Abstract
La presente invención se refiere a polímeros proteicos o mezclas de los mismos que comprenden dominios capaces de promover la nucleación/crecimiento y/o adhesión de fosfatos de calcio de interés biológico, y dominios capaces de promover la adhesión celular a través de péptidos de las proteínas de la matriz extracelular, a procedimientos para activación de materiales para implantes óseos que emplean dichos polímeros proteicos y a los implantes obtenibles mediante dichos procedimientos.
Claims (17)
- Reivindicaciones1. Un polímero proteico o una mezcla de polímeros proteicos cuyos polímeros comprenden:i. uno o más dominios con la secuencia de aminoácidos (VPGX1G)n [(SEQ ID NO: 11)n] en la que n es un entero entre 1 y 50 y X1 es isoleucina,ii. uno o más dominios con la secuencia de aminoácidos (VPGX2G) (SEQ ID NO: 12) en la que X2 es lisina, yiii. uno o más dominios que se seleccionan entre:iiia. dominios que promueve(n) la nucleación/crecimiento y/o adhesión de fosfatos de calcio de interés biológico que comprenden la secuencia de aminoácidos DDDEEKFLRRIGRFG (SEQ ID NO: 16).iiib. dominios que promueve(n) la adhesión celular a través de péptidos de las proteínas de la matriz extracelular que comprenden la secuencia de aminoácidos RGD.con la condición que el polímero proteico o la mezcla de polímeros proteicos comprenden tanto el dominio (iiia) como el dominio (iiib).

- Polímero proteico según la reivindicación 1 en el que los dominios (iiia) y (iiib) están presentes simultáneamente en dicho polímero proteico.

- Mezcla de al menos dos tipos de polímeros proteicos según la reivindicación 1 en la que al menos uno de los polímeros proteicos (P1) comprende:
i. uno o más dominios con la secuencia de aminoácidos (VPGX1G)n [(SEQ ID NO: 11)n] en la que n es un entero entre 1 y 50 y X1 es isoleucina,ii. uno o más dominios con la secuencia de aminoácidos (VPGX2G) (SEQ ID NO: 12) en la que X2 es lisina, yiiia. uno o más dominios que promueve(n) la nucleación/crecimiento y/o adhesión de fosfatos de calcio de interés biológico que comprenden la secuencia de aminoácidos DDDEEKFLRRIGRFG (SEQ ID NO: 16);y al menos otro de los polímeros proteicos (P2) comprende:i. uno o más dominios con la secuencia de aminoácidos (VPGX1G)n [(SEQ ID NO: 11)n] en la que n es un entero entre 1 y 50 y X1 es isoleucina,ii. uno o más dominios con la secuencia de aminoácidos (VPGX2G) (SEQ ID NO: 12) en la que X2 es lisina, yiiib. uno o más dominios que promueve(n) la adhesión celular a través de péptidos de las proteínas de la matriz extracelular que comprenden la secuencia de aminoácidos RGD. -
- 4.
- Mezcla según la reivindicación 3 en la que únicamente hay un polímero proteico (P1) y un polímero proteico (P2).

- Polímero proteico o mezcla de polímeros proteicos según cualquiera de las reivindicaciones anteriores, en los que n es un entero entre 2 y 15.

- Polímero proteico o mezcla de polímeros proteicos según cualquiera de las reivindicaciones 1 a 4, en los que el polímero proteico comprende una o más secuencias de aminoácidos seleccionadas entre los que comprenden la secuencia [(VPGX1G)a VPGX2G (VPGX1G)b]c [(SEQ ID NO: 11)a (SEQ ID NO:12) (SEQ ID NO: 11)b]c en la que a, b y c son enteros entre 1 y 30 estando el número total de aminoácidos de dicha secuencia comprendido entre 2 y 500.

- Polímero proteico o mezcla de polímeros proteicos según cualquiera de las reivindicaciones 1 a 6 en los que el dominio (iiib) que promueve la adhesión celular comprende la secuencia de aminoácidos AVTGRGDSPASS (SEQ ID NO: 30).

- Polímero proteico según cualquiera de las reivindicaciones 1, 2, 5, 6 ó 7 de SEQ ID NO: 37 o mezcla de polímeros proteicos según cualquiera de las reivindicaciones 3 a 7 donde la secuencia de aminoácidos del polímero proteico (P1) es SEQ ID NO: 39 y la secuencia de aminoácidos del polímero proteico (P2) es SEQ ID NO: 41.

- Mezcla de polímeros proteicos según cualquiera de las reivindicaciones 1, 3, 5, 6 y 7 donde las secuencias de aminoácidos de dichos polímeros proteicos son SEQ ID NO: 37, SEQ ID NO: 39 y SEQ ID NO: 41.

- Un procedimiento para la bioactivación de materiales para implantes óseos seleccionados entre el grupo que comprende metales, aleaciones metálicas, materiales cerámicos y combinaciones de los mismos, que comprende las etapas de:
a) fijar moléculas de anclaje sobre la superficie activada químicamente del material de implante, presentando dichas moléculas de anclaje grupos funcionales que pueden unirse mediante enlace covalente con polímeros proteicosb) inmovilizar el polímero proteico o la mezcla de polímeros proteicos según las reivindicaciones 1 a 7 mediante dichos grupos funcionales sobre el material de implante. - 11. Procedimiento según la reivindicación 10 en el que el material para implante5 óseo se selecciona entre titanio y sus aleaciones metálicas, aleaciones basadas en Co-Cr o aceros inoxidables.

- Procedimiento según la reivindicación 11 en el que el material para implante óseo es titanio.

- Procedimiento según cualquiera de las reivindicaciones 10 a 12 caracterizado
10 por que las moléculas de anclaje que se fijan sobre la superficie del material de implante son silanos funcionalizados. - 14. Procedimiento según la reivindicación 13 caracterizado por que los silanos son trialcoxisilanos funcionalizados.

- Procedimiento según la reivindicación 14 caracterizado por que el silano es 315 cloropropiltrietoxisilano.
- 16. Procedimiento según la reivindicación 15 caracterizado por que la mezcla de polímeros proteicos está formada por los polímeros proteicos de SEQ ID NO: 37, SEQ ID NO: 39 y SEQ ID NO: 41.

- Implantes óseos obtenibles mediante el procedimiento de las reivindicaciones 20 10 a 16.
