Abstract

Viruses, antigens or purified vaccines are prepd. by fractional centrifugation of a crude virus conc. in a sterilised rotor (40-90% of vol. of rotor), spinning the rotor at 2000-5000 rev./min. and adding successively to the periphery of the rotor >=2 lots of liq of increasing density, the first being less dense than that of the virus particles but dense enough to prevent migration of the greater part of the impurities, and the density of the second being >= than that of the virus particles. Having completely filled with rotor it is accelerated to such a speed so as to separate the virus from the impurities, slowing the rotor to 2000-5000 rev./min. and introducing a liq. of density >= that of the contents of the bowl to discharge the contents through the central orifice of the rotor and collecting purified virus. Process is simple and cheap.
