Abstract
Ribosome inactivator proteins known as Alpha-nigritin, Beta-nigritin and Gamma-nigritin, from the plant Sambucus nigra L., process for obtaining them and their use Alpha-nigritin, Beta-nigritin and Gamma-nigritin have relative molecular masses of 29,000, 40,000 and 27,500 daltons respectively. Alpha-nigritin has the following amino acid sequence at its amine terminal end: Ala-Ser-Val-Leu-Asp-Ser-Ile-Gln-Phe-Val-Ser-Leu-Glu-Lys- Xxx-Ala-Val-Tyr-Glu-Gln-Asn. Gamma-nigritin has three tryptic peptides with the following amino acid sequences: a) Val-Thr-Asn-Ser-Phe-Tyr-Leu b) Tyr-Xxx-Pro-Ser-Ile-Ile-Arg c) Trp-Xxx-Asn-Thr-His-Ser-Tyr-Leu-Asn-Glu-Ile-Phe-Ile- Xxx-Leu. The general process for obtaining RIPs from Sambucus nigra according to this invention comprises isolating these from the leaves of Sambucus nigra including a first extraction operation with an aqueous solution based on sodium chloride and monosodium phosphate to obtain an extract which is capable of inhibiting protein synthesis, followed by purification of the extract using ion exchange and molecular exclusion chromatography techniques. Alpha-nigritin, Beta-nigritin and Gamma-nigritin are used as ribonucleic acid inactivators and as inhibitors of protein biosynthesis.
