Abstract
Un método para producir un producto alimenticio de base vegetal tratado térmicamente que comprende: (a) poner en contacto un material alimenticio de base vegetal con asparaginasa a una temperatura de 67-75 °C; (b) secar el material alimenticio de base vegetal a una temperatura del aire de 40 a 90°C; y (c) tratar térmicamente el material alimenticio de base vegetal tratado con asparaginasa para obtener el producto alimenticio de base vegetal tratado térmicamente; en donde la asparaginasa tiene una actividad residual después de 4 horas de incubación en agua desionizada con SAPP al 0,5% a 70°C, pH 5, de al menos 20%, con preferencia al menos 40%, más preferiblemente al menos 60%, todavía más preferiblemente al menos 80%, de la actividad sin tal incubación; y en donde la asparaginasa tiene una actividad a 35 °C, pH 6, de al menos 20%, preferiblemente al menos 30%, y más preferiblemente al menos 35%, de su actividad a 50°C, pH 6.

Comparación de la estabilidad térmica de las variantes termoestables y la enzima de tipo salvaje del tratamiento continuo de patatas fritas para mitigación de la acrilamida
Se pelaron manualmente patatas (Maris Piper o Bintje) para freír y se cortaron en patatas fritas (tamaño 0,8 x 0,8 x 5 cm) utilizando una cortadora de patatas fritas (Coupe Frites). Los palitos de patata de las patatas individuales se mezclaron y se mantuvieron en agua desionizada hasta su utilización. Porciones de 20 g de palitos de patata se escaldaron en dos pasos; primeramente a 85 °C durante 4 min (4 L de agua desionizada que se reutilizó) y subsiguientemente en 250 mL de agua desionizada a 70°C durante 15 min (agua nueva para cada muestra). El tratamiento con la enzima se realizó por mantenimiento de los palitos de patata escaldados durante 5 min a 70°C en 200 mL de solución de enzima (pirofosfato ácido de sodio al 0,5% en agua desionizada) dosificando 10.000 ASNU/L de las variantes o la enzima de tipo salvaje. Cada 5 mL se sumergió en el baño de enzima una nueva porción de palitos de patata a fin de mimetizar el uso continuo del baño de enzima. Se tomaron muestras del baño de enzima cada 30 min durante un periodo total de 4 horas y se congelaron para análisis posterior de la actividad.
Los resultados se muestran a continuación.

Como se ve por la tabla, todas las variantes exhiben un aumento significativo de estabilidad a 70°C comparadas con la enzima de tipo salvaje también en condiciones de uso continuo en un esquema de aplicación.
Ejemplo 3
Eficiencia de aplicación de las variantes termoestables en el tratamiento de patatas fritas para mitigación de la acrilamida

Porciones de 75 g de palitos de patata se escaldaron en dos pasos; primeramente a 85 °C durante 4 min (4 L de agua desionizada que se reutilizó) y subsiguientemente en 250 mL de agua desionizada a 70°C durante 15 min (agua nueva para cada muestra). El tratamiento con la enzima se realizó por inmersión de los palitos de patata escaldados durante 1 min a 70°C en 250 mL de solución de enzima (pirofosfato ácido de sodio al 0,5%, pH 5 en agua desionizada) utilizando una dosis de
10.000 ASNU/L de las variantes. Para comparación, se incluyó una muestra de control sumergida en SAPP al 0,5% sin enzima. El ensayo de la enzima de tipo salvaje se realizó a 55 °C para asegurar la actividad. Las muestras se prepararon por triplicado. Después del tratamiento con la enzima, los palitos de patata se secaron en un armario de calentamiento ventilado durante 10 min a 85 °C, 20% RH (humedad relativa), y se frieron parcialmente en aceite vegetal durante 1 min a 175 °C. Las muestras se congelaron rápidamente y se frieron finalmente por segunda vez durante 3 min a 175 °C.
Las patatas fritas se mezclaron y la acrilamida se extrajo utilizando acetonitrilo y un Extractor Automático con Disolvente (ASE de Dionex). El extracto se trató con solución Carrez I y II (la solución Carrez I contiene 15 g/100 mL de hexacianoferrato(II) de potasio trihidratado (K4[Fe(CN)6] x 3H2O); la solución Carrez II contiene 30 g/100 ml de sulfato de zinc heptahidratado (ZnSO4 x 7H2O)), se dejó durante una noche en el frigorífico y se filtró utilizando un filtro de 0,22 µm antes del análisis por HPLC (columna: Dionex IonPac ICE-AS1, 9x250 mm, eluyente: HCl 5 mM,
detección: UV a 202 nm). La acrilamida se identificó y se cuantificó por comparación con estándares conocidos.
Los resultados se dan a continuación.

Actividad a temperatura
20 Se diluyó enzima purificada con tampón de acetato de sodio 20 mM de pH 5,5 a una concentración aproximada de 2 rASNU/ml. Se añadieron 10 µL de solución de enzima a 100 µL de solución sustrato, que se ha pre-incubado a temperatura diferente, de ensayo rASNU en una placa PCR de 96 pocillos y se incubaron durante 20 min. Estabilidad térmica de la enzima en SAPP al 0,5%
5 La enzima purificada se diluyó con dihidrógeno-pirofosfato de sodio (SAPP) al 0,5% de pH 5,0 a una concentración aproximada de 6 rASNU/mL. Se transfirieron 50 µL de solución diluida de enzima a una placa PCR de 96 pocillos y se incubaron a 70°C-80° C durante 4 horas utilizando un ciclador térmico. Inmediatamente después de la incubación, se añadieron 10 µL de solución incubada a 100 µL de solución sustrato preincubada del ensayo rASNU en una placa
10 de 96 pocillos y se incubaron a 37 °C o 50°C durante 20 min. La reacción se paró por adición de 100 µL de solución de parada y se midió la absorbancia a 490 nm. La actividad residual se determinó utilizando la actividad de una muestra similar incubada a 25 °C como control.
