Abstract
Die Erfindung betrifft substituierte 3-Phenylpiperidin-Derivate, Verfahren zu deren Herstellung, Arzneimittel enthaltend diese Verbindungen sowie die Verwendung dieser Stoffe zur Herstellung von Arzneimitteln, vorzugsweise zur Behandlung von Schmerz.
Description
Substituierte 3-Phenylpiperidin-Derivate
Die Erfindung betrifft substituierte 3-Phenylpiperidin-Derivate, Verfahren zu deren Herstellung, Arzneimittel enthaltend diese Verbindungen sowie die Verwendung dieser Stoffe zur Herstellung von Arzneimitteln, vorzugsweise zur Behandlung von Schmerz.
Die Behandlung chronischer und nichtchronischer Schmerzzustände hat in der Medizin eine große Bedeutung. Es besteht ein weltweiter Bedarf an gut wirksamen Schmerztherapien. Der dringende Handlungsbedarf für eine patientengerechte und zielorientierte Behandlung chronischer und nicht chronischer Schmerzzustände, wobei hierunter die erfolgreiche und zufriedenstellende Schmerzbehandlung für den Patienten zu verstehen ist, dokumentiert sich in der großen Anzahl von wissen¬ schaftlichen Arbeiten, die auf dem Gebiet der angewandten Analgetik bzw. der Grundlagenforschung zur Nociception in letzter Zeit erschienen sind.
Klassische Opioide wie Morphin sind bei der Therapie starker bis stärkster Schmerzen gut wirksam. Ihr Einsatz wird jedoch durch die bekannten Nebenwirkungen, z. B. Atemdepression, Erbrechen, Sedierung, Obstipation und Toleranzentwicklung, limitiert. Außerdem sind sie bei neuropathischen oder inzidentiellen Schmerzen, unter denen insbesondere Tumorpatienten leiden, weniger wirksam.
Unter den Opioiden, die sich in der Behandlung von Schmerz in der Anwendung befinden, nimmt Tramadol eine Sonderstellung ein, da sich Tramadol in seinem Nebenwirkungsspektrum von den klassischen Opioiden unterscheidet (Analgesics - from Chemistry and Pharmacology to Clinical application, 228-230, Wiley 2002). Ausgehend von Tramadol wurde jetzt überraschenderweise gefunden, dass das Ersetzen der Cyclohexyldimethylamino-Gruppe durch ein am Stickstoff substituiertes Piperidin zu schmerzaktiven Substanzen führt.

Aus der Klasse der substituierten 3-Phenylpiperidin-Derivate ist bislang nur ein Derivat aus der Literatur bekannt. WO 0039091 offenbart 3-(3-Methoxyphenyl)-1- (phenylmethyl)-3-piperidinol als Zwischenprodukt bei der Synthese von Bisarylpiperidinen. In dieser Verbindung ist der Piperidin-Stickstoff mit einer Benzyl- Schutzgruppe versehen, wobei Funktionalisierungen der Benzylgruppe nicht beschrieben sind. Auch in DE 31497023 wird 3-(3-Methoxyphenyl)-1-(phenylmethyl)- 3-piperidinol als Zwischenprodukt genannt.
Aufgabe der Erfindung war es, neue Strukturen aufzufinden, die entsprechende physiologische Wirksamkeiten in Hinblick auf Analgesie aufweisen. Weitere Aufgaben bestanden darin, Wirkstoffe zu finden, die sich auch zur Verwendung als Antiarrhythmikum und/oder Antiemetikum und/oder Nootropikum (Neurotropikum) und/oder zur Behandlung/Therapie von cardiovaskulären Erkrankungen und/oder Harninkontinenz und/oder Diarrhöe und/oder Pruritus und/oder Alkohol- und/oder Drogen- und/oder Medikamentenabhängigkeit und/oder Entzündungen und/oder zur Behandlung von Depressionen und/oder zur Vigilanzsteigerung und/oder Libido- Steigerung eignen.
Gegenstand der Erfindung sind daher 3-Phenylpiperidin-Derivate der allgemeinen Formel I

R1 für C(O)R3 oder CH2R4 steht R2 für Ci-3-Alkyl oder H steht R3 für Aryl, unsubstituiert oder einfach oder mehrfach substituiert; Thiophenyl,
Benzothiophenyl, Benzofuranyl oder Furyl, unsubstituiert oder einfach oder mehrfach substituiert; über eine (CHR5)n-Gruppe oder eine (CH2)nO-Gruppe verbrücktes Aryl, Thiophenyl oder Furanyl, jeweils unsubstituiert oder einfach oder mehrfach substituiert, mit n = 1 , 2, 3; steht; R4 für einfach oder mehrfach substituiertes Phenyl; Naphthyl, Fluoranthenyl,
Fluorenyl, Tetralinyl, Indanyl oder Anthracenyl, unsubstituert oder einfach oder mehrfach substituiert; Thiophenyl, Benzothiophenyl, Benzofuranyl oder Furyl, unsubstituiert oder einfach oder mehrfach substituiert; über eine (CHR5)n- Gruppe oder eine (CH2)nO-Gruppe verbrücktes Aryl, Thiophenyl oder Furyl, jeweils unsubstituiert oder einfach oder mehrfach substituiert, mit n = 1 , 2, 3; steht;
R5 für H, d-3-Alkyl, Phenyl oder Benzyl, jeweils unsubstituiert oder einfach oder mehrfach substituiert, steht,
in Form des Razemats; der Enantiomere, Diastereomere, Mischungen der
Enantiomere oder Diastereomere oder eines einzelnen Enantiomers oder Diastereomers; der Basen und/oder Salze physiologisch verträglicher Säuren oder Kationen.
Bevorzugt im Sinne dieser Erfindung sind substituierte 3-Phenylpiperidin-Derivate, worin R3 für Aryl, unsubstituiert oder einfach oder mehrfach substituiert, Thiophenyl oder Furyl, unsubstituiert oder einfach oder mehrfach substituiert; über eine CHR5- Gruppe oder eine CH2O-Gruppe verbrücktes Aryl, unsubstituiert oder einfach oder mehrfach substituiert, steht.
Besonders bevorzugt sind substituierte 3-Phenylpiperidin-Derivate, worin R3 für Phenyl oder Naphthyl, unsubstituiert oder einfach oder mehrfach substituiert, unsubstituiertes Thiophenyl oder Furyl; über eine CHR5-Gruppe oder eine CH2O-
Gruppe verbrücktes Phenyl, unsubstituiert oder einfach oder mehrfach substituiert, steht.
Außerdem bevorzugt sind substituierte 3-Phenylpiperidin-Derivate, worin R4 für einfach oder mehrfach substituiertes Phenyl, Naphthyl, Fluorenyl, Indanyl oder Anthracenyl; Thiophenyl, oder Furyl, unsubstituiert oder einfach oder mehrfach substituiert; über eine CHR5-Gruppe oder eine CH2O-Gruppe verbrücktes Aryl, einfach oder mehrfach substituiert, steht.
Besonders bevorzugt sind substituierte 3-Phenylpiperidin-Derivate, worin R4 für einfach oder mehrfach substituiertes Phenyl, Naphthyl; Thiophenyl, oder Furyl, unsubstituiert; über eine CHR5-Gruppe oder eine CH2O-Gruppe verbrücktes Phenyl, einfach oder mehrfach substituiert, steht,
Weiterhin bevorzugt sind 3-Phenylpiperidin-Derivate, worin R2 für Methyl oder H steht.

Die erfindungsgemäßen Verbindungen weisen eine ausgeprägte Affinität zum L-Typ- Ca-Kanal auf. Sie sind daher wirksam gegen Schmerzen (Analgesics - from chemistry and pharmacology to clinical application, S. 353-378, Wiley 2002), verstärken den Effekt weiterer Analgetika (Methods Find. Exp. Clin. Pharmacol. 2000 Dec, 22, 741-745; lndian J. Exp. Biol. 2001 Jul;.39, 636-642) und eignen sich darüber hinaus auch zur Verwendung als Antiarrhythmikum und/oder Antiemetikum und/oder Nootropikum (Neurotropikum) und/oder zur Behandlung/Therapie von cardiovaskulären Erkrankungen und/oder Harninkontinenz und/oder Diarrhöe und/oder Pruritus und/oder Bluthochdruck und/oder Alkohol- und/oder Drogen- und/oder Medikamentenabhängigkeit und/oder Entzündungen und/oder zur

Der Ausdruck "C-j_3-Alky!" umfasst im Sinne dieser Erfindung acyclische gesättigte oder ungesättigte Kohlenwasserstoffreste, die verzweigt- oder geradkettig sowie unsubstituiert oder ein- oder mehrfach substituiert sein können, mit 1 , 2 oder 3 C- Atomen, d.h. C<|_3-Alkanyle, C2_3-Alkenyle und C2_3-Alkinyle. Dabei weisen Alkenyle mindestens eine C-C-Doppelbindung und Alkinyle mindestens eine C-C- Dreifachbindung auf. B. zwei- oder dreifach, substituiert sind. So ist es erfindungsgemäß möglich, dreifach am gleichen C-Atom wie im Falle von CF3 oder -CH2CF3 oder an verschiedenen Stellen wie im Falle von -CH(OH)-CH=CH-CHCI2 H ZU substituierten. Die Mehrfachsubstitution kann mit dem gleichen oder mit verschiedenen Substituenten erfolgen. Ggf. kann ein Substituent auch seinerseits substituiert sein; so umfaßt -OAlkyl u.a. auch -0-CH2-CH2-O-CH2- CH2-OH. Bevorzugt sind Substituenten ausgewählt aus der Gruppe F, Cl, Br, I, -CN, CF3, NH2, OH, COOH, =0, OCH3, NO2, COCH3, COOCH3, COOC2H5 oder N(CH3)2. Vorteilhaft ist Alkyl aus der Gruppe ausgewählt, die Methyl, Ethyl, n-Propyl, 2-Propyl umfaßt.
Der Ausdruck "Aryl" bedeutet im Sinne dieser Erfindung carbocyclische Ringsysteme mit mindestens einem aromatischen Ring, aber ohne Heteroatome in nur einem der Ringe, u.a. Phenyle, Naphthyle und Phenanthrenyle, Fluoranthenyle, Fluorenyle,
Indanyle und Tetralinyle. Die Aryl-Reste können auch mit weiteren gesättigten, (partiell) ungesättigten oder aromatischen Ringsystemen kondensiert sein. Jeder Aryl-Rest kann unsubstituiert oder einfach oder mehrfach substituiert vorliegen, wobei die Aryl-Substituenten gleich oder verschieden und in jeder beliebigen und möglichen Position des Aryls sein können. Besonders vorteilhaft sind Phenyl- oder Naphthyl-Reste.
Der Ausdruck "Heteroaryl" steht für einen 5-, 6- oder 7-gliedrigen cyclischen aromatischen Rest, der mindestens 1 , ggf. auch 2, 3, 4 oder 5 Heteroatome, enthält, wobei die Heteroatome gleich oder verschieden sind und der Heterocyclus unsubstituiert oder ein- oder mehrfach substituiert sein kann; im Falle der Substitution am Heterocyclus können die Substituenten gleich oder verschieden sein und in jeder beliebigen und möglichen Position des Heteroaryls sein. Der Heterocyclus kann auch Teil eines bi- oder polycyclischen Systems sein. Bevorzugte Heteroatome sind Stickstoff, Sauerstoff und Schwefel. Es ist bevorzugt, daß der
Heteroaryl-Rest ausgewählt ist aus der Gruppe, die Pyrrolyl, Indolyl, Furyl (Furanyl), Benzofuranyl, Thienyl (Thiophenyl), Benzothienyl, Benzothiadiazolyl, Benzothiazolyl, Benzotriazolyl, Benzodioxolanyl, Benzodioxanyl, Phtalazinyl, Pyrazolyl, Imidazolyl, Thiazolyl, Oxazolyl, Isoxazoyl, Pyridinyl, Pyridazinyl, Pyrimidinyl, Pyrazinyl, Pyranyl, Indazolyl, Purinyl, Indolizinyl, Chinolinyl, Isochinolinyl, Chinazolinyl, Carbazolyl, Phenazinyl, Phenothiazinyl oder Oxadiazolyl enthält, wobei die Bindung an die Verbindungen der allgemeinen Struktur I über jedes beliebige und mögliche Ringglied des Heteroaryl-Restes erfolgen kann.

Die Mehrfach¬ substitution erfolgt dabei mit dem gleichen oder mit unterschiedlichen Substituenten. Bevorzugt sind Substituenten ausgewählt aus der Gruppe F, Cl, Br, I, -CN, NH2, OH, COOH, OCF3, CF3, OCH3, NO2, OCOCH3, COOCH3, COOC2H5 oder N(CH3)2.
Unter dem Begriff Salz ist jegliche Form des erfindungsgemäßen Wirkstoffes zu verstehen, in dem dieser eine ionische Form annimmt bzw. geladen ist und mit einem Gegenion (einem Kation oder Anion) gekoppelt ist bzw. sich in Lösung befindet. Darunter sind auch Komplexe des Wirkstoffes mit anderen Molekülen und Ionen zu verstehen, insbesondere Komplexe, die über ionische Wechselwirkungen komplexiert sind. Insbesondere versteht man darunter (und dies ist auch eine bevorzugte Ausführungsform dieser Erfindung) physiologisch verträgliche Salze, insbesondere physiologisch verträgliche Salze mit Kationen oder Basen und physiologisch verträgliche Salze mit Anionen oder Säuren oder auch ein mit einer physiologisch verträglichen Säure oder einem physiologisch verträglichen Kation gebildetes Salz.
Unter dem Begriff des physiologisch verträglichen Salzes mit Anionen oder Säuren versteht man im Sinne dieser Erfindung Salze mindestens einer der erfindungsgemäßen Verbindungen - meist, vorzugsweise am Stickstoff, protoniert - als Kation mit mindestens einem Anion, die physiologisch - insbesondere bei Anwendung im Menschen und/oder Säugetier - verträglich sind. Insbesondere versteht man darunter im Sinne dieser Erfindung das mit einer physiologisch verträglichen Säure gebildete Salz, nämlich Salze des jeweiligen Wirkstoffes mit anorganischen bzw. organischen Säuren, die physiologisch - insbesondere bei Anwendung im Menschen und/oder Säugetier - verträglich sind. Beispiele für physiologisch verträgliche Salze bestimmter Säuren sind Salze der: Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Methansulfonsäure, Ameisensäure, Essigsäure, Oxalsäure, Bernsteinsäure, Apfelsäure, Weinsäure, Mandelsäure,
Fumarsäure, Milchsäure, Zitronensäure, Glutaminsäure, Saccharinsäure, Monomethylsebacinsäure, 5-Oxo-prolin, Hexan-1-sulfonsäure, Nicotinsäure, 2-, 3- oder 4-Aminobenzoesäure, 2,4,6-Trimethylbenzoesäure, α-Liponsäure, Acetylglycin, Phosphorsäure, Maleinsäure, Malonsäure, Hippursäure und/oder Asparaginsäure. Besonders bevorzugt ist das Hydrochlorid-Salz.
Unter dem Begriff des physiologisch verträglichen Salzes mit Kationen oder Basen versteht man im Sinne dieser Erfindung Salze mindestens einer der erfindungsgemäßen Verbindungen - meist einer (deprotonierten) Säure - als Anion mit mindestens einem, vorzugsweise anorganischen, Kation, die physiologisch - insbesondere bei Anwendung im Menschen und/oder Säugetier - verträglich sind. Besonders bevorzugt sind die Salze der Alkali- und Erdalkalimetalle aber auch Ammoniumsalze, insbesondere aber (Mono-) oder (Di-) Natrium-, (Mono-) oder (Di-) Kalium-, Magnesium- oder Calzium-Salze.
Die erfindungsgemäßen Substanzen eignen sich als pharmazeutische Wirkstoffe in Arzneimitteln. Ein weiterer Gegenstand der Erfindung sind daher Arzneimittel enthaltend wenigstens ein erfindungsgemäßes substituiertes 3-Phenylpiperidin- Derivat, sowie gegebenenfalls geeignete Zusatz- und/oder Hilfsstoffe und/oder gegebenenfalls weitere Wirkstoffe.
Die erfindungsgemäßen Arzneimittel enthalten neben mindestens einem erfindungsgemäßen substituierten 3-Phenylpiperidin-Derivat gegebenenfalls geeignete Zusatz- und/oder Hilfsstoffe, so auch auch Trägermaterialien, Füllstoffe, Lösungsmittel, Verdünnungsmittel, Farbstoffe und/oder Bindemittel und können als flüssige Arzneiformen in Form von Injektionslösungen, Tropfen oder Säfte, als halbfeste Arzneiformen in Form von Granulaten, Tabletten, Pellets, Patches, Kapseln, Pflaster oder Aerosolen verabreicht werden. Die Auswahl der Hilfsstoffe etc. sowie die einzusetzenden Mengen derselben hängen davon ab, ob das Arzneimittel oral, peroral, parenteral, intravenös, intraperitoneal, intradermal, intramuskulär, intranasal, buccal, rektal oder örtlich, zum Beispiel auf die Haut, die Schleimhäute oder in die Augen, appliziert werden soll. Für die orale Applikation
eignen sich Zubereitungen in Form von Tabletten, Dragees, Kapseln, Granulaten, Tropfen, Säften und Sirupen, für die parenterale, topische und inhalative Applikation Lösungen, Suspensionen, leicht rekonstituierbare Trockenzubereitungen sowie Sprays. Erfindungsgemäße 3-Phenylpiperidin-Derivate in einem Depot, in gelöster Form oder in einem Pflaster, gegebenenfalls unter Zusatz von die Hautpenetration fördernden Mitteln, sind geeignete perkutane Applikationszubereitungen. Oral oder perkutan anwendbare Zubereitungsformen können die erfindungsgemäßen 3- Phenylpiperidin-Derivate verzögert freisetzen. Prinzipiell können den erfindungsgemäßen Arzneimitteln andere dem Fachmann bekannte weitere Wirkstoffe zugesetzt werden.
Die an den Patienten zu verabreichende Wirkstoffmenge variiert in Abhängigkeit vom Gewicht des Patienten, von der Applikationsart, der Indikation und dem Schweregrad der Erkrankung. Üblicherweise werden 0,005 bis 1000 mg/kg, bevorzugt 0,05 bis 5 mg/kg wenigstens eines erfindungsgemäßen 3-Phenylpiperidin-Derivats appliziert.
In einer bevorzugten Form des Arzneimittels liegt ein enthaltenes erfindungsgemäßes 3-Phenylpiperidin-Derivat als reines Enantiomer, als Razemat oder als nicht-äquimolare oder äquimolare Mischung der Enantiomere vor. Grundsätzlich ist es auch möglich, wenn ein Substituent ein oder mehrere weitere Stereozentren enthält, reine Diastereomere oder nicht-äquimolare oder äquimolare Mischung der Diastereomeren einzusetzen.
Ein weiterer Gegenstand der Erfindung ist die Verwendung eines erfindungs- gemäßen 3-Phenylpiperidin-Derivats zur Herstellung eines Arzneimittels zur Behandlung von Schmerz, insbesondere von akutem, neuropathischem oder chronischem Schmerz.
Ein weiterer Gegenstand der Erfindung ist die Verwendung eines erfindungs- gemäßen 3-Phenylpiperidin-Derivats in Kombination mit einem weiteren
Analgetikum, beispielsweise einem Opioid, zur Herstellung eines Arzneimittels zur
Behandlung von Schmerz, insbesondere von akutem, neuropathischem oder chronischem Schmerz.
Überraschenderweise hat sich gezeigt, dass die substituierten 3-Phenylpiperidin- Derivate der allgemeinen Formel I sich zur Verwendung als Antiarrhythmikum, Anti- emetikum, Nootropikum (Neurotropikum), zur Behandlung/Therapie von cardio- vaskulären Erkrankungen, Harninkontinenz, Diarrhöe, Pruritus, Bluthochdruck, Alkohol- und/oder Drogen- und/oder Medikamentenabhängigkeit, Entzündungen, zur Behandlung von Depressionen, zur Vigilanzsteigerung und Libidosteigerung eignen.
Gegenstand der Erfindung ist daher auch die Verwendung eines substituierten 3- Phenylpiperidin-Derivats der allgemeinen Formel I zur Herstellung eines Arzneimittels zur Verwendung als Antiarrhythmikum, Antiemetikum, Nootropikum (Neurotropikum), zur Behandlung/Therapie von cardiovaskulären Erkrankungen, Harninkontinenz, Diarrhöe, Pruritus, Bluthochdruck, Alkohol- und/oder Drogen- und/oder Medikamentenabhängigkeit, Entzündungen, zur Behandlung von Depres¬ sionen, zur Vigilanzsteigerung und Libidosteigerung.
Besonders bevorzugt werden die erfindungsgemäßen substituierten 3- Phenylpiperidin-Derivate, die zur Herstellung eines Arzneimittels zur Behandlung von Schmerz, insbesondere von akutem, neuropathischem oder chronischem Schmerz, von Depressionen, zur Behandlung von cardiovaskulären Erkrankungen, Harninkontinenz, Diarrhöe, Pruritus, Bluthochdruck, Alkohol- und Drogenmißbrauch, Medikamentenabhängigkeit, Entzündungen, zur Verwendung als Antiarrythmikum, Antiemetikum, Nootropikum oder zur Vigilanz- und Libidosteigerung verwendet werden, ausgewählt aus folgender Gruppe:

Ein weiterer Gegenstand der Erfindung sind Verfahren zur Herstellung der erfindungsgemäßen 3-Phenylpiperidin-Derivate.
Ib Ia
Zur Herstellung der erfindungsgemäßen 3-Phenylpiperidin-Derivate wird N-Benzyl- Piperidon mit einem 3-Alkyloxyphenylhalogenid und einem Metall oder einer metallorganischen Verbindung, beispielsweise Magnesium oder n-BuLi, in einem geeigneten Lösungsmittel, beispielsweise Diethylether oder THF, zu einer 1-Benzyl- 3-(3-alkoxy-phenyl)-piperidin-3-ol-Verbindung umgesetzt. Die Benzylgruppe wird durch Hydrierung an einem Katalysator, beispielsweise Pd/C, entfernt. Durch Umsetzung mit Säurechloriden der allgemeinen Formel R3(CO)CI unter Zugabe von Base, beispielsweise Triethylamin, erhält man erfindungsgemäße Verbindungen der allgemeinen Formel Ia, die durch Reduktion der Ketogruppe, beispielsweise mit Lithiumaluminiumhydrid in Gegenwart von Aluminiumtrichlorid, zu Verbindungen der allgemeinen Formel Ib umgesetzt werden können.

Die folgenden Beispiele dienen zur näheren Erläuterung der Erfindung, schränken aber den allgemeinen Erfindungsgedanken nicht ein.
Die Ausbeuten der hergestellten Verbindungen sind nicht optimiert.
Als stationäre Phase für die Säulenchromatographie wurde Kieselgel 60 (0.040 - 0.063 mm) der Firma E. Merck, Darmstadt, eingesetzt.
Die dünnschicht-chromatographischen Untersuchungen wurden mit HPTLC- Fertigplatten, Kieselgel 60 F 254, der Firma E. Merck, Darmstadt, durchgeführt.
Die Mischungsverhältnisse von Laufmitteln für chromatographische Untersuchungen sind stets in Volumen/Volumen angegeben.
1-Benzyl-3-(3-methoxy-phenyl)-piperidin-3-ol
In einem Dreihalskolben mit Kühler, Innenthermometer und Tropftrichter wurden unter Stickstoffatmosphäre 4,22 g Mg in 50 ml THF und einem Körnchen I2 vorgelegt. Unter Rührung wurde auf 500C erwärmt und innerhalb von 30 min 22,1 ml 3-Bromanisol in 50 ml THF zugetropft. Anschließend wurde weitere 30 min bei 700C gerührt. Nach Abkühlung mit Eis auf 5°C wurden 22,34 g N-Benzyl-Piperidon in 100 ml THF in 60 min zugetropft. Bei Raumtemperatur wurde 15 h weitergerührt. Zur Hydrolyse wurden 400 ml 10 % NH4HSO4-Lösung zugegeben, mit verdünnter
HCl angesäuert und anschließend mit 100 ml Diethylether extrahiert. Die organische Phase wurde einmal mit verdünnter HCl extrahiert. Die vereinigten wäßrigen Phasen wurden mit NaOH alkalisch gestellt und dreimal mit je 100 ml Diethylether extrahiert. Die vereinigten organischen Phasen wurden eingeengt. Ausbeute 35,05 g
3-(3-Methoxy-phenyl)-piperidin-3-ol
32,41 g 1-Benzyl-3-(3-methoxy-phenyl)-piperidin-3-ol wurde in Methanol gelöst und mit 12,4 g 32% HCI-Lösung auf pH 6 gestellt. Unter Stickstoffatmosphäre wurden 3,3 g Pd/C 10% (in gekühltem Methanol) zugegeben. Bei 5 bar H2-Druck wurde über Nacht hydriert. Anschließend wurde der Katalysator über Filtererde abgetrennt und das Filtrat eingeengt. Der ölige Rückstand wurde in H2O aufgenommen, mit NaOH alkalisch gestellt und dreimal mit Diethylether extrahiert. Die vereinigten organischen Phasen wurden über MgSO4 getrocknet und eingeengt.
Ausbeute: 20,92 g
1-Benzyl-3-[3-(tetrahydropyran-2-yloxy)-phenyl]-piperidin-3-ol
In einem Dreihalskolben mit Kühler, Innenthermometer und Tropftrichter wurden unter Stickstoffatmosphäre 0,583 g Mg in 40 ml THF und einem Kömchen I2 vorgelegt. Unter Rührung wurde auf 500C erwärmt und innerhalb von 30 min 5,14 g 2-(3-Bromphenoxy)-tetrahydropyran in 50 ml THF zugetropft. Anschließend wurde weitere 60 min zum Rückfluß erhitzt. Nach Abkühlung 15°C wurden 4,83 g N-Benzyl- Piperidon in THF zugetropft. Bei Raumtemperatur wurde 15 h weitergerührt. Zur Hydrolyse wurden 50 ml gesättigte NH4HSO4-Lösung zugegeben, die wäßrige Phase wurde dreimal mit Ethylacetat extrahiert und die organischen Phasen wurden eingeengt. Ausbeute: 9,24 g
1 -Benzyl-3-(3-hydroxy-phenyl)-piperidin-3-ol
8,64 g 1-Benzyl-3-[3-(tetrahydropyran-2-yloxy)-phenyl]-piperidin-3-ol wurden in 30 ml
Ethanol gelöst und mit 8,9 ml HCl versetzt. Nach 60 h Rühren wurde das
Lösungsmittel entfernt.
Ausbeute: 6 g
3-(3-Hydroxy-phenyl)-piperidin-3-ol
2,7 g 1-Benzyl-3-(3-hydroxy-phenyl)-piperidin-3-ol wurden in Methanol gelöst. Unter
Stickstoffatmosphäre wurden 2,5 g Pd/C 10% (in gekühltem Methanol) zugegeben.
Bei 5 bar H2-DmCk wurde über Nacht hydriert. Anschließend wurde der Katalysator über Filtererde abgetrennt und das Filtrat eingeengt.
Ausbeute: 1 ,61 g
Allgemeine Vorschrift A:
Zur Herstellung der Amide wurden 14,47 mmol eines entsprechenden Säurechlorids in 10 ml Dichlormethan gelöst. Nach Zugabe von 19,2 mmol Triethylamin und einer Spatelspitze DMAP (N,N-Dimethylaminopyridin) wurden unter Eiskühlung 9,6 mmol des Amins in Dichlormethan zugetropft. Das Reaktionsgemisch wurde 15 h bei Raumtemperatur gerührt. Anschließend wurde das Gemisch mit 20ml NH4CI versetzt und dreimal mit Dichlormethan extrahiert. Nach Trocknung über MgSO4 wurde das
Lösungsmittel entfernt und das Produkt durch Säulenchromatographie gereinigt (Kieselgel, n-Hexan/Ethylacetat 1 :10).
Allgemeine Vorschrift B: Zur Reduktion der Ketogruppe wurden 0,452 g Lithiumaluminiumhydrid unter
Stickstoffatmosphäre in 50 ml THF vorgelegt und langsam 0,528 g AICb zugegeben.
Nach 45 min Rühren wurden 3,3 mmol des entsprechenden Amids in 50 ml THF zugegeben. Das Reaktionsgemisch wurde 3 h refluxiert, mit 50 ml verdünnter NaOH versetzt und weitere 15 h gerührt. Das Gemisch wurde dreimal mit Diethylether extrahiert, die organischen Phasen über MgSO4 getrocknet und das Lösungsmittel entfernt.
Zur Fällung des HCI-Salzes wurde das Produkt in 25 ml Ether aufgenommen und mit
0,386 ml Trimethylsilylchlorid versetzt. Nach 30 min Rühren im Eisbad wurde das
Produkt abgesaugt.

Beispiel 55 (4-Fluorphenyl)-[3-hydroxy-3-(3-hydroxyphenyl)-piperidin-1-yl]-methanon
Beispiel 55 wurde gemäß der allgemeinen Vorschrift A aus 4-Fluorbenzoylchlorid und 3-(3-Hydroxyphenyl)-piperidin-3-ol hergestellt.
Biologische Daten:
Bindungsuntersuchungen am L-Kalziumkanal Benzothiazepin-Bindungsstelle (Diltiazem-Bindung) Das biologische Membranmaterial wurde aus dem Cerebrocortex der Ratte isoliert. Als Ligand wurde [3H]-cis-(+)-Diltiazem (5 nM im Ansatz) verwendet. Inkubation für 20 Minuten bei 25 0C. Als unspezifische Bindung ist die Radioaktivität definiert, die bei Anwesenheit von (±)-Diltiazem (10"^ M im Ansatz) gemessen wird. Der nicht gebundene Anteil des radioaktiven Liganden wird nach Abschluß der Inkubation mit Hilfe eines Filtrationsprozesses über Whatman Glasfiber GF/B-Membranen abge¬ trennt. Die Membranen werden anschließend nach einem Waschprozeß am ß- Counter vermessen. Die Methode ist in Anlehnung an die Veröffentlichung von Schoemaker und Langer (H. Schoemaker und S.Z. Langer (1985) Eur. J. Pharmacol. 111. 273-277) erstellt worden. Der KD-Wert für diese hoch affine Bindungsstelle wurde mit 4,10 ± 0,75 nM bestimmt (N = 3, d.h. Mittelwerte ± SEM aus 3 unabhängigen Versuchsreihen, die in Dreifach-Parallelversuchen durchgeführt worden sind).
Claims
1. Substituierte 3-Pheny!piperidin-Derivate der allgemeinen Formel I
worin
R1 für C(O)R3 oder CH2R4 steht
R2 für d-3-Alkyl oder H steht
R3 für Aryl, unsubstituiert oder einfach oder mehrfach substituiert; Thiophenyl,
Benzothiophenyl, Benzofuranyl oder Furyl, unsubstituiert oder einfach oder mehrfach substituiert; über eine (CHR5)n-Gruppe oder eine (CH2)nO-Gruppe verbrücktes Aryl, Thiophenyl oder Furanyl, jeweils unsubstituiert oder einfach oder mehrfach substituiert, mit n = 1 , 2, 3; steht;
R4 für einfach oder mehrfach substituiertes Phenyl; Naphthyl, Fluoranthenyl,
Fluorenyl, Tetralinyl, Indanyl oder Anthracenyl unsubstituiert oder einfach oder mehrfach substituiert; Thiophenyl, Benzothiophenyl, Benzofuranyl oder Furyl, unsubstituiert oder einfach oder mehrfach substituiert; über eine (CHR5)n-
Gruppe oder eine (CH2)nO-Gruppe verbrücktes Aryl, Thiophenyl oder Furyl, jeweils unsubstituiert oder einfach oder mehrfach substituiert, mit n = 1 , 2, 3; steht;
R5 für H, Ci-3-Alkyl, Phenyl oder Benzyl, jeweils unsubstituiert oder einfach oder mehrfach substituiert, steht,
in Form des Razemats; der Enantiomere, Diastereomere, Mischungen der Enantiomere oder Diastereomere oder eines einzelnen Enantiomers oder Diastereomers; der Basen und/oder Salze physiologisch verträglicher Säuren oder Kationen.
2. Substituierte 3-Phenylpiperidin-Derivate gemäß Anspruch 1 , dadurch gekennzeichnet, dass R1 für C(O)R3 steht.
3. Substituierte 3-Phenylpiperidin-Derivate gemäß Anspruch 1 , dadurch gekennzeichnet, dass R1 für CH2R4 steht.
4. Substituierte 3-Phenylpiperidin-Derivate gemäß einem der Ansprüche 1 oder 2, dadurch gekennzeichnet, dass R3 für Aryl, unsubstituiert oder einfach oder mehrfach substituiert, Thiophenyl oder Furyl, unsubstituiert oder einfach oder mehrfach substituiert; über eine CHR5-Gruppe oder eine CH2O-Gmppe verbrücktes Aryl, unsubstituiert oder einfach oder mehrfach substituiert, steht.

Substituierte 3-Phenylpiperidin-Derivate gemäß einem der Ansprüche 1 oder 2, dadurch gekennzeichnet, dass worin R3 für Phenyl oder Naphthyl, unsubstituiert oder einfach oder mehrfach substituiert, unsubstituiertes Thiophenyl oder Furyl; über eine CHR5-Gruppe oder eine CH2O-Gruppe verbrücktes Phenyl, unsubstituiert oder einfach oder mehrfach substituiert, steht.

Substituierte 3-Phenylpiperidin-Derivate gemäß einem der Ansprüche 1 oder 3, dadurch gekennzeichnet, dass R4 für einfach oder mehrfach substituiertes Phenyl, Naphthyl, Fluorenyl, Indanyl oder Anthracenyl; Thiophenyl, oder Furyl, unsubstituiert oder einfach oder mehrfach substituiert; über eine CHR5-Gruppe oder eine CH2O-Gruppe verbrücktes Aryl, einfach oder mehrfach substituiert, steht.

Substituierte 3-Phenylpiperidin-Derivate gemäß einem der Ansprüche 1 oder 3, dadurch gekennzeichnet, dass R4 für einfach oder mehrfach substituiertes Phenyl oder Naphthyl; Thiophenyl oder Furyl, unsubstituiert; über eine CHR5-Gruppe oder eine CH2O-Gruppe verbrücktes Phenyl, einfach oder mehrfach substituiert, steht.
8. Substituierte 3-Phenylpiperidin-Derivate gemäß einem der Ansprüche 1 bis 7, dadurch gekennzeichnet, dass R2 für Methyl oder H steht.
9. Substituierte 3-Phenylpiperidin-Derivate gemäß einem der Ansprüche 1 bis 8 aus der Gruppe
(2-Fluorphenyl)-[3-hydroxy-3-(3-methoxyphenyl)-piperidin-1-yl]-methanon (3-Fluorphenyl)-[3-hydroxy-3-(3-methoxyphenyl)-piperidin-1-yl]-methanon (4-Fluorphenyl)-[3-hydroxy-3-(3-methoxyphenyl)-piperidin-1-yl]-methanon (2,3-Difluorphenyl)-[3-hydroxy-3-(3-methoxyphenyl)-piperidin-1-yl]-methanon

10. Verfahren zur Herstellung von substituierten 3-Phenylpiperidin-Derivaten gemäß Anspruch 2, dadurch gekennzeichnet, dass ein 3-Phenyl-piperidin-3-ol-Derivat der allgemeinen Formel Il
I! mit einem Säurechlorid der allgemeinen Formel R3C(O)CI umgesetzt wird.
11.Verfahren zur Herstellung von substituierten 3-Phenylpiperidin-Derivaten gemäß Anspruch 3, dadurch gekennzeichnet, dass die Ketogruppe von 3-Phenyl- piperidin-3-ol-Derivate der allgemeinen Formel Ia
Ia mit geeigneten Reduktionsmitteln, beispielsweise Lithiumaluminiumhydrid, reduziert wird.
12 Arzneimittel enthaltend wenigstens ein substituiertes 3-Phenylpiperidin -Derivat 5 gemäß Anspruch 1 , gegebenenfalls in Form seines Razemats, der reinen
Stereoisomeren, insbesondere Enantiomeren und Diastereomeren, in einem beliebigen Mischungsverhältnis; in Form seiner Säuren oder seiner Basen oder in Form seiner Salze, insbesondere der physiologisch verträglichen Salze oder Salze physiologisch verträglicher Säuren oder Kationen; oder in Form seiner IO Solvate, insbesondere der Hydrate, sowie gegebenenfalls enthaltend geeignete
Zusatz- und/oder Hilfsstoffe und/oder gegebenenfalls weiterer Wirkstoffe.
13. Verwendung eines substituierten 3-Phenylpiperidin-Derivates gemäß Anspruch 1 , gegebenenfalls in Form seines Razemats, der reinen Stereoisomeren,
L5 insbesondere Enantiomeren und Diastereomeren, in einem beliebigen
Mischungsverhältnis; in Form seiner Säuren oder seiner Basen oder in Form seiner Salze, insbesondere der physiologisch verträglichen Salze oder Salze physiologisch verträglicher Säuren oder Kationen; oder in Form seiner Solvate, insbesondere der Hydrate; zur Herstellung eines Arzneimittels zur Behandlung
.0 von Schmerz, insbesondere von akutem, neuropathischem oder chronischem
Schmerz.
14. Verwendung eines substituierten 3-Phenylpiperidin-Derivates gemäß Anspruch 1 , gegebenenfalls in Form seines Razemats, der reinen Stereoisomeren,
.5 insbesondere Enantiomeren und Diastereomeren, in einem beliebigen
Mischungsverhältnis; in Form seiner Säuren oder seiner Basen oder in Form seiner Salze, insbesondere der physiologisch verträglichen Salze oder Salze physiologisch verträglicher Säuren oder Kationen; oder in Form seiner Solvate, insbesondere der Hydrate; in Kombination mit einem weiteren Analgetikum,
50 beispielsweise einem Opioid, zur Herstellung eines Arzneimittels zur Behandlung von Schmerz, insbesondere von akutem, neuropathischem oder chronischem Schmerz.
15. Verwendung eines substituierten 3-Phenylpiperidin-Derivates gemäß Anspruch 1 , gegebenenfalls in Form seines Razemats, der reinen Stereoisomeren, insbesondere Enantiomeren und Diastereomeren, in einem beliebigen Mischungsverhältnis; in Form seiner Säuren oder seiner Basen oder in Form seiner Salze, insbesondere der physiologisch verträglichen Salze oder Salze physiologisch verträglicher Säuren oder Kationen; oder in Form seiner Solvate, insbesondere der Hydrate; zur Herstellung eines Arzneimittels zur Verwendung als Antiarrhythmikum, zur Behandlung/Therapie von cardiovaskulären Erkrankungen und/oder Bluthochdruck.
16. Verwendung eines substituierten 3-Phenylpiperidin-Derivates gemäß Anspruch 1 , gegebenenfalls in Form seines Razemats, der reinen Stereoisomeren, insbesondere Enantiomeren und Diastereomeren, in einem beliebigen Mischungsverhältnis; in Form seiner Säuren oder seiner Basen oder in Form seiner Salze, insbesondere der physiologisch verträglichen Salze oder Salze physiologisch verträglicher Säuren oder Kationen; oder in Form seiner Solvate, insbesondere der Hydrate; zur Herstellung eines Arzneimittels zur Verwendung als Antiemetikum, Nootropikum (Neurotropikum), zur Behandlung/Therapie von Harninkontinenz, Diarrhöe, Pruritus, Alkohol- und/oder Drogen- und/oder Medikamentenabhängigkeit, Entzündungen, zur Behandlung von Depressionen, zur Vigilanzsteigerung und Libidosteigerung.
