Abstract
Die vorliegende Erfindung betrifft magnetische Nanopartikel, insbesondere Magnetit-Partikel mit einem Durchmesser zwischen 10 und 50 nm sowie ein Verfahren zu deren Herstellung.

Magnetische Nanopartikel und Verfahren zu deren Herstellung
Die vorliegende Erfindung betrifft magnetische Nanopartikel, insbesondere Magnetit-Partikel mit einem Durchmesser zwischen 10 und 50 nm sowie ein Verfahren zu deren Herstellung.
Magnetische Nanopartikel werden in einer Vielzahl unterschiedlicher Anwendungsfelder eingesetzt. Beispielsweise spielen sie in der Elektronik auf dem Gebiet der Informationsspeicherung und der Sensortechnik eine wichtige Rolle. Für Sicherheitsanwendungen, beispielsweise zur fälschungssicheren Codierung von Produkten, Dokumenten, Wertpapieren, usw. können magnetische Nanopartikel als sogenannte Security Marker verwendet werden. In Form von sogenannten Ferrofluiden - stabilen Flüssigdispersionen, deren Fließeigenschaften durch äußere Magnetfelder beeinflusst werden können - kommen magnetische Nanopartikel unter anderem beim Bau von Schwingungsdämpfern und Hochtonlautsprechern zum Einsatz. In der Molekularbiologie, Biochemie und Diagnostik werden magnetische Mikro- und Nanopartikel schon seit langem als selektive Sorbentien zur Abtrennung, Anreicherung oder Aufreinigung von biologischen Molekülen wie Nukleinsäuren oder Proteinen angewendet. Ein sehr großes Potential wird magnetischen Materialien in der Medizin vorausgesagt. Bereits jetzt werden magnetische Nanopartikel als Kontrastmittel in der Magnetresonanztomographie (MRI) verwendet. Weitere potenzielle Einsatzmöglichkeiten sind magnetisches drug targeting, und die Hyperthermiebehandlung von erkranktem Gewebe, beispielsweise x
Tumoren.
Für die beschriebenen Anwendungen werden in der Regel Kompositpartikel eingesetzt, die aus einer magnetischen Komponente (ferro- oder ferrimagnetische Nanopartikel) bestehen und einer funktionellen Komponente, z.B. einer Schicht aus Siliciumdioxid oder einem anderen Metalloxid wie etwa Aluminiumoxid, Titandioxid, oder Zirkondioxid, einem Polymer wie Polystyrol, Polymethacrylat, Polyvinylalkohol oder Polysiloxan,
oder einer natürlichen Komponente wie Dextran oder Chitosan. Zur chemischen Anknüpfung von Wirkstoffmolekülen, Antikörpern, Oligonukleotiden, usw. kann diese Beschichtung mit Reaktivgruppen, Ladungsträgern oder ähnlichen Funktionalitäten modifiziert sein.
Für die meisten der oben beschriebenen Einsatzfelder kommen bevorzugt Eisenoxide wie Maghämit (γ -Fe2Oa) oder Magnetit (Fe3Ü4) in Frage. Vor allem für Anwendungen in der Biotechnologie und der Medizin werden Eisenoxide wegen ihrer nicht-toxischen Eigenschaften anderen ferro- oder ferrimagnetischen Materialien wie Barium-, Zink- oder Cobaltferrit, elemetarem Cobalt etc. vorgezogen. Darüber hinaus sind Maghämit und Magnetit in nanopartikulärer Form besonders einfach durch eine
Fällungsreaktion herzustellen. So wird in fast allen einschlägigen
Publikationen und Patenten Magnetit durch Ausfällen aus einer stark alkalischen Lösung von Fe(II) und Fe(lll)-salzen im stöchiometrischen Verhältnis 1 :2 hergestellt. Das Verfahren wurde von Massart beschrieben (Massart, IEEE Trans. Magn. 1981 , MAG-17, 1247) und ist mit Abstand das am häufigsten verbreitete Verfahren zur Herstellung von
Magnetitnanopartikeln. Die Reaktionsbedingungen (Temperatur,
Konzentrationen, Reaktionsdauer, Art der Lauge, usw.) können in weiten Bereichen variiert werden, obwohl dadurch in Bezug auf Ausbeute, Produktreinheit und Partikelgröße kaum Änderungen bewirkt werden. Die auf diese Weise hergestellten Partikel haben stets einen sehr kleinen
Durchmesser (7 - 10 nm). Eine nachträgliche Oxidation des Magnetits liefert Maghämit, das ähnliche magnetische Eigenschaften aufweist.
Die sehr geringe Partikelgröße von <10 nm führt dazu, dass das Eisenoxid superparamagnetisch ist, d.h. dass es seine ferrimagnetischen Eigenschaften nur in Gegenwart eines äußeren Magnetfeldes zeigt und keine magnetische Remanenz besitzt. Dies ist ein generelles Phänomen aller ferri- und ferromagnetischen Materialien bei hinreichend geringen
Partikelgrößen. Es resultiert daher, dass die Partikelgröße in derselben Größenordnung liegt wie die Weiss'schen Bezirke, die als kleinste elementarmagnetische Domänen angesehen werden können. Im Fall von Magnetit liegt diese Größenordnung bei 20 - 30 nm. Magnetitnanopartikel mit wesentlich größerem Durchmesser sind also nicht mehr superparamagnetisch. Superparamagnetismus ist in der Mehrzahl der oben beschriebenen Anwendungen - mit Ausnahme der Informationsspeicherung - eine wünschenswerte oder sogar unabdingbare Eigenschaft, da Nanopartikel mit Remanenzmagnetismus als kleine Permanentmagnete wirken und sich aufgrund der magnetischen
Anziehungskräfte zusammenballen würden. Insbesondere bei in vivo Anwendungen wie MRI, drug targeting oder Hyperthermie könnte ein solches Verhalten sogar lebensbedrohliche Auswirkungen haben.
Trotz der Einfachheit seiner Herstellung und des Superparamagnetismus hat das über das Massart-Verfahren und seiner Varianten gewonnene Magnetit einige Nachteile. So entstehen meist auch andere, nichtmagnetische Eisenoxide als Nebenprodukt, wie z.B. α -Fβ2θ3 (Hämatit) oder FeOOH (Goethit). Diese lassen sich nur schwer abtrennen und führen häufig dazu, dass der Überstand einer magnetisch sedimentierten Partikelsuspension bräunlich-gelb gefärbt ist. Weiterhin sind derart kleine Magnetitpartikel aufgrund der hohen spezifischen Oberfläche wesentlich empfindlicher gegenüber Luftsauerstoff und neigen daher in nicht inerter Umgebung stärker zur Oxidation als größere Partikel. Der Hauptnachteil ist jedoch die aus der geringen Partikelgröße resultierende schlechte makroskopische Magnetisierbarkeit, die sich darin äußert, dass die Sedimentation der Partikel in einem Magnetfeld sehr lange dauert. Zum einen kann diese schlechte Abtrennbarkeit auf die geringe Sättigungsmagnetisierung zurückgeführt werden, die aus der geringen Ausdehnung der elementarmagnetischen Domänen resultiert. Zum Zweiten ist der Sedimentationsprozess überlagert von der Brownschen Molekularbewegung, die mit abnehmender Partikelgröße immer deutlicher

in Erscheinung tritt. Zur Abhilfe könnten stärkere homogene Magnetfelder eingesetzt werden oder spezielle Hochfeldgradientenabscheider, die beispielsweise für den Einsatz in der Abwasserreinigung konstruiert wurden. Für medizinische Anwendungen sind solche Konzepte allerdings nicht oder nur eingeschränkt verwendbar.
Die Auswirkung der schlechten Magnetisierbarkeit sind z.B. lange Prozesszeiten für .Separationen, geringe Heizleistung bei Hyperthermieanwendungen und schlechte Dosisverfügbarkeit bei drug Q targeting.
Es wäre daher für viele Anwendungen vorteilhaft, Partikel einzusetzen, die größer und damit besser magnetisierbar sind. Dies ist jedoch mit der beschriebenen Fällungsmethode nicht machbar. Obwohl mit einer speziellen Variante des Massart-Verfahrens Partikel mit einer '° durchschnittlichen Größe von 20 nm erzeugt werden konnten (Goetze et al., J. Magn. Magn. Mater. 252, 359, 2002), ist diese Methode jedoch nicht besonders vorteilhaft, da die Größenverteilung der erzeugten Teilchen sehr stark schwankt (Polydispersität). o Als Alternative zum Massart-Verfahren hat sich bislang nur das erstmals von Sugimoto und Matijevic beschriebene Oxidationsverfahren durchgesetzt (Sugimoto et al., J. Colloid Interface Sei. 74, 227, 1979). Hierbei wird keine stöchiometrische Mischung aus Fe(II) und Fe(III) eingesetzt, sondern nur eine Fe(ll)-salzlösung. Dabei wird in alkalischem 5 Medium aus einer Fe(ll)-Salzlösung zunächst dunkelgrünes Fe(OH)2 ausgefällt (sog. „green rust"), das anschließend durch ein zugesetztes Oxidationsmittel in der Hitze zu sehr reinem kristallinen Magnetit oxidiert wird. Als Oxidationsmittel wird in der Regel Nitrat eingesetzt, es können jedoch prinzipiell auch andere Oxidationsmittel wie Luftsauerstoff 0 verwendet werden. Die Partikel, die mit dieser Methode gewonnen werden, können durch geeignete Wahl der Reaktionsbedingungen in ihrer Größe in gewissen Grenzen variieren. Sie sind mit durchschnittlich 50 - 200 nm
allerdings wesentlich größer als die bisher beschriebenen und damit auch nicht superparamagnetisch. Da sie extrem gut magnetisierbar sind, eignen sie sich hervorragend für Adsorptions- und Bioseparationsaufgaben, bei denen ein 100%iger Superparamagnetismus nicht notwendig ist.
Auch zahlreiche weitere Syntheseansätze wurden in der Literatur beschrieben. So ist es beispielsweise möglich, magnetische Eisenoxidnanopartikel durch laserinduzierte Pyrolyse von Eisenpentacarbonyldämpfen in Ethylen zu synthetisieren (Veintemillas- Verdaguer et al., Mater. Lett. 35, 227, 1998). Andere Autoren beschreiben die partielle Reduktion von Fβ2θ3 Nanopartikeln mit Wasserstoff. Diese Methoden haben sich jedoch bislang nicht auf breiter Ebene durchgesetzt, da sie erheblich komplizierter in der Durchführung sind.
Bislang gibt es keine Methode, die es erlaubt, Eisenoxid-Nanopartikel monodispers in einem variablen Größenordnungsbereich zwischen 10 und 50 nm herzustellen. Insbesondere der Größenbereich 20-40 nm ist mit den bisherigen Verfahren nach Massart und Matijevic nicht zugänglich. Gerade dieser Größenordnungsbereich, in dem der Übergang von Remanenz zum Superparamagnetismus erfolgen sollte, ist aber äußerst interessant. Es wird erwartet, dass die Magnetisierbarkeit und damit die für Hyperthermieanwendung so wichtige Heizleistung im magnetischen Wechselfeld durch den Einsatz größerer Partikel wesentlich erhöht werden kann, ohne auf nicht-superparamagnetische Teilchen zurückgreifen zu müssen.
Aufgabe der vorliegenden Erfindung war es daher, ein Verfahren zur Verfügung zu stellen, mit dem Nanopartikel in einem variablen Größenordnungsbereich zwischen ca. 10 und 50 nm einfach und möglichst monodispers hergestellt werden können.
Es wurde gefunden, dass Nanopartikel in einem variablen Größenordnungsbereich zwischen ca. 10 und 50 nm hergestellt werden können, wenn eine Mischung aus mindestens einem Fe(II)-SaIz und mindestens einem Fe(III)-SaIz unter alkalischen Bedingungen oxidiert werden. Nach den Erfahrungen aus den bekannten Verfahren, bei denen entweder eine Mischung aus Fe(ll)-und Fe(lll)-Salzen eingesetzt wird (Massart-Verfahren) oder ein Oxidationsmittel eingesetzt wird (Verfahren nach Matijevic et al.) war zu erwarten, dass eine Kombination zu völlig unkontrollierbaren Reaktionsbedingungen führen würde. Überraschend stellte sich jedoch heraus, dass gerade diese Kombination ein effizientes und sehr leicht kontrollierbares Verfahren zur Herstellung von Nanopartikeln zur Verfügung stellt.
Gegenstand der vorliegenden Erfindung ist daher ein Verfahren zur Herstellung von Nanopartikeln, gekennzeichnet durch zumindest folgende Verfahrensschritte: a) Herstellung einer basischen Mischung enthaltend mindestens ein Me(II)-SaIz, ein Me(III)-SaIz und ein Oxidationsmittel, wobei das molare Verhältnis von Me(II) zu Me(III) in der Mischung zwischen 100:1 und 1 :1 ,5 liegt, Me(II) ausgewählt ist aus der Gruppe Fe(II), Co(II), Cr (II) und/oder Mn(II) und Me(III) ausgewählt ist aus der Gruppe Fe(III), Co(III), Cr(III) und/oder Mn(III); b) Optional Waschen und/oder Isolieren des entstandenen Niederschlags der Nanopartikel
In einer bevorzugten Ausführungsform wird die Mischung in einem Verfahrensschritt a2) nach ihrer Herstellung für mindestens eine Minute, typischerweise bei einer Temperatur zwischen 0 und 1000C temperiert.
In einer bevorzugten Ausführungsform enthält die in Verfahrensschritt a) hergestellte Mischung mindestens ein Fe(II)-SaIz und mindestens ein Fe(III)-SaIz.
In einer besonders bevorzugten Ausführungsform enthält die in Verfahrensschritt a) hergestellte Mischung Fe(ll)sulfat und Fe(lll)nitrat.
In einer bevorzugten Ausführungsform wird die Mischung in Schritt a) erzeugt, indem eine Lösung zumindest enthaltend ein Me(II)-SaIz und ein Me(III)-SaIz gemischt wird mit einer basischen Lösung, die mindestens ein Oxidationsmittel enthält.
In einer bevorzugten Ausführungsform wird als Oxidationsmittel in Schritt a) ein Nitratsalz wie Kaliumnitrat, Natriumnitrat oder Ammoniumnitrat eingesetzt.
In einer bevorzugten Ausführungsform wird in Schritt a2) zwischen 10 Minuten und 4 Stunden bei einer Temperatur zwischen 60 und 900C temperiert.
In einer Ausführungsform des erfindungsgemäßen Verfahrens wird die Oberfläche der erhaltenen Nanopartikel in einem zusätzlichen Verfahrensschritt ganz oder teilweise beschichtet oder derivatisiert.
Gegenstand der vorliegenden Erfindung sind auch Nanopartikel herstellbar nach dem erfindungsgemäßen Verfahren.
In einer bevorzugten Ausführungsform haben die Nanopartikel einen Durchmesser zwischen 10 und 50 nm.
Gegenstand der vorliegenden Erfindung ist auch die Verwendung der erfindungsgemäßen Nanopartikel in der in der Elektronik z.B. auf dem Gebiet der Informationsspeicherung und der Sensortechnik, als Security Marker, als Ferrofluide z.B. beim Bau von Schwingungsdämpfern und Hochtonlautsprechern, in der Molekularbiologie, Biochemie oder Diagnostik
z.B. als Sorbentien zur Abtrennung, Anreicherung oder Aufreinigung von biologischen Molekülen wie Nukleinsäuren oder Proteinen sowie in der Medizin z.B. in der Magnetresonanztomographie (MRI) für magnetisches drug targeting und zur Hyperthermiebehandlung von erkranktem Gewebe.
Die oben geschilderten Ausführungsformen der vorliegenden Erfindung können miteinander kombiniert werden, so dass Ausführungsformen entstehen, die die Merkmale zweier oder mehrerer der genannten Ausführungsformen aufweisen.
Nanopartikel im Sinne der vorliegenden Erfindung sind Partikel, die einen Durchmesser unter 80 nm, bevorzugt einen Durchmesser zwischen 10 und 50 nm, aufweisen.
Die erfindungsgemäß hergestellten Nanopartikel sind typischerweise polyedrisch, z.B. oktaedrisch. Der Durchmesser eines Nanopartikels wird erfindungsgemäß so bestimmt, dass eine Kugel um einen Partikel gelegt wird, auf deren Oberfläche möglichst viele Ecken des Partikels liegen. Der Durchmesser dieser hypothetischen Kugel entspricht dann dem Durchmesser des Partikels.
Bei dem erfindungsgemäßen Verfahren zur Herstellung der Nanopartikel wird eine basische Mischung erzeugt, die zumindest ein Me(II)-SaIz und ein Me(III)-SaIz enthält sowie mindestens ein Oxidationsmittel. Ein Me(II)-SaIz ist erfindungsgemäß ein Salz, das zumindest ein Metallion in der
Oxidationsstufe (II) enthält. Ein Me(III)-SaIz ist erfindungsgemäß ein Salz, das zumindest ein Metallion in der Oxidationsstufe (III) enthält.
Als Me(II)-SaIz können beispielsweise Fe(II)-, Co(II)- Cr(II)- oder Mn(II)- Salze oder Mischungen davon eingesetzt werden.
Als Me(III)-SaIz können beispielsweise Fe(III)-, Co(III)-, Cr(III)- oder Mn(III)- Salze oder Mischungen davon eingesetzt werden.
Bevorzugt weisen Me(II)-SaIz und Me(III)-SaIz die gleiche Metall- Komponente auf. Besonders bevorzugt werden ein oder mehrere Fe(II)- Salze und ein oder mehrere Fe(lll)-Salze eingesetzt.
Bevorzugte Fe(ll)-Salze sind Fe(ll)sulfat, Fe(ll)halogenide, insbesondere Fe(ll)chlorid, Fe(I I Perchlorat, Fe(ll)nitrat, Fe(ll)carbonat, Fe(ll)phosphat, Fe(ll)arsenat, Fe(ll)oxid, Fe(ll)hydroxid, Fe(ll)thiocyanat, Fe(ll)acetylacetonat, sowie die Fe(ll)-Salze organischer Säuren, insbesondere Fe(ll)formiat, Fe(ll)acetat, Fe(ll)citrat, Fe(ll)oxalat,
Fe(ll)fumarat, Fe(ll)tartrat, Fe(ll)gluconat, Fe(ll)-succinat, Fe(ll)-Iactat,. Besonders bevorzugt ist Fe(ll)sulfat. Die Salze können neben Fe weitere Kationen enthalten, wie z.B. Ammonium, Natrium oder Kalium.
Bevorzugte Fe(lll)-Salze sind Fe(lll)nitrat, Fe(lll)sulfat, Fe(lll)halogenide, insbesondere Fe(lll)chlorid, Fe(lll)perchlorat, Fe(lll)phosphat, Fe(lll)arsenat, Fe(lll)oxid, Fe(lll)hydroxid, Fe(lll)thiocyanat, Fe(lll)acetylacetonat, sowie die Fe(lll)salze organischer Säuren, insbesondere Fe(lll)formiat, Fe(lll)acetat, Fe(lll)citrat, Fe(lll)oxalat, Fe(ll)fumarat, Fe(ll)tartrat, Fe(ll)gluconat, Fe(ll)-succinat, Fe(ll)-Iactat.
Besonders bevorzugt ist Fe(lll)nitrat. Die Salze können neben Fe weitere Kationen enthalten, wie z.B. Ammonium, Natrium oder Kalium.
Die Me(II)- und Me(lll)-Salze werden in der Mischung typischerweise in einer Konzentration zwischen 0, 1 mmol/l und 5 mol/l eingesetzt. Bei höheren Konzentrationen kann es durch die ausfallenden Nanopartikel zum
Verstopfen der Apparaturen kommen, weil durch den hohen Anteil der
Nanopartikel die Mischung nicht mehr ausreichend fließfähig ist.
Bevorzugte Konzentrationsbereiche liegen für Me(ll)-Salze zwischen 0,05 mol/l und 0,2 mol/l, für Me(lll)-Salze zwischen 0, 5 mmol und 0,1 mol/l, wobei jeweils das molare Verhältnis von Me(II) zu Me(III) in der Mischung zwischen 100:1 und 1 :1 ,5 liegen muss.
Zur Herstellung der Mischung können die Me(ll)-Salze und die Me(III)- Salze zusammen oder getrennt eingebracht werden. Die Me-SaIz- Lösungen sollten vor Herstellung der Mischung nicht basisch sein, da sich sonst Me-Hydroxide bilden und ausfallen könnten. Bevorzugt liegt der pH-
Wert der Me-Salz-Lösungen vor der Mischung zwischen 0 und 7.
Als Oxidationsmittel können alle Oxidationsmittel eingesetzt werden, die geeignet stöchiometrisch dosiert werden können. Da dies bei Luftsauerstoff häufig problematisch ist, wird dieser bevorzugt nicht als Oxidationsmittel eingesetzt sondern beispielsweise durch Entgasung der eingesetzten Lösungen mit Stickstoff oder Edelgasen weitgehend eliminiert. Geeignete Oxidationsmittel sind beispielsweise Wasserstoffperoxid, anorganische Peroxoverbindungen wie beispielsweise Peroxide, Hydroperoxide, Peroxodisulfate, Peroxomonosulfate, Peroxoborate, Peroxochromate,
Peroxophosphate, Peroxocarbonate, organische Peroxoverbindungen wie beispielsweise Acetonperoxid oder Peroxocarbonsäuren, Chloramin T, Chlorate, Bromate, lodate, Perchlorate, Perbromate, Periodate, Permanganate, Chromate, Dichromate, Hypochlorite, Chloroxide oder Nitrate. Besonders bevorzugt werden als Oxidationsmittel erfindungsgemäß Nitrate, wie Kaliumnitrat, Natriumnitrat oder Ammoniumnitrat eingesetzt.
Die Menge an Oxidationsmittel richtet sich typischerweise nach der Menge des zu oxidierenden Metallsalzes. Ist der Me(lll)-Salz-Anteil sehr gering, so wird das Oxidationsmittel bevorzugt in ca. äquimolarer Menge zum Me(II)- SaIz eingesetzt. Liegt ein hoher Anteil an Me(III)-SaIz vor, so reduziert sich der Anteil an Oxidationsmittel entsprechend. Bei Verwendung milder Oxidationsmittel wie Nitrat kann das Oxidationsmittel auch im Überschuss eingesetzt werden, ohne dass eine Weiteroxidation des Fällungsprodukts erfolgt.
Das Oxidationsmittel kann zur Herstellung der Mischung einzeln zugesetzt werden oder vorab mit den Me-Salzen oder der Base gemischt werden.
Die in dem erfindungsgemäßen Verfahren hergestellte Mischung muss basisch sein, d.h. einen pH-Wert > 7 aufweisen, damit die gebildeten
Nanopartikel ausgefällt werden. Bevorzugt liegt der pH-Wert der Mischung zwischen pH 9 und 13, besonders bevorzugt zwischen pH 11 und 12. Dazu wird der Mischung in der Regel mindestens eine Base zugesetzt, die den pH-Wert der Mischung schnell entsprechend alkalisch macht. Geeignet sind alle starken Basen, wie z.B. Alkali- oder Erdalkalihydroxide, Amine oder Ammoniak. Bevorzugt wird als Base Natriumhydroxid eingesetzt.
Die basische Mischung enthaltend mindestens ein Me(II)-SaIz, ein Me(III)- SaIz und ein Oxidationsmittel, wobei das molare Verhältnis von Me(II) zu Me(III) in der Mischung zwischen 100:1 und 1:1 ,5 liegt, Me(II) ausgewählt ist aus der Gruppe Fe(II), Co(II), Cr(II) und/oder Mn(II) und Me(III) ausgewählt ist aus der Gruppe Fe(III), Co(III), Cr(III) und/oder Mn(III), kann auf jede Weise hergestellt werden, insbesondere so, dass sichergestellt ist, dass die Me-Salze vorher keinen Kontakt zu einer Base oder basischen Lösung haben. Typischerweise werden jedoch um eine besonders schnelle Durchmischung aller Komponenten zu erzielen, alle Komponenten zuvor in Lösung gebracht. Dazu wird bevorzugt eine Lösung enthaltend mindestens ein Me(II)-SaIz und ein Me(III)-SaIz (Me-Salz-Lösung) hergestellt und eine basische Lösung. Das Oxidationsmittel kann der Me-Salz-Lösung oder bevorzugt der basischen Lösung zugesetzt werden.
Als Lösungsmittel dient typischerweise Wasser oder Mischungen von Wasser mit wasserlöslichen organischen Lösungsmitteln. Bevorzugt wird als Lösungsmittel Wasser eingesetzt. Weiterhin können die Lösungen Zusätze, wie z.B. oberflächenaktive Substanzen, enthalten.
Um die Eigenschaften der Nanopartikel zusätzlich zu beeinflussen, können der Mischung weiterhin andere Bestandteile, wie z.B. andere Metallsalze, Metalle, Pigmente oder organische Verbindungen, zugesetzt werden.
Die Mischung der einzelnen Komponenten (Me-Salze, Oxidationsmittel,
Base) erfolgt dann durch Mischung der Lösungen, in denen die Komponenten enthalten sind. Dies kann bei einem Batch-Verfahren in einem Rührkessel oder einer entsprechenden Apparatur erfolgen. Bevorzugt erfolgt die Mischung in einem kontinuierlichen Verfahren. Dazu werden typischerweise zwei Flüssigkeitsströme erzeugt, die ineinander geführt und so gemischt werden. Dies kann z.B. unter Einsatz eines statischen Mischers oder bevorzugt eines einfachen T- oder Y-Stücks erfolgen. Die Flussraten liegen bei der kontinuierlichen Mischung typischerweise zwischen 50 und 150 mL/min, können aber je nach Dimensionierung der Anlage beliebig vergrößert oder verringert werden.
Die Herstellung der Mischung erfolgt typischerweise bei Temperaturen zwischen 0 und 100 0C, bevorzugt zwischen 10 und 600C, besonders bevorzugt bei Raumtemperatur.
Durch die Herstellung der basischen Mischung enthaltend mindestens ein Me(II)-SaIz, ein Me(III)-SaIz und ein Oxidationsmittel, wobei das molare Verhältnis von Me(II) zu Me(III) in der Mischung zwischen 100:1 und 1 :1 ,5 liegt, Me(II) ausgewählt ist aus der Gruppe Fe(II), Co(II), Cr(II) und/oder Mn(II) und Me(III) ausgewählt ist aus der Gruppe Fe(III), Co(III), Cr(III) und/oder Mn(III), bildet sich ein Niederschlag der Nanopartikel. Nach der Herstellung der Mischung wird diese bevorzugt temperiert, um die Fällung der Nanopartikel und deren Größenwachstum zu vervollständigen.
Das Temperieren kann im Batch-Verfahren im Rührkessel oder einer entsprechenden anderen temperierbaren Apparatur erfolgen. Bei einer kontinuierlichen Verfahrensführung wird die Mischung beispielsweise in ein
Temperier-Gefäß (z.B. einen temperierbaren Rührkessel) oder bevorzugt in eine Verweilschleife überführt. Durch Länge und Durchmesser sowie die Fließgeschwindigkeit kann die Verweildauer in Verweilschleife genau festgelegt werden und ist für die gesamte Mischung gleich.
Das Temperieren erfolgt typischerweise bei Temperaturen zwischen 0 und 100 0C, bevorzugt zwischen 20 und 1000C1 besonders bevorzugt zwischen 60 und 900C.
Die Temperierdauer beträgt in Abhängigkeit von der Temperatur (niedrigere Temperaturen bedingen eine längere Temperierdauer) typischerweise zwischen einer Minute und einem Tag, bevorzugt zwischen 10 Minuten und 4 Stunden, besonders bevorzugt zwischen 20 Minuten und einer Stunde.
Nach dem Temperieren können die erhaltenen Nanopartikel optional gewaschen, filtriert, zentrifugiert oder auf andere Weise aufgereinigt oder isoliert werden. Typischerweise wird mehrfach mit entsalztem Wasser gewaschen.
Genauso können die Nanopartikel direkt oder nach Waschen bzw. Isolierung weiteren Derivatisierungsreaktionen unterzogen werden. Dabei werden die Partikel beispielsweise ganz oder teilweise mit einer Schicht aus Siliciumdioxid (WO 98/31461) oder einem anderen anorganischen Oxid (EP 0343934), einem Polymer wie beispielsweise Polystyrol (US 4421660), Polymethacrylat (Horäk et al. Biotechnol. Prog. 17, 447, 2001), Polyvinylalkohol (WO 97/04862) oder Polysiloxan (US 4985166) oder einer natürlichen Komponente wie Dextran (US 4452773) oder Chitosan (US 5864025) belegt. Zur chemischen Anknüpfung von Wirkstoffmolekülen, Antikörpern, Oligonukleotiden, usw. kann diese Beschichtung mit Reaktivgruppen, Ladungsträgern oder ähnlichen
Funktionalitäten modifiziert sein (Liu et al. Colloids & Surfaces A 238, 127,
2004). Auch das Aufbringen von Pfropfpolymerisaten z.B. entsprechend EP 0 337 144, WO 98/01480 oder WO 2000/011043 ist möglich.
Auch eine direkte Derivatisierung der Nanopartikel mit funktionellen
Molekülen, wie z.B. Silanen, die Reaktivgruppen, Ladungsträger oder 5 ähnliche Funktionalitäten tragen, ist möglich (Kobayashi et al. J. Colloid
Interface Sei. 141 , 505, 1991 ). Auch kann die Aggregationsneigung der Nanopartikel z.B. durch die Adsorption oberflächenaktiver Moleküle reduziert werden (Badescu et al. J. Magn. Magn. Mater. 202, 197, 1999).

Bevorzugt werden mit dem erfindungsgemäßen Verfahren Magnetit-Partikel hergestellt. Diese weisen je nach ihrer Größe superparamagnetische Eigenschaften auf. Eine nachträgliche Oxidation der Magnetit-Nanopartikel liefert Maghämit-Partikel, die ähnliche magnetische Eigenschaften aufweisen. (Cornell, Schwertmann, The Iron Oxides, Wiley-VCH, 2003, S. 402)
Die nach dem erfindungsgemäßen Verfahren erhaltenen Nanopartikel haben typischerweise einen Durchmesser von < 80 nm; bevorzugt haben sie einen Durchmesser zwischen 10 und 50 nm. Da die Nanopartikel durch die erfindungsgemäße Herstellung schon sehr monodispers erhalten werden, ist zumeist keine weitere Klassierung der Partikel notwendig.
Durch die Wahl des Molverhältnisses zwischen Me(II)- und Me(lll)-Salzen,
O(_ kann die Größe der erhaltenen Nanopartikel in sehr engem Bereich Zo eingestellt werden. Je größer der prozentuale Anteil an Me(III) zu dem Gesamtmetallgehalt gewählt wird, desto kleiner werden die erhaltenen Nanopartikel.
Gegenstand der vorliegenden Erfindung ist auch die Verwendung der erfindungsgemäßen Nanopartikel in der in der Elektronik z.B. auf dem Gebiet der Informationsspeicherung und der Sensortechnik, als Security
marker, als Ferrofluide z.B. beim Bau von Schwingungsdämpfern und Hochtonlautsprechern, in der Molekularbiologie, Biochemie oder Diagnostik z.B. als Sorbentien zur Abtrennung, Anreicherung oder Aufreinigung von biologischen Molekülen wie Nukleinsäuren oder Proteinen sowie in der Medizin z.B. in der Magnetresonanztomographie (MRI) für magnetisches drug targeting und zur Hyperthermiebehandlung von erkranktem Gewebe.
Auch ohne weitere Ausführungen wird davon ausgegangen, daß ein Fach-0 mann die obige Beschreibung im weitesten Umfang nutzen kann. Die bevorzugten Ausführungsformen und Beispiele sind deswegen lediglich als beschreibende, keineswegs als in irgendeiner Weise limitierende Offenbarung aufzufassen. 5 Die vollständige Offenbarung aller vor- und nachstehend aufgeführten Anmeldungen, Patente und Veröffentlichungen, insbesondere der korrespondierenden Anmeldungen DE 102008015365.6, eingereicht am 20.03.2008 ist durch Bezugnahme in diese Anmeldung eingeführt. 0
Beispiele
Beispiel 1 : Vergleichsbeispiel: Kontinuierliche Herstellung von Magnetit-5 Nanopartikeln mit einer mittleren Partikelqröße von ca. 80 nm analog Suqimoto et aL J. Colloid Interface Sei. 74, 227. 1979
In zwei 2,5 L Glasflaschen werden jeweils 1 ,5 L VE-Wasser vorgelegt. Der im Wasser gelöste Luftsauerstoff wird durch Einleiten von StickstoffQ weitgehend ausgetrieben. Anschließend werden in dem einen
Vorlagegefäßi 38,2 g Eisen(ll)sulfat-Heptahydrat (497 mmol) Wasser gelöst, in dem zweiten 45,9 g Kaliumnitrat (454 mmol) und 56 g
Natriumhydroxid (1 ,4 mol). Die beiden Lösungen werden mit Hilfe zweier Büchi-Kolbenhubpumpen mit einer Flussrate von jeweils 100 mL/min in ein T-Stück gepumpt, welches als statischer Mischer dient. Die austretende Mischung wird in eine 200 m lange Verweilstrecke (Teflonschlauch mit einem Innendurchmesser von 4 mm) geleitet, welche in einem
Thermostaten auf 750C temperiert wird. Die Verweilzeit im Thermostaten beträgt demnach ca. 12 min. Die Mischung wird in einer 5 L Glasflasche aufgefangen und dort abkühlen gelassen. Zur Aufarbeitung wird der schwarze Niederschlag mittels eines Permanentmagneten (Bakker, 200 mT) sedimentiert und der klare
Überstand abgegossen. Das Präzipitat wird mit 3 L VE-Wasser aufgerührt und die Sedimentation auf die beschriebene Weise mehrfach wiederholt, bis die elektrische Leitfähigkeit des Überstands auf einen Wert von < 200 μS abgesunken ist. Anschließend wird das Produkt wie folgt charakterisiert: 1. Trocknen einer Probe von ca. 0,5 g im Vakuumtrockenschrank bei 1000C und Ermitteln der spezifischen Oberfläche Asp des Magnetits nach der Methode von Brunauer, Emmett und Teller (BET). Anhand der spezifischen Oberfläche und der spezifischen Dichte von Magnetit p = 5 g/mL kann näherungsweise der mittlere Partikeldurchmesser D errechnet werden: D = 6/(pAsp)
2. Anfertigen einer rasterelektronenmikroskopischen Aufnahme zur visuellen Charakterisierung.
3. Durchführung einer Magnetisierungsmessung mittels SQUID- Magnetometer.
Ergebnisse: spezif. Oberfläche: 15 m2/g errechnete mittlere Partikelgröße: 79 nm magnetische Remanenz: 25 emu/g
Da für die Fällung nur Eisen(ll)-salz eingesetzt worden war (0% Fe(III), entspricht das Produkt dem nach dem Matijevic-Verfahren hergestellten Magnetit.
Beispiel 2: Kontinuierliche Herstellung von Maqnetit-Nanopartikeln mit einer mittleren Partikelgröße von ca. 46 nm
Der in Beispiel 1 beschriebene Versuchsaufbau wird beibehalten. Es wird jedoch in das eine Vorlagegefäß neben 128,4 g Eisen(ll)-sulfat-Heptahydrat (462 mmol) 9,9 g Eisen(lll)-nitrat-Nonahydrat (24 mmol) eingewogen. In das zweite Vorlagegefäß werden anstatt 45,9 g nur 35 g Kaliumnitrat (346 mmol) eingewogen. Die Gesamtmenge an Eisenionen (Fe(II) + Fe(III)) bleibt insgesamt also gleich. Die Menge an Nitrat ist geringfügig reduziert, da etwas weniger Eisen oxidiert werden muss. Der prozentuale Anteil an Fe(III) in Relation zum Gesamteisengehalt liegt bei 5%. Die Durchführung des Versuchs erfolgt wie in Beispiel 1 beschrieben.
Ergebnisse: spezif. Oberfläche: 26 m2/g errechnete mittlere Partikelgröße: 46 nm magnetische Remanenz: 25 emu/g
Beispiel 3: Kontinuierliche Herstellung von Magnetit-Nanopartikeln mit einer mittleren Partikelgröße von ca. 28 nm
Durchführung wie in Beispiel 2 beschrieben, jedoch mit 124,3 g Eisen(ll)- sulfat-Heptahydrat (447 mmol), 20,1 g Eisen(lll)-nitrat-Nonahydrat (50 mmol) und 24,2 g Kaliumnitrat (239 mmol). Der prozentuale Anteil an Fe(III) in Relation zum Gesamteisengehalt liegt bei 10%.
Ergebnisse: spezif. Oberfläche: 43 m2/g errechnete mittlere Partikelgröße: 28 nm magnetische Remanenz: 6 emu/g
Beispiel 4: Kontinuierliche Herstellung von Magnetit-Nanopartikeln mit einer mittleren Partikelgröße von ca. 21 nm
Durchführung wie in Beispiel 2 beschrieben, jedoch mit 117,4 g Eisen(ll)- sulfat-Heptahydrat (422 mmol), 30,2 g Eisen(lll)-nitrat-Nonahydrat (75 mmol) und 13 g Kaliumnitrat (129 mmol). Der prozentuale Anteil an Fe(III) in Relation zum Gesamteisengehalt liegt bei 15%.
Ergebnisse: spezif. Oberfläche: 57 m2/g errechnete mittlere Partikelgröße: 21 nm magnetische Remanenz: 5 emu/g
Beispiel 5: Kontinuierliche Herstellung von Maqnetit-Nanopartikeln mit einer mittleren Partikelgröße von ca. 18 nm
Durchführung wie in Beispiel 2 beschrieben, jedoch mit 110,5 g Eisen(ll)- sulfat-Heptahydrat (397 mmol), 40,2 g Eisen(lll)-nitrat-Nonahydrat (99 mmol) und 2 g Kaliumnitrat (20 mmol). Der prozentuale Anteil an Fe(III) in Relation zum Gesamteisengehalt liegt bei 20%.

Durchführung wie in Beispiel 2 beschrieben, jedoch mit 104 g Eisen(ll)- sulfat-Heptahydrat (374 mmol), 50,2 g Eisen(lll)-nitrat-Nonahydrat (124 mmol) und 0 g Kaliumnitrat (0 mmol). Der prozentuale Anteil an Fe(III) in Relation zum Gesamteisengehalt liegt bei 25%.

Beispiel 8: Kontinuierliche Herstellung von Magnetit-Nanopartikeln mit einer mittleren Partikelgröße von ca. 11 nm
Durchführung wie in Beispiel 5 beschrieben, jedoch mit 90,1 g Eisen(ll)- sulfat-Heptahydrat (324 mmol) und 70,3 g Eisen(lll)-nitrat-Nonahydrat (174 mmol). Der prozentuale Anteil an Fe(III) in Relation zum
Gesamteisengehalt liegt bei 35%.

Claims
1. Verfahren zur Herstellung von Nanopartikeln, gekennzeichnet durch zumindest folgende Verfahrensschritte: a) Herstellung einer basischen Mischung enthaltend mindestens ein
Me(II)-SaIz, ein Me(III)-SaIz und ein Oxidationsmittel, wobei das molare Verhältnis von Me(II) zu Me(III) in der Mischung zwischen 100:1 und 1 :1 ,5 liegt, Me(II) ausgewählt ist aus der Gruppe Fe(II), Co(II), Cr(II) und/oder Mn(II) und Me(III) ausgewählt ist aus der Gruppe Fe(III), Co(III), Cr(III) und/oder Mn(III); b) Optional Waschen und/oder Isolieren der entstandenen Nanopartikel.
2. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass die Mischung in einem Verfahrensschritt a2) nach der Herstellung für mindestens eine Minute temperiert wird.
3. Verfahren nach einem der Ansprüche 1 oder 2, dadurch gekennzeichnet, dass die in Verfahrensschritt a) hergestellte Mischung mindestens ein Fe(II)-SaIz und mindestens ein Fe(III)-SaIz enthält.
4. Verfahren nach einem oder mehreren der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass die in Verfahrensschritt a) hergestellte Mischung Fe(ll)-sulfat und Fe(lll)nitrat enthält.
5. Verfahren nach einem oder mehreren der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass die Mischung in Schritt a) erzeugt wird, indem eine Lösung, die zumindest ein Me(II)-SaIz und ein Me(III)-SaIz enthält, gemischt wird mit einer basischen Lösung, die mindestens ein Oxidationsmittel enthält.
6. Verfahren nach einem oder mehreren der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass als Oxidationsmittel in Schritt a) ein Nitratsalz eingesetzt wird.
7. Verfahren nach einem oder mehreren der Ansprüche 2 bis 6, dadurch gekennzeichnet, dass in Schritt a2) zwischen 10 Minuten und 24 Stunden bei einer Temperatur zwischen 60 und 900C temperiert wird.
8. Verfahren nach einem oder mehreren der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass die Oberfläche der erhaltenen Nanopartikel in einem zusätzlichen Verfahrensschritt ganz oder teilweise beschichtet oder derivatisiert wird.
9. Nanopartikel herstellbar nach dem Verfahren entsprechend einem oder mehreren der Ansprüche 1 bis 8.
10. Nanopartikel nach Anspruch 9, dadurch gekennzeichnet, dass sie einen Durchmesser zwischen 10 und 50 nm haben.
11. Verwendung der Nanopartikel nach Anspruch 9 oder 10 in der
Elektronik, als Security marker, als Ferrofluide, in der Molekularbiologie, Biochemie oder Diagnostik, sowie in der Medizin.
