Abstract
PROCEDE DE PREPARATION DE SOLUTIONS AQUEUSES D'ALLOXANE STABLES INJECTABLES CONDITIONNEES EN AMPOULES STERILES, CARACTERISE EN CE QU'ON INTRODUIT DANS DE L'EAU POUR SOLUTIONS INJECTABLES, UNE QUANTITE PREDETERMINEE D'ALLOXANE, QU'ON AJOUTE 5 A 25 FOIS PLUS GRANDE D'UN SUCRE REDUCTEUR, QU'ON FILTRE SUR MEMBRANE FILTRANTE UTILISEE EN BACTERIOLOGIE, PUIS QU'ON INTRODUIT EN ATMOSPHERE STERILE LE SOLUTE OBTENU DANS UNE AMPOULE DE 5 ML QUE L'ON SCELLE AU CHALUMEAU ET SOLUTIONS AQUEUSES D'ALLOXANE STABLES EN RESULTANT.
Description
La demande de brevet nO 79-15363 du 15 Juin 1979 a pour objet un procédé de préparation de solutions aqueuses d1alloxane injectables, caractérisé en ce que l'on introduit dans du sérum physiologique une quantité prédéterminée d'alloxane et une quantité 8 à 25 fois plus grande d'un sucre réducteur, qu'on filtre, introduit dans une ampoule de 5 ml qu'on scelle au chalumeau et stérilise par tyndallisation à 60-700C.
La présente addition concerne un perfectionnement à ce procédé pour la préparation d'ampoules stables injectables, consistant à réaliser la stérilisation de la solution avant la mise en ampoules, en procédant à une filtration sur membrane filtrante utilisée en bactériologie, à température ambiante.
On choisit avantageusement une quantité de sucre réducteur égale à environ 5 à 15 fois la quantité d'alloxane anhydre introduite dans la solution.
L'exemple suivant illustre la mise en oeuvre de l'invention.
On utilise de l'eau pour solutions injectables et on ajoute par millilitre d'eau
- 10 mg d'alloxane pur anhydre ou bien 12,27 mg d'alloxane
monohydraté, ou bien 15,07 mg d'alloxane tétrahy draté,
- 50 mg de lévulose (ce qui correspond à une solution isoto
nique de lévulose).

On filtre la solution obtenue sur membrane filtrante à la température ambiante.
On remplit une ampoule de 5 ml et on la scelle en atmosphère stérile.
Cette ampoule contient 50 mg d'alloxane et 250 mg de lévulose. Les ampoules sont conservées à 40C maximum et de préférence à l'obscurité.
Le contrle de l'absence d'altération de l'alloxane a été étudié grâce à la spectrophotométrie en lumière ultra-violette.
On sait qu'en milieu alcoolique l'alloxane donne un spectre d'absorption en lumière ultra-violette, caractérisé par un maximum à 272 millimicrons et un minimum à 262 millimicrons.
Des mesures comparatives ont été réalisées sur des so lutions d'alloxane préparées selon le procédé de l'invention (avec sucre réducteur et notamment le lévulose) et sur des solutions contenant la meme quantité d'alloxane mais en sérum physiologique sans lévulose.
On a établi les courbes spectrales de l'alloxane dans ces deux types de solutions en fonction du temps.
Il a été ainsi constaté que seules les solutions dans lesquelles l'alloxane est associée au lévulose sont d'une stabilité remarquable pendant au moins un an.
Claims (4)
1. Procédé de préparation de solutions aqueuses d'alloxane stables injectables conditionnées en ampoules stériles, caractérisé en ce qu'on introduit dans de l'eau pour solutions injectables, une quantité prédéterminée d'alloxane, qu'on ajoute 5 à 25 fois plus grande d'un sucre réducteur, qu'on filtre sur membrane filtrante utilisée en bactériologie, puis qu'on introduit en atmosphère stérile le soluté obtenu dans une ampoule de 5 ml que l'on scelle au chalumeau.
2. Procédé de préparation de solutions aqueuses d'alloxane stables injectables conditionnées en ampoules stériles, selon la revendication 1, caractérisé en ce qu'on introduit dans de l'eau pour solutions injectables 10 mg d'alloxane pur anhydre et 50 mg de lévulose par ml d'eau.
3. Solutions aqueuses d'alloxane stables injectables caractérisées en ce qu'elle contiennent, par millilitre d'eau, pour solutions injectables, une quantité prédéterminée d'alloxane et une quantité 5 à 25 fois plus grande de sucre réducteur.
4. Solutions aqueuses d'alloxane stables injectables selon la revendication 3, caractérisées en ce qu'elles contiennent par millilitre d'eau : 10 mg d'alloxane et 50 mg de levulose.
