Abstract

Thyroxine-binding globulin (TBG) is extracted from the protein fraction of human plasma, which has been treated by cohn's method, and is purified as follows: (a) chromatography on DEAE Sephadex (RTM) eluting with phosphate buffer and 0.13M NaCl; (b) the eluted fractions are submitted to affinity chromatography using a suspension of Sepharose (RTM) to which thyroxin is attached, eluting with K carbonate which increases in strength from 0M to 0.02M as the elution progresses; (c) the eluted fractions undergo another ion exchange chromatography using DEAE Sephadex A 50, eluting with TRIS buffered NaCl which increases in strength from 0.1-0.25M as the elution progresses, and (d) the eluted fractions are purified. TBG may be used to make anti-TBG serum. The process is easier to carry out than known methods of preparing TBG and gives a highly pure prod. having the same electrophoretic and immunological properties as natural TBG.
