Abstract
Racemic and optically active compounds of the general formula (R = isopropyl or tert. -butyl R 1 = -(CH 2 )x- CN, -(CH 2 )x-NH 2 or -(CH 2 ) x+1 -OH (x = 0-3), -COOH or -COOCH 3 R 2 = C 1-4 alkoxy or alkylthio, C 2-4 alkenyl or alkynyl, CN or, when R 1 is not -CN or -NH 2 , H R 3 = H, halogen or C 1-4 alkyl or alkoxy) and their salts are prepared by (a) reacting a compound of the formula (Z = oxiranyl or 2-halo-1-hydroxyethyl) with RHNCONHR or RNH 2 , (b) alkylating a compound of the formula (c) hydrolysing a compound of the formula (G = hydrolysable group), (d) removing the protecting group in a compound of the formula (Sch = protecting group), (e) hydrolysing a compound of the formula (f) hydrolysing or pyrolysing a compound of the formula (R 4,5 = alkyl, aralkyl, aryl), (g) converting A to R 1 in a compound of the formula (A = group convertible to R 1 ), (h) converting B to R 2 in a compound of the formula (B = group convertible to R 2 ) or (i) halogenating or converting C to R 3 in a compound of the formula (C = H or group convertible to R 3 ), optionally followed in each case by salt formation and/or resolution. The preparation of 1 - (4-methoxycarbonyl phenoxy) - 2 - tetrahydropyranyloxy - 3 - isopropylaminopropane oxalate is described. The above compounds have #-adrenolytic and hypotensive activity, and may be administered in the form of pharmaceutical preparations containing them in association with a carrier. Reference has been directed by the Comptroller to Specification 1,084,793.
