can limit the accumulation of fatty acids in human adipocytes. It is believed that the fatty acids contained in said extract are responsible for an effect of inhibiting the action of an enzyme involved in lipogenesis, including the synthesis of fatty acids by fatty acid synthetase FAS. By using a product which contains such an active principle, at least the lipogenesis of the fatty acids is inhibited or at least limited and their concentration in the adipocytes is thus reduced.
Il en est de même des acides gras présentant un degré It has been shown that fatty acids having a carbon number between 12 and 22 can be used. It is the same with fatty acids having a degree
<Desc/Clms Page number 2><Desc / Clms Page number 2>
Elle fait partie des diatomés dont la classe est celle des Bacilariophycées, l'ordre celui des Bidulphiales, et la famille celle des Eurodiscaceaes. of unsaturation between 0 and 5 It is still the same of free fatty acids, or monoglyceric, or diglyceric or triglycerical
According to another characteristic of the invention, said extract is a lipid extract of an alga or a microalga
An example of a cosmetic product based on a lipid extract of a microalgae which is, in this case, a cyanobacterium named Odontella aurita. It is one of the diatoms of which the class is that of the Bacilariophyceae, the order that of the Bidulphiales, and the family that of the Eurodiscaceae.
This particular microalgae contains polyunsaturated fatty acids such as
eicopentanoic acid (5, 8, 11, 14, 17-eicosapentaenoic acid) known as EPA in a concentration of about 22% of the total fatty acids, or approximately 2% of the dry weight of the microalgae. It is therefore rich in EPA.
This microalga is obtained by controlled culture in outdoor pools (race way type)
The present invention also relates to a process for preparing a product which contains as an active ingredient a lipid extract of plants or algae. It is characterized in that said extract is produced by supercritical CO2 gas extraction from said previously dried plant or said previously frozen algae which is then lyophilized.
We will now describe the procedure of the experiments that were conducted to prove the inhibitory effect of fatty acids on the neosynthesis of lipids.
In all these experiments, the adipocytes were isolated from abdominal plasties taken from women aged 34 to 52 years. The experiments took place on the day of collection or the day after sampling. The fragments of adipose tissue were incubated for 30 minutes at 37 ° C. in the presence of collagenase, then the isolated adipocytes were washed and taken up in a bicarbonate-buffered MEM medium, in the presence of antibiotics (penicillin / streptomycin), glutamine ( 2 mM), and serum bovine albumin delipidated (0.5%)
To 90 l of the same suspension of adipocytes of predetermined concentration, were added 100 l of a culture medium, then after a pre-incubation of about 1 hour, 10 l of a marker of [2-14C ] -acetate 50 μCi / ml. The samples thus formed undergo incubation at 37 ° C., 5% CO 2, for 4 hours, after which they are subjected to freezing at -80 ° C. The experiments are always carried out in double or triple
<Desc/Clms Page number 3> <Desc / Clms Page number 3>
The lipids are then extracted according to the method of Bligh and Dyer (extraction methanol / chloroform / water), dried under nitrogen and the incorporated radioactivity (Radioactivity resulting from the incorporation of the carbon 14C in the neosynthesized lipids in the adipocytes) is counted in liquid scintillation. The raw count data was transferred and processed under appropriate software; intergroup comparisons were performed by ANOVA using Dunnett's multiple comparison test.
The background noise (radioactivity measured at time to) is taken into account.
This procedure was validated using cerulenin which is known to inhibit the expression of fatty acid synthetase.
The incorporation of acetate after 4 hours of incubation is strong (133,000 cpm); the background noise at to was negligible (1% of the time 4 hours).
It has thus been found that at concentrations of 10 M (gmoles per liter) and 1 M, cerulenin effectively inhibits the incorporation (respectively 93 and 56% inhibition). On the other hand, it is inactive at a concentration of 0.1 M. These results therefore validate the above procedure.
In addition, it has also been found that the presence of mineral oil, such as liquid petrolatum, in the culture medium has not inhibited the incorporation or extraction of adipocyte lipids. Vaseline oil is therefore used as a buffer medium in which in particular the fatty acids tested will be previously diluted.
It has been sought to demonstrate the inhibitory activity of fatty acids on the neosynthesis of lipids produced by human adipocytes and to determine the properties of those which give maximum activity.
We therefore, first, study the effect of the length of the carbon chain of the fatty acids tested.
heure. Puis, mélangé à IOjj.1 d'un marqueur de [2-14C]-acétate 50 Ci/ml, il a été incubé pendant 4 heures To form the incubation medium, 12,14,16,18, 20 and 22 carbon fatty acids were diluted in petrolatum oil, which is recalled that the presence does not inhibit incorporation or extraction of adipocyte lipids. This medium was therefore mixed with a suspension of 90 l of adipocytes and was preincubated for 1 hour.
hour. Then, mixed with 10 μl of a [2-14 C] -acetate marker 50 Ci / ml, it was incubated for 4 hours.
<Desc/Clms Page number 4> <Desc / Clms Page number 4>
The incorporation of acetate after 4 hours of labeling is very strong (of the order of 200,000 cpm); the background noise at to is negligible (1% of the time 4 hours). This background noise has been subtracted from the final results.
These results are given in Table 1 below.

Note that the inhibition rate is the complement of the rate of incorporation given in% control in Table 1 above (inhibition rate = 100 -% control).
It can be seen that all the fatty acids tested inhibited the incorporation of acetate in a dose-dependent manner compared with the control Vaseline oil. At the lowest concentration tested (10 M), the inhibition was about 10-20% for saturated C12 to C16: 0 saturated fatty acids, and 30-40% for Cl 8: 0 and C20 saturated fatty acids. : 0. At the average concentration of 100 μM, the inhibition is 25% for the C12.0 and C14: 0 fatty acids, and 45-50% for the C16: 0 and C18: 0 fatty acids. It is greater than 60% for the C20 fatty acid: with the exception of the C22: 0 fatty acid, all the fatty acids, at the highest concentration of 1 mM, inhibited the incorporation with an efficacy of order of 80-85%.
C22 0 fatty acid at a concentration of 1 mM inhibited incorporation by only 40%.
It can therefore be concluded that the inhibition of lipogenesis is optimal for the 18- and 20-carbon fatty acids, but then decreases for the 22-carbon fatty acids.
The effect of the degree of unsaturation was evaluated on the fatty acids containing 20 carbons, namely the fatty acids C20 0, C20 2, C20: 3, C20: 4 and C20: 5. recalls that these 20-carbon fatty acids are among the most efficient, as has been demonstrated in the previous experiment. The results are given in Table II below.

Table II: Effects of the number of fatty acid unsaturation on the incorporation of [14 C] -acetate into adipocyte lipids. sd, standard deviation
<Desc/Clms Page number 8> <Desc / Clms Page number 8>
As can be seen, the incorporation of acetate after 4 hours of marking is high (of the order of 50 000 cpm), the background noise at to is low (2-3% of the time 4 hours). This background noise has been subtracted from the final results.
All C20: 0, C20: 2, C20: 3 and C20: 4 fatty acids inhibited the incorporation of acetate in a dose-dependent manner with respect to the petrolatum oil control. As for the fatty acid C20: 5, called EPA (eicopentanoic acid), it has, in oily solution at the 3 concentrations tested (0.01 mM, 0. 1 mM and 1 mM, in petrolatum oil), strongly inhibited the incorporation (90% inhibition).
It can thus be seen that the inhibition of lipogenesis increases with the degree of unsaturation.
The effect of the type of binding of the fatty acids concerned was also evaluated, in particular on the 18-carbon and 3-unsaturated fatty acids, namely the C18: 3L free fatty acid, the C18: 3MG monoglycerol fatty acid, diglyceric fatty acid C 18: 3DG and triglyceric fatty acid C18: 3TG. The results are given in Table III below.

As can be seen, at the lowest concentration tested (10 M), the tested fatty acids have no significant inhibitory effect (> 10%) unlike EPA (22% inhibition) A 100 M C18: 3DG, C18: 3MG and C18: 3L fatty acids inhibited incorporation by 25%, 82% and 32%, respectively. The fatty acid C 18: 3MG was therefore the most effective at this concentration.
At the highest concentration tested (1mM), the C18: 3DG fatty acid inhibited the incorporation by 54%, whereas the C18: 3MG and C18: 3L fatty acids inhibited the incorporation by an amount greater than 95%. %, like the EPA (98%).
It can also be noted that at the 3 concentrations tested, the triglyceric fatty acid C18: 3TG did not show any effect on the incorporation of acetate under these experimental conditions.
Fatty acids in free and monoglyceric form are therefore more effective than in the diglyceric form Fatty acids in triglycerol form have no effect.
An oily extract derived from a micro-alga containing polyunsaturated fatty acids, in particular free EPA (C20: 05L) in a concentration of approximately 10%, was also tested. This extract was previously diluted to 10% in a vegetable oil (HV) containing no EPA. The tested extract (ODON) therefore contained about 1% EPA. More particularly, were tested such an extract preserved oxidation by the presence of an antioxidant (ODON 0.5) and another extract without antioxidant (ODON 0). The oily extracts were diluted in petrolatum oil.
The results are listed in Table IV below.

As can be seen from Table V above, these oily extracts strongly inhibited acetate incorporation:% inhibition at 10% concentration; 70-80% inhibition at 1% and 0.1% concentrations.
These results were confirmed by thin layer chromatography (TLC) measurements which furthermore proved that these effects do not come from quenching (this control was necessary since the products are colored and are extracted by the solvents used).
Finally, the vegetable oil HV which contains polyunsaturated fatty acids, in particular linoleic acid (> 50%) in triglyceride (C18: 2TG) form, has been tested alone. This test was conducted in conjunction with EPA (C20: 5), sialic acid (C18: 0) and linoleic acid (C18: 2) in their free form. These fatty acids were diluted in petrolatum oil. The results are shown in Table V below.

Acetate incorporation after 4 hours of labeling was strong (54,000 cpm); the background noise at to was negligible (2% of the time 4 hours)
<Desc/Clms Page number 14> <Desc / Clms Page number 14>
As can be seen in the table above, Vegetable Oil (HV) at 9% and
0.9% significantly inhibited the incorporation of acetate in lipids, respectively 53% and 76% of the control against only 2% n and 26% of the control with the ODON extract at the same concentrations of HV oil. On the other hand, the inhibition of HV is no longer significant at the 0.09% concentration corresponding to approximately 500 μM of C18: 2TG (88% of the control) whereas the 0.1% ODON extract inhibited strongly and significantly statistic incorporation of acetate (only 22% of the control).
Thin layer chromatography measurements confirmed these effects.
It will therefore be noted that the effects obtained with the vegetable oil HV are very much lower than those obtained with the oily extracts containing the free polyunsaturated fatty acids of the ODON microalgae.
L'EPA en solution huileuse à 1 mM (huile de vaseline) a fortement inhibé l'incorporation (80 % d'inhibition). EPA in oily solution at 1 mM (petrolatum oil) strongly inhibited the incorporation (80% inhibition).
Sialic acid (AS or C18: 0) at 1000 M and 100 M strongly inhibited acetate incorporation (respectively 74% and 69% inhibition). The inhibition was no longer significant at the 10M concentration (84% of the control).
As for linoleic acid in free form (LA or C18: 2) at 1000 M and 100 M, it strongly inhibited the incorporation of acetate (respectively 88% and 63% inhibition). The inhibition was no longer significant at the 10M concentration (97% of the control).
This experiment confirms the inhibition of lipid neosynthesis in human adipocytes by fatty acids with 18 or 20 carbons, in particular, saturated or unsaturated, preferably in free form or monoglycerides.
