Abstract

Die Erfindung betrifft ein Verfahren zum Verhindern von Luftblasen in einer im wesentlichen spaltförmigen Hybridisierkammer (5) eines Systems (1) zum Hybridisieren von auf Objektträgern (27) immobilisierten Nukleinsäureproben, Proteinen oder Gewebeschnitten. Dabei umfasst das Verfahren das Bereitstellen einer im Wesentlichen spaltförmigen Hybridisierkammer (5) zwischen einem Objektträger (27) und einem Deckel (26) (wobei der Deckel (26) derart gegenüber dem Objektträger (27) angeordnet wird, dass die Hybridisierkammer (5) gegenüber der Umgebungsluft abgedichtet ist) und das im wesentlichen Füllen der bereitgestellten Hybridisierkammer (5) mit einer Flüssigkeit. Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass es zudem das Ausbilden eines Kammer-Drucks in der Hybridisierkammer (5) mit einer Druckeinrichtung dieses Systems (1) und das Aufrechterhalten dieses Kammer-Drucks während eines Hybridisiervorgangs auf einem Wert, der mindestens 100 mbar über dem in der Umgebungsluft herrschenden, normalen atmosphärischen Druck liegt, umfasst.

The invention relates to a method for preventing air bubbles in a substantially slit-shaped hybridization chamber (5) of a system (1) for hybridizing nucleic acid samples, proteins or tissue sections immobilized on slides (27). The method comprises providing a substantially gap-shaped hybridization chamber (5) between a slide (27) and a lid (26) (wherein the lid (26) is positioned opposite the slide (27) such that the hybridization chamber (5) faces the ambient air is sealed) and substantially filling the provided Hybridisierkammer (5) with a liquid. The inventive method is characterized in that it further comprises forming a chamber pressure in the Hybridisierkammer (5) with a pressure device of this system (1) and maintaining this chamber pressure during a hybridization process at a value which is at least 100 mbar above the ambient atmospheric pressure prevails.
