Abstract

Process for analysing a serum sample for a non-protein cpd., in which the non-protein mols. are released from a binding protein comprises incubating the serum with an acid pepsin soln. and then adjusting the pH to >=6 to inactivate the pepsin. Pref. the determination of thyroxine (T4) content of a serum sample comprises incubating the sample with an enzyme soln. to release the T4 from T4-binding globulin (TBG). The mixt. is then treated with a TBG-T4-I125 reagent and contacted with a material contg. secondary binding sites. The mixt. is incubated and its initial radioactivity is measured. After this, the material with sec. binding sites is removed, washed, and its residual radioactivity measured. Pref. determination of the triiodothryonine (T3) content of a serum sample comprises incubating the sample with an enzyme soln. to release the T3 from TBG. The mixt. is then treated with a T3-antibody-T3-I125 reagent and treated as thyroxine (T4). A part from the determn. of T3 and T4, process (A) can also be used for the determn. of vitamin B12, steroids and other hormones and drugs.
