Abstract

Medicaments contg. a thieno 2,3-c and/or 3,2-c pyrdine (I) and/or (II) or their acid addn. salts (where R is cyano, carbamoyl or carboxy), are new. Process for the prepn. of (I) and (II) by treating the corresp. bromo cpds. (I and II where R=Br) with CuCN to give (and II;r=cyano) is alsoclaimed. Hydrolysis of this prod. with a strong mineral acid gives (I and II; R=carboxy) or careful hydrolysis of (I or II; R=CN) to give (I and II; R=carbamoyl). The reaction with cuprous cyanide is effected at 100-200 degrees C in DMF, N-methyl pyrrolidone, hexamethylphosphorotriamide, or benzonitrile. Hydrolysis of the acid is effected by refluxing in HCl, HBr or H2SO4. Hydrolysis of the amide is effected by passing dry HCl or HBr into a solution in formic acid at 20-100 degrees C. Cpds. of formula (I) where R=cyano or carbamoyl and their salts are claimed and cpds. of formula (II) where R = cyano, carbamoyl, or carboxy and their salts are claimed. The medicaments may be given orally, rectally or parenterally.
