Abstract

This invention describes a number of novel tacrine analogues, and, for the first time, the synthesis of 4H- pyrano[2,3-b]quinoline derivatives, a novel heterocyclic system, is described. This protocol is based on the Friedlander reaction between polyfunctionalized 2-amino-3- cyano-4H-pyrans and cycloalkanones, in the presence of Lewis acids. It is the first time that this reaction with polyfunctionalized 4H-pyrans has been described in the literature. The resulting 5-amino-4-aryl-3-ethoxycarbonyl- 2-methyl-6,7,8,9-tetrahydro-4H-pyrano[2,3-b]quinoline heterocycles were thus obtained in a single step, with a yield of about 60%. These compounds are novel potential neuroprotective agents for use in the treatment of Alzheimer's and Parkinson's diseases.
