Abstract

Novel substituted morphanthridines of the general formula wherein R is ethylene, trimethylene or propylene, R 1 is hydrogen, or C 1-3 alkyl, C 1-3 hydroxyalkyl, or C 2-3 alkanoyloxy-C 1-3 alkyl, or C 2-5 alkoxyalkyl group, R 2 is hydrogen or methyl, and R 3 is C 1-3 alkyl and physiologically acceptable acid addition salts thereof are prepared by any one of the following methods: (a) by heating a reaction mixture containing nitrilium and immonium cations of the formul[mu] with the appropriately substituted piperazine or homo-piperazine. The necessary cations are produced by intramolecular Ritter's reaction in 0- cyano-diphenylmethanes, by Beckmann's transformation of anthronosimes, or by Schmidt's reaction of anthrones with hydrozoic acid or as the dissociation products of the corresponding 6-mercapto-, 6-alkoxy-, 6-alkylthio-, or 6 - chloro - 8 - R 3 - morphanthridine, the latter is obtained from 5,6-dihydro-6-oxo-8-R 3 -morphanthridine, by reaction with a halogenating agent such as phosphorus oxychloride (b) by the dehydration of the corresponding 2-[(4-R 1 -1- piperazinyl (-homo - piperazinyl))] carboxyamido - 41 - R 3 - diphenylmethane (c) by ring closure of an acid amide or thioamide of the formula wherein Y is oxygen or sulphur, the amides being obtained by reduction of a suitable nitro compound, and the thioamide by treating the corresponding amide with phosphorus pentasulphide (d) by treating 6-amino-8-R3-morphanthridine with a reactive ester of an alcohol of formula the amines being prepared by treating corresponding cations (II) with ammonia (e) when R 1 is other than hydrogen, by reacting unsubstituted compound with a reactive ester of an alcohol of formula R 1 OH. Therapeutic compositions, having neuroplegic, neuroleptic and analgesic acitivity and suitable for oral or parenteral administration, contain the above novel morphanthridines or physiologically acceptable acid addition salts thereof as the active ingredient.
