- composition according to the invention furthermore necessarily contains a special combination of the three enzymes lipase, amylase and mannanase.
- lipase a special combination of the three enzymes lipase, amylase and mannanase.
- variant is at the level of proteins the term corresponding to "mutant” at the level of the nucleic acids.
- the precursor or parent molecules may be wild-type enzymes, that is, those obtainable from natural sources. They may also be enzymes which in themselves already represent variants, that is to say have already been changed with respect to the wild-type molecules. These include, for example, point mutants, those with changes in the amino acid sequence, multiple positions or longer contiguous regions, or else hybrid molecules composed of complementary sections of various wild-type enzymes.
- amino acid substitutions are meant substitutions of one amino acid against another amino acid. According to the invention, such substitutions are indicated by designation of the positions in which the exchange takes place, if appropriate combined with the relevant amino acids in the internationally used single-letter code.
- position 320 replacement means that a variant in position having position 320 in the sequence of a reference protein has a different amino acid.
- exchanges are carried out at the DNA level via mutations of single base pairs (see above).
- R320K means that the reference enzyme at position 320 has the amino acid arginine, while the variant under consideration has the amino acid lysine at the homologizable position.
- 320K means that any, that is usually a naturally given, amino acid at a position corresponding to position 320 is replaced with a lysine, which is located in that molecule in the same place.
- R320K, L means that the amino acid arginine at position 320 is replaced with lysine or leucine.
- R320X means that the amino acid arginine at position 320 is replaced with an essentially any other amino acid.
- amino acid substitutions according to the invention designated by the present application are not limited to being the only substitutions in which the variant in question differs from the wild-type molecule. It is known in the art that the beneficial properties of single point mutations can complement each other. Thus, embodiments of the present invention include all variants which, in addition to other exchanges with the wild-type molecule, also comprise the exchanges according to the invention.
- nucleic acid or amino acid sequences are carried out according to the invention by a sequence comparison. Such a comparison is made by assigning similar sequences in the nucleotide sequences or amino acid sequences to each other. This sequence comparison is preferably carried out based on the BLAST algorithm established and commonly used in the prior art (cf., for example, US Pat Altschul, SF, Gish, W., Miller, W., Myers, EW & Lipman, DJ (1990) "Basic local alignment search tool.” J. Mol. Biol. 215: 403-410 , and Altschul, Stephan F., Thomas L. Madden, Alejandro A. Schaffer, Jinghui Zhang, Hheng Zhang, Webb Miller, and David J.
- T-Coffee cf., for example Notredame et al. (2000): T-Coffee: A novel method for multiple sequence alignments. J. Mol. Biol. 302, 205-217 ) or programs based on these programs or algorithms.
- Clustal cf., for example, Chenna et al. (2003): Multiple sequence alignment with the Clustal series of programs.
- T-Coffee cf., for example Notredame et al. (2000): T-Coffee: A novel method for multiple sequence alignments. J. Mol. Biol.
- sequence comparisons and alignments are preferably created with the computer program Vector NTI® Suite 10.3 (Invitrogen Corporation, 1600 Faraday Avenue, Carlsbad, California, USA) with the default parameters specified.
- Such a comparison allows a statement about the similarity of the compared sequences to each other. It is usually given in percent identity, that is, the proportion of identical nucleotides or amino acid residues at the same or in an alignment corresponding positions.
- the broader concept of homology involves conserved amino acid substitutions in the consideration of amino acid sequences, that is, amino acids with similar chemical activity, as these usually perform similar chemical activities within the protein. Therefore, the similarity of the compared sequences may also be given in percent homology or percent similarity.
- Identity and / or Homology information can be found on whole polypeptides or genes or only on individual regions. Homologous or identical regions of different nucleic acid or amino acid sequences are therefore defined by matches in the sequences. Such areas often have identical functions.
- nucleic acid or amino acid sequence can be small and comprise only a few nucleotides or amino acids. Often, such small regions exert essential functions for the overall activity of the protein. It may therefore be useful to relate sequence matches only to individual, possibly small areas. Unless stated otherwise, identity or homology information in the present application, however, refers to the total length of the respectively indicated nucleic acid or amino acid sequence.
- Fragments are understood as meaning all polypeptides, proteins or peptides which are smaller than corresponding comparison proteins or those which correspond to completely translated genes and, for example, can also be obtained synthetically. Due to their amino acid sequences, they can be assigned to the respective complete comparison proteins. For example, they may adopt the same spatial structures or perform proteolytic or partial activities, such as the complexation of a substrate. Fragments and deletion variants of starting proteins are in principle similar; while fragments are rather small fragments, the deletion mutants tend to lack only short regions (possibly only one or more amino acids). Thus, for example, it is possible to delete further single amino acids at the termini or in the loops of the enzyme, without thereby losing or reducing the enzymatic activity.

- lipase-containing samples are added to an olive oil emulsion in emulsifier-containing water and incubated at 30 ° C and pH 9.0. This fatty acids are released. These are done with an autotitrator over 20 min. titrated continuously with 0.01 N sodium hydroxide solution so that the pH remains constant ("pH-stat titration"). Based on the sodium hydroxide consumption, the determination of the lipase activity takes place by reference to a reference lipase sample. Another suitable The method for measuring the lipase activity is the release of a dye from a suitable pNP-labeled substrate.

Description

J. Mol. Biol. 215:403-410 , und Altschul, Stephan F., Thomas L. Madden, Alejandro A. Schaffer, Jinghui Zhang, Hheng Zhang, Webb Miller, and David J. Lipman (1997): "Gapped BLAST and PSI-BLAST: a new generation of Protein database search programs"; Nucleic Acids Res., 25, S. 3389-3402) und geschieht prinzipiell dadurch, dass ähnliche Abfolgen von Nukleotiden oder Aminosäuren in den Nukleinsäure- bzw. Häufig genutzt werden beispielsweise die Clustal-Serie (vgl. beispielsweise Chenna et al. (2003): Multiple sequence alignment with the Clustal series of programs. Nucleic Acid Research 31, 3497-3500), T-Coffee (vgl. beispielsweise Notredame et al. (2000): T-Coffee: A novel method for multiple sequence alignments. J. Mol. Biol. 302, 205-217) oder Programme, die auf diesen Programmen bzw. Algorithmen basieren. Häufig genutzt werden beispielsweise Clustal (vgl. beispielsweise Chenna et al. (2003): Multiple sequence alignment with the Clustal series of programs. Nucleic Acid Research 31, 3497-3500) oder T-Coffee (vgl. beispielsweise Notredame et al. (2000): T-Coffee: A novel method for multiple sequence alignments. J. Mol. Biol.

J. Biochem. 1994, 223, 1-5; Eur. J. Biochem. 1995, 232, 1-6; Eur. J. Biochem. 1996, 237, 1-5; Eur. J. Biochem. 1997, 250, 1-6; and Eur. J. Biochem. 1999, 264, 610-650.

Gornall, C.S. Bardawill und M.M. David, J. Biol. Chem.
