Abstract

Ind. scale process for producing inducers of interferon in which a cell or tissue-culture is inoculated with an RNA virus, after the intracellular growth of the virus is finished the cells and debris are collected, e.g. by centrifugation of 2000-4000g, and extracted to give double-chain RNA which is finally inactivated. The collected cells or tissue may be disintegrated by mechanical or chemical means before deproteination e.g. with phenol/CKCl3. Double-chain RNA is pptd. with alcohol, opt. in presence of NaCl, or Na K acetates. Final purification from DNA and single-chain RNA is by chromatography or enzyme treatment. Final inactivation by UV-irradiation or treatment with formalin or beta-propiolactone.
