Abstract
A process for the production of antibiotic A16884 or a salt thereof, whose monoammonium salt is characterized as a white, amorphous substance, which decomposes at about 180ºC, very soluble in water, soluble in dimethyl sulfoxide, slightly soluble in lower alkanols and insoluble in acetonitrile; which is amphoteric, containing four valuable groups of pK'B1 = 3.9, pK'a2 = 5.3, pK'a3 = 9.2 and pK'a4 = 10.5, when titrated in 66% dimethylformamide at an initial pH of 5.8; that it has an apparent molecular weight of approximately 435, determined from the titration data; it has the following approximate composition: 44.01% carbon, 5.73% hydrogen, 10.65% nitrogen, 31.27% oxygen and 6.86% sulfur, which has a specific optical rotation, [ α] D25, +140.9 (c = 1% w/v in water); a suspension in mineral oil of which it has the following perceptible bands in its infrared absorption spectrum at the following wavelengths, expressed in microns: 3.18 (wide band), 5.66, 6.26 6.57, 6, 89, 7.15, 7.28, 7.40, 7.73, 8.00, 8.14, 8.79, 9.24, 9.65, 9.79 and 10.4; and that of positive tests with the reagents ninhidrina, Pan Dutsdher, Benedict, Molisch, iodine and dansyl chloride and negative tests with the reagents of Pehling, ferric chloride, biuret and Sakaguehi; the procedure of which is to cultivate an organism that is a variety of Strentomyces lipmanii NRRL 3584 that produces antibiotic A16884, in a culture medium that contains assimilable sources of carbon, nitrogen and inorganic salts, under submerged aerobic conditions, until a substantial amount is produced of A16884 by said organism in said culture medium.
