Abstract

1-(2-Methoxy-4-amino-5-chlorobenzamido)-4-piperonyl-piperazine (I) and its pharmaceutically active salts (pref. the maleate, hydrochloride and methanelsulphonate) are new cpds. (I) is prepd. by preparing a solution of piperonylpiperazine (II) in C6H6 in the presence of PCl3, heating to reflux, cooling, treating with 2-methoxy-4-amino-k-chlorobenzoic acid (III), heating to reflux, and separating (I) from the reaction mixture. (I) is useful as a tranquilliser. Results of tests with (I) maleate (LD50 1 g./kg. p.o. in mice) are given in the specification. Thus, a dosage of 100 mg./kg. p.o. in mice protects 40% of mice against the lethal effects of 125 mg./kg. pentetrazole s.c. and 50 mg./kg. p.o. gives a 30% reduction in the activity of mice. Dosage for humans in the treatment of neuroses, neurovegetative disorders, anxiety states and psychosomatic disorders is 50-200 mg.
