Abstract

Ribosome inactivator protein known as nigrin f from the plant Sambucus nigra L., process for obtaining it and preparation Type 2 nigrin f (comprising two polypeptide chains) has a relative molecular mass of 58,000 daltons. There are two B chains and one A chain joined by disulphide bridges which in the presence of a reducer have an Mr of 31,600 and 30,000 daltons for the B chains, and 26,200 and 28,200 for the A chain. It has the following sequence of amino acids at its amine terminal end: Chain A Ile-Asp-Tyr-Pro-Ser-Val-Ser-Phe-Asn-Leu-Ala-Gly-Ala-Xaa- Ser-Ala-Thr-Tyr-Arg-Asp-Phe-Leu-Ser-Asn. Chain B Asp-Gly-Glu-Thr-Xaa-Pro-Ile-Pro-Ala-Ser-Phe-Thr-Arg-Xaa- Ile-Val-Gly-Arg-Asp-Gly-Leu-Xaa-Val-Asp. The general process for obtaining RIPs from Sambucus nigra L. according to this invention comprises isolating these from the fruits of Sambucus nigra L., including a first extraction operation using an aqueous solution based on sodium chloride and monosodium phosphate to obtain an extract which is capable of inhibiting protein synthesis, followed by purification of this using affinity, molecular exclusion and ion exchange chromatography techniques. Nigrin f is used as an inactivator of ribonucleic acid and an inhibitor of protein biosynthesis.
