Abstract
Description
The present invention relates to a novel series of condensation products of silanols and their derivatives; it relates more particularly to such products, formed by the condensation of silanols with organic compounds, bearing at least one carboxylic function. These novel products also include condensates of silanols with organic compounds which, in addition to the carboxylic function, have one or more hydroxyls and / or one or more amino groups. They may have siloxane groups in their molecule. The invention also relates to a process for preparing these condensation products, as well as their applications, in particular medical, cosmetic, rheological or as adjuvants of plastics.
The futility of silanols, siloxanes and their various derivatives is well known. It is known, for example, that alkyl silanes, such as methyl silanetriol and their salicylates, exert on the connective tissues a very beneficial action; they prevent sclerosis, stimulate the regeneration of the tissue and ensure the stabilization of it. On the other hand, organic polysiloxanes are used as additives of plastics which they facilitate the demolding; it is also used for the lubrication of organic fibers and as emulsifiers and foaming agents.
The present invention provides an improvement over the known technique of organic silicon compounds, in that it is aimed at condensation products which are very stable to heat, which can be handled, transported and preserved in solid or oily form and not necessarily in solution as are the products mentioned above. It is possible to have, according to the invention, compounds which hydrolyze or alcoholise in solution, thus giving incipient, particularly active products.
The novel products according to the invention, which comprise an Sn atom bearing two organic radicals and two oxygen atoms linked to organic groups other than said radicals, are characterized in that at least one of these oxygen atoms is separated from its organic group by a carbonyl.
One or both organic groups of these products may be cyclic or together form a common ring.
The products according to the invention can be represented by the general formula
where the radicals R and R1, similar or different; are hydrocarbon or oxyhydrocarbon, aliphatic or aryl radicals; R 2 and R 3, which are similar to or different from each other and different from each of R 1 and R 1, are linear or cyclic organic groups which may carry various substituents, in particular amine or acyl functions.
R3 peut comprendre un C=O. R3 can include a C = O.
Similarly, R and R3 can be C1-C18 alkyl or alkenyl, aryl, alkoxy, aryloxy, cycloalkyl or cycloalkenyl, defined as above, but different from R and R1.
de la molécule.R2 and R3 can be linked to each other, in which case they form a cycle with the part
of the molecule.
An important case is where Rss and R3 form a common ring, for example benzene or furanic; the product then comprises two rings having two common C atoms, that is to say one which comprises the part
of the molecule and the second benzene, furanic or other.
This is the case for example benzo carboxy siloxane type
When the products according to the invention are intended for biological uses, especially as medicaments or in cosmetics, it is much preferable for only one of the radicals R and R 1 to be hydrocarbon, that is to say al-kyl, alkenyl or aryl, the second being of a different nature, in particular alkoxy, alkenoxy, aryloxy, hydroxyalkyl or amino. It has indeed been found that silanols and their derivatives, whose silicon atom carries more than one hydrocarbon radical, linked to Si by a C-Si bond, have a lower efficiency than compounds having a single bond of this type. type.
This is why those of the products according to the invention, which are intended for pharmaceutical applications, preferably correspond to formulas (1) or (2), in which R can be - as indicated above - a hydrocarbon radical or oxyhydrocarbon, while R1 is never a hydrocarbon radical. In other words, in this form of the invention, preferred for pharmaceuticals, the part of the molecule
derived from a mono-alkyl-, alkenyl- or aryl- (R) silane di-tri-ol, where R1 may be a R40-bond -O-Si group where R4 is allyl, alkenyl, aryl, cycloalkyl, optionally hydroxylated hydrogen, ether or polyether, this group R40 can be blocked, for example by an acid molecule, R1 can, on the other hand, be constituted by an amine or amide function, optionally blocked.
Here are, as purely illustrative examples, the formulas of the part
of some of the condensation products according to the invention, particularly recommendable for pharmaceutical use
Cette illustration n'a, bien entendu, aucun caractère limi tatif. This illustration has, of course, no limiting character.
The condensation products according to the invention can be prepared by the action of a silane or a siloxane, having at least two reactive groups, with a carboxylic acid which can carry a second carboxyl function, a hydroxyl or / and a group amino, the reaction taking place in an anhydrous organic solvent, in the presence of a ne u tr li li ntl, the preparation is completed by the elimination of the solvent at boiling,
The silanes containing at least two reactive groups are in particular di- and tri-halides of di- and mono- hydrocarbyl silicon respectively, or R2SiX2 and RSiX3, where R is a hydrocarbon radical and X is a halogen; eg SiLanes are, for example: (CH 3) 2 SiCl 2 and CH SiCl 3. Toute-
Three times, dihalogenated compounds are much more preferred, because of the subsequent complications, namely the crosslinking polymerization, which may cause the third halogen.
The siloxanes that can be used in the process according to the invention are organosilicon of the R2, SiQ2 and R'5iQ3 types, R 'being a hydrocarbon or oxy-hydrocarbon radical, for example -CH3 or CH30-, while Q is -OH or -OR, R denoting, as above, a hydrocarbon radical. Examples of such siloxanes are, without limitation (CH 3) 2 Si (OH) 2; (C2H5) 2Si (OCH3) 2; C6H5-Si (OC2H5) 3
C4HgO-Si (OH) 3; If (OC2H5) 4; C2H5-Si (OH) 3
and the like.
The compounds, which are preferred for the process of the invention, are those of which R is a lower alkyl (C1-C4), leading to the formation of the corresponding alcohol, easily separable by distillation.
As carboxylic acids, to be condensed with the siloxane, it is possible to use different aliphatic or aryl acids, diacids and acids carrying other functions, especially OH and / or NH 2. Can one thus use saturated or ethylenic monoacids C1 to C18, in particular at 2 mol per atom of Si. The diacids, both aliphatic and aryl, are interesting, especially when their two -COOH groups are in & or Y in relation to each other; such are, for example, malic, maleic, fumaric, malonic, succinic, tartaric, ortho and metaphthalic, glutaric, aspartic, glutamic, succinylsalicylic, phenylene diacetic, mellic, etc.
Ainsi conviennent des acides tels que glucuronique, lactique, butanol-4 orque, hydroxy-3 propionique, ortho-hydroxycinnamique (coumarique) s- our7- hydroxy benzoSque, -hydroxy napht-ofqueldihydroxy ben osque, sérine, di-sérine, thréonine, hydroxytoline, tyrosine, caséine, acide amino phényl lactique, pyridyl orthodicarboxylique, (fonction amine d préférence bloquée).Another important class of carboxylic acids making it possible to obtain the products according to the invention are the alcoholic acids, among which the hydroxylated amino acids are very interesting. Thus acids such as glucuronic, lactic, 4-butanol, 3-hydroxypropionic, ortho-hydroxycinnamic (coumaric) s-7-hydroxybenzoic, -hydroxy-naphth-of-dihydroxy-benosque, serine, di-serine, threonine, hydroxytoline are suitable. , tyrosine, casein, amino phenyl lactic acid, pyridyl orthodicarboxylic acid, (amine function preferably blocked).
As a nonlimiting illustration, here are some diagrams giving an idea of the condensation reaction of organic silicon compounds.
As regards the anhydrous organic solvents used in the preparation of the condensation products according to the invention, they are chosen from the liquid of the solvents dissolving the silicon compound and the acid used, and preferably - boiling at temperatures which do not do not exceed about 150OC. These are, of course, liquids not reacting with the reagents involved. Thus, it is possible to use, as the case may be, aliphatic hydrocarbons, especially C 6 to C 12 hydrocarbons, aromatic compounds such as benzene, toluene, xylene, etc., chlorinated solvents, terpenes, ethers such as dioxane, tetrahydrofuran, and the like. It is very convenient to employ, as a solvent, a silane or silanol, particularly that which is desired to be reacted. As it is indicated to terminate the reaction by heating, preferably between 500 and 100 ° C. this heating serves at the same time for the removal of the solvent, and it is carried out under a pressure, possibly reduced, under which the solvent boils at the appropriate temperature.
Neutralizers are well known in chemical technology, so there is no need to mention them here; it is recalled only that various amines, such as trimethylamine, dibutylamine or pyridine can be used in the process according to the invention.

As indicated above, in the present description, for pharmaceutical applications, the compounds whose Si atom carries only one hydrocarbon group are particularly suitable.
