Abstract

A process which permits the commercial scale isolation of large amounts of homogeneous populations of Chymopapain B or Chymopapain C, free from cross-contamination and other proteins and with low levels of pyrogens, from either papaya latex or crude chymopapain preparations. This process comprises mixing the papaya latex or crude chymopapain with increments of cation exchange resin to selectively absorb some but not all of the chymopapain onto the resin until the point is reached at which protease activity in the supernatant has decreased to a predetermined minimum level. The chymopapain is then eluted from the cation exchange resin. Therapeutic chymopapain-containing compositions are also disclosed, as is a standardized, reproducible enzyme assay for Chymopapain B and C.
