Abstract

Pure beta-trypsin from pigs is prepd. from a commercial crystalline pig trypsin by chromatographic purification of a soln. of this material in a buffer with an elution coefficient equivalent to that of tris-(hydroxymethyl)aminomethane buffer with a resistance of >0.21x103 OMEGA, pref. between 0.23x103 and 0.35x103 OMEGA. Sulphonated ion exchange agents are used in the separation, esp. SE-Sephadex C-50 or Sp-Sephadex C-50 (RTM). Pref. buffer contains 0.05M tris buffer in Cl-form as well as 0.012-0.02M CaCl2 and 1mM benzamidine. Pref. pH is 7-8.2 at 1-4 degrees C. The prod. is more stable than customary bovine trypsin and uses previously unsuitable starting material.
