Abstract
PROCEDIMIENTO PARA LA OBTENCION DE UNA SOLUCION DE CULTIVO QUE CONTIENE LA SECUENCIA DE AMINOACIDOS DE LA PROINSULINA DE MONOS. A PARTIR DE PANCREAS DE MONOS POR DIGESTION PROTEOLITICA SE OBTIENEN CELULAS INDIVIDUALES. POR CENTRIFUAGACION DE DENSIDADES SE OBTIENEN A TRAVES DE UN GRADIENTE UNA CAPA RICA EN CELULAS B QUE SE DESNATURALIZA, OBTENIENDOSE ACIDO RIBONUCLEICO COMO PRECIPITADO. TRAS UNA NUEVA PRECIPITACION, Y CON AYUDA DE LA ENCIMA TRANSCRIPTOSA INVERSA, SE OBTIENE UNA DOBLE HEBRA DE ACIDO ROBONUCLEICO-ACIDO DESOXIRIBONUCLEICO. IGUALMENTE CON TRANSCERIPTOSA INVERSA SE COMPLETA UNA DOBLE HEBRA DE ACIDO DESOXI-ROBONUCLEICO. SE INCORPORA ESTA DOBLE HEBRA A UN PLASMIDO, POR EJEMPLO, EL PBR 322, REALIZANDOSE UN APARAMIENTO DE BASES ENTRE EL ACIDO DESOXIRIBONUCLEICO Y SE PLAQUEAN EN PLACAS DE AGAR Y SE SELECCIONAN EN CUANTO A LAS RESISTENCIAS A ANTIBIOTICOS. MEDIANTE ENSAYOS INMUNOLOGICOS SE SELECCIONAN LOS CLONES QUE EXPERIMENTAN PROTEINAS QUE CONTIENEN INSULINA. ESTOS CLONES SE CULTIVAN, EXTRAYENDOSE DE ELLOS EL CONTENIDO DE INSULINA.
