Suspensionen enthaltend A) mindestens eine oxidationsempfindliche Substanz, ausgewählt aus der Gruppe, bestehend aus Carotinoiden, Retinoiden und ungesättigten Fettsäuren und B) feste Teilchen eines oder mehrerer Salze der Ascorbinsäure in einem Dispersionsmittel, in dem die Salze der Ascorbinsäure unlöslich sind.

Suspensionen von Salzen der Ascorbinsäure und ihre Verwendung- als Antioxidantien
Beschreibung
Die vorliegende Erfindung betrifft Suspensionen von Salzen der Ascorbinsäure, deren Herstellung und Verwendung als Anti- oxidantien in der Kosmetik, Pharmazie sowie in der Human- und Tierernährung.
Die Oxidation von Fetten und Ölen sowie von lipophilen Wirkstoffen wie z.B. Carotinoiden und Retinoiden stellt bei der- Herstellung und Lagerung von kosmetischen und pharmazeutisctien Zubereitungen sowie von Lebensmitteln und Tierfuttermitteln, häufig ein großes Problem dar. Als Folge einer Oxidation von Fetten und ölen tritt u.a. die Ranzigkeit von Lebens- und Tierfuttermitteln sowie von kosmetischen und pharmazeutischen Zubereitungen auf, was in der Regel zur Unbrauchbarkeit dieser Produkte führt.
Bei Wirkstoffen wie Carotinoiden, die als Farbpigmente im Food- und Feed-Bereich eine breite Anwendung finden, kann die Oxiclation zu unerwünschten Abbaureaktionen und zu einer Verminderung 3ozw. einem Verlust der Farbintensität der Carotinoide führen.
Die Stoffklasse der Retinoide zählt mit zu den aktivsten Wirkstoffen, die in der Kosmetik und in der Dermatologie eingesetzt werden. Sie wirken u.a. regulierend auf das normale Zellwachstum und beeinflussen die Differenzierung von Epithelzellen. So werden Retinsäuren zur Behandlung von Akne und Retinol beispielsweise in Antifaltencremes eingesetzt.
Die Verwendung der Retinoide ist' jedoch ebenfalls stark ein— geschränkt, was unter anderem auf die hohe Instabilität der" Verbindungen zurückzuführen ist. Aus diesem Grund müssen bei 61er Herstellung Retinoid-haltiger Zubereitungen strenge Vorsichtsmaßnahmen eingehalten werden. Beispielsweise muss die Herstellung vollständig unter Schutzgas erfolgen und das fertige Produkt Sauerstoff-undurchlässig verpackt werden.
Antioxidativ hingegen wirken der Ausschluss von Licht und Sauerstoff, der Zusatz von Schwermetall-komplexierenden Stoffen wie z.B. Citrate, Gallate oder Tartrate, sowie der Zusatz von natürlichen (Vitamin E, Vitamin C, Carnosin, Gallate) oder synthetischen Antioxidantien (Butylhydroxyanisol , Butylhydroxy- toluol ) .
Die Verwendung der oben genannten synthetischen Antioxidantien ist aus toxikologischer und - aufgrund ihrer mangelnden biologischen Abbaubarkeit - aus ökologischer Sicht nicht immer unbedenklich .
Es sind eine Reihe von Verfahren zur Stabilisierung von Retinoiden beschrieben worden . So offenbart EP-A-1 055 720 die Stabilisierung von sauerstoffempfindlichen Verbindungen durch Verwendung von Thioverbindungen oder Glycoproteinen unter Ausschluss von Sauerstoff .
Gemäß WO 93 / 00085 und EP-A-0 440 398 werden sowohl wasser- als auch fettlösliche Antioxidantien zusammen mit Chelat-bildenden Mitteln zur Stabilisierung von Retinoiden eingesetzt .
EP-A-0 590 029 beschreibt Suspensionen von Ascorbinsäure-Partikel im Mikrometerbereich und ihre Verwendung als Antioxidantien u . a . für Fette , Öle , Carotinoide und Lebensmittel .
WO 01/67896 beschreibt ein Verfahren zur Herstellung öliger Suspensionen von festen Teilchen wasserlöslicher Vitamine durch Mahlung dieser Teilchen bis auf eine mittlere Partikelgröße von 0 , 1 bis 100 μ .
Die beschriebenen Verfahren führen j edoch nicht immer zu einer ausreichenden Stabilisierung im Sinne der Erfindung . Vielmehr zeigen sowohl wasserlösliche Antioxidantien, insbesondere Ascorbinsäure , als auch fettlösliche Antioxidantien , insbesondere Tocopherol , im Zusammenhang mit Retinoiden in bestimmten Konzentrationsbereichen eine destabilisierende' Wirkung .
Zudem kann es unter Umständen bei der Verwendung bestimmter Anti- oxidantienkombinationen zusammen mit Retinoiden zu unerwünschten Nebeneffekten, z.B. gelbliche Verfärbungen der Zubereitungen kommen, die die Verwendung dieser Systeme in der Kosmetik oder im Nahrungsmittelbereich unbrauchbar werden lassen.
Es war daher Aufgabe der vorliegenden Erfindung, eine Anti- oxidans-Zusammensetzung bereitzustellen, die die o.g. Nachteile des Standes der Technik nicht aufweist. Diese Aufgabe wurde gelöst d ch Suspensionen enthaltend
A) mindestens eine oxidationsempfindliche Substanz , ausgewählt aus der Gruppe , bestehend aus Carotinoiden, Retinoiden und ungesättigten Fettsäuren und
B) feste Teilchen eines oder mehrerer Salze der Ascorbinsäure
in einem Dispersionsmittel , in dem die Salze der Ascorbinsäure unlöslich sind.
Unter Carotinoide sind erfindungsgemäß folgende Verbindungen zu verstehen : ß-Carotin, Lycopin, Lutein, Astaxanthin, Zea- xanthin, Cryptoxanthin, Citranaxanthin, Canthaxanthin , Bixin , ß-Apo-4-carotinal , ß-Apo-8-carotinal , ß-Apo-8-carotinsäureester, einzeln oder als Mischung . Bevorzugt verwendete Carotinoide . sind ß-Carotin, Lycopin, Lutein, Astaxanthin , Zeaxanthin, Citranaxanthin und Canthaxanthin.
Die Carotinoide können dabei in kristalliner Form oder als Formulierung - beispielsweise als Trockenpulver , gemäß EP-A-0 065 193 eingesetzt werden.
Im Falle von Lycopin, Astaxanthin und Canthaxanthin werden bevor- zugt Lycopin- , Astaxanthin- bzw. Canthaxanthin-haltige Trockenpulver , beispielsweise LycoVit® , Lucantin® Pink bzw. Lucantin® Rot ( 10%ige Lycopin- , Astaxanthin- bzw . Canthaxanthin-Trocken- pulver, Fa . BASF AG, Ludwigshafen, Deutschland) zusammen mit den Salzen der Ascorbinsäure eingesetzt .
Unter Retinoide sind im Rahmen der vorliegenden Erfindung Vitamin A Alkohol - (Retinol) und seine Derivate wie Vitamin A Aldehyd (Retinal) , Vitamin A Säure (Retinsäure ) und Vitamin A Ester ( z . B. Retinylacetat , Retinylpropionat und Retinylpalmitat ) gemeint . Der Begriff Retinsäure umfasst dabei sowohl all-trans Retinsäure als auch 13-cis Retinsäure . Die Begriffe Retinol und Retinal umfassen bevorzugt die all-trans Verbindungen. Als bevorzugtes Retinoid verwendet man für die erfindungsgemäßen Suspensionen all-trans- Retinol , im folgenden als Retinol bezeichnet .
Ungesättigte Fettsäuren sind z . B . Undecylensäure , Palmitoleinsäure , Ölsäure , Linolsäure , Linolensä r e , Arachidonsäure , Eicosa- pentaensäure , Docosahexaensäure . Der Begriff Ascorbinsäure umfasst sowohl die L-Ascorbinsäure als auch deren diastereomere Form, die D-Ascorbinsäure (Isoascorbin- säure) . Als bevorzugte Ausführungsfor der erfindungsgemäßen festen Teilchen verwendet man die Salze der L-Ascorbinsäure.
Beispiele für Salze der L-Ascorbinsäure sind Alkali- oder Erdalkalimetallsalze der L-Ascorbinsäure wie Natrium-L-asσorbat , Kalium-L-ascorbat oder Calcium-L-ascor at , aber auch Salze der L-Ascorbinsäure mit organischen Aminverbindungen wie Cholin- ascorbat oder L-Carnitinascorbat . Bevorzugt verwendet man Alkalimetallsalze der L-Ascorbinsäure, besonders bevorzugt Natrium— L-ascorbat. Der Begriff Ascorbinsäure bzw. Ascorbat steht im folgenden für die L-Form.
Die oben genannten Salze der Ascorbinsäure können sowohl in kristalliner Form mit einer Reinheit größer 95 %, bevorzugt größer 98 %, besonders bevorzugt größer 99 %, als auch in formulierter Form, beispielsweise als Granulat, Beadlet oder als sprühgetrocknetes Pulver eingesetzt werden. Bevorzugt sind die o.g. Salze in ihrer kristallinen Form.
Als Dispersionsmittel, in dem die Salze der Ascorbinsäure unlöslich sind, kommen generell solche Substanzen in Frage, in denen die Löslichkeit der Ascorbat-Salze kleiner 0,01 g/g Dispersions- mittel ist. Dazu gehören beispielsweise Fette, Öle, Wachse sowie organische Lösungsmittel wie Ether, Petrolether, aromatische Kohlenwasserstoffe wie Toluol oder Xylol, chlorierte Kohlenwasserstoffe wie Dichlormethan oder Chloroform.
Die im Rahmen der vorliegenden Erfindung bevorzugt verwendeten Dispersionsmittel, in denen die Salze der Ascorbinsäure unlöslich sind, sind die für die Anwendung in der Kosmetik sowie in der Human- oder Tierernährung geeigneten Öle gemeint.
Als kosmetische Ölkörper kommen beispielsweise Guerbetalkohole auf Basis von Fettalkoholen mit 6 bis 18, vorzugsweise 8 bis 10 Kohlenstoffatomen, Ester von linearen C6~C2-Fettsäuren mit linearen C6-C22-Fettalkoholen, Ester von verzweigten C6-Ci3-Carbonsäuren mit linearen C6-C22-Fettalko ol n, Ester von linearen C6-C22-Fettsäuren mit verzweigten Alkoholen, insbesondere 2-Ethyl- hexanol, Ester von Hydroxycarbonsäuren mit linearen oder verzweigten C6-C22-Fettalkoholen, insbesondere Dioctyl Malate, Ester von linearen und/oder verzweigten Fettsäuren mit mehrwertigen Alkoholen (wie z.B. Propylenglycol, Di erdiol oder Trimertriol) und/oder Guerbetalkoholen, Triglycericle auf Basis Cδ-Cio-Fett- säuren, flüssige Mono-/Di-/Triglycericlmischungen auf Basis von C6-Ci8-Fettsäuren, Ester von C6-C22-Fettalkoholen und/oder Guerbet- alkoholen mit aromatischen Carbonsäuren, insbesondere Benzoe- säure , pflanzliche Öle, verzweigte primäre Alkohole, substituierte Cyclohexane, lineare und verzweigte C6-C22-Fettalkoh.ol- carbonate, Guerbetcarbonate, Ester der Benzoesäure mit linearen -und/oder verzweigten C6-C22-Alkoholen ( z . B . Finsolv® TN) , lineare oder verzweigte , symmetrische oder unsymmetrische Dialkylether mit 6 bis 22 Kohlenstoffatomen pro Alkylgruppe, Ringöffnungs- produkte von epoxidierten Fettsäureestern mit Polyolen, Siliconöle und/oder aliphatische bzw. naphthenische Kohlenwasserstoffe in Betracht .
Geeignete Siliconverbindungen sind beispielsweise Dimethylpoly- siloxane, Methylphenylpol siloxane, cyclische Silicone sowie - a ino-, fett säure- , alkohol-, polyether-, epoxy-, fluor-, glykosid- und/oder alkylmodifizierte Siliconverbindungen, die bei Raumtemperatur sowohl flüssig als auch harzförmig vorliegen können. Typische Beispiele für Fette sind Glyceride, als Wachse kommen u. a . Bienenwachs , Carnaubawachs , Candelillawachs , Montanwachs , Paraffinwachs oder Mikrowachse gegebenenfalls in Kombi - nation mit hydrophilen Wachsen, z .B. Cetylstearylalkohol oder
Partialglyceriden in Frage . Als Stabilisatoren können Metallsalze von Fettsäuren, wie z .B. Magnesium-, Aluminium- und/oder Zink- stearat eingesetzt werden .
Als essbare Öle kommen in der Regel alle physiologisch unbedenklichen Öle - sowohl pflanzlichen als auch tierischen Ursprungs - in Frage, insbesondere solche Öle, die bei 20°C flüssig sind bzw. die in der Suspension bei 20°C allein oder zusammen mit anderen Ölen die flüssige Phase ilden. Bevorzugt zu nennen sind in diesem Zusammenhang Sonnenblumenöl, Palmöl , Sesa öl, Maiskei- möl , Baumwollsaatöl , Soj abohnenöl oder Erdnussöl , Ester ittel- kettiger Triglyceride sowie außerdem Fischöle wie beispielsweise Makrelen- , Sprotten- oder Lachsöl . Für die Tierernährung besonders bevorzugt sind Fischöle, Maiskeimöl , Sonnenblumenöl und Erdnussöl . Für den Food- / Pharma-/Kosmetikbereich zusätzlich von Vorteil sind die Ester mittelkettiger Triglyceride (z .B . Delios® SK der Fa . Grünau; n-Octansäure/n-Decansäure Triglycerid) .
Die Menge der oben genannten oxidations empfindlichen Substanzen in den erfindungsgemäßen Suspensionen liegt im Bereich von 0, 1 bis 40 Gew. -%, bevorzugt von 0 , 5 bis 20 Gew. -% , besonders bevorzugt von 1 bis 15 Gew; -% , ganz besonders bevorzugt im Bereich von 3 bis 12 Gew. -% , wobei sich die Gew. -% Angaben auf die Gesamtmenge der Suspensionen beziehen. Der Gehalt an einem oder mehreren der oben genannten Salze der Ascorbinsäure liegt im Bereich von 1 bis 50 Gew.-%, bevorzugt von 5 bis 40 Gew.-%, besonders bevorzugt von 10 bis 35 Gew.—%, ganz besonders bevorzugt im Bereich von 15 bis 25 Gew.-%, wobei sich 5 die Gew.-% Angaben auf die Gesamtmenge der Suspensionen beziehen.
Bevorzugte Suspensionen im Sinne der vorliegenden Erfinctung enthalten
3.0 A) mindestens ein Retinoid, besonders bevorzugt Retinol und
B) feste Teilchen eines oder mehrerer Alkali- und/oder Erdalkalimetallsalze der Ascorbinsäure, besonders bevorzugt Natriumascorbat .

Bevorzugt sind ebenfalls Suspensionen, enthaltend als Komponente B) feste Teilchen eines oder mehrerer Salze der Ascorbinsäure, deren mittlere Partikelgröße D[4, 3] im Bereich von 0, 01 bis 1000 um, bevorzugt von 0 , 1 bis 500 um, besonders bevorzugt von
20 0 , 2 bis 200 Um, ganz besonders bevorzugt im Bereich von 0 , 5 bis 50 Um, insbesondere 0, 5 bis 20 um liegt . Der Begriff D [ 4 , 3] bezeichnet den volumengewichteten mittleren Durchmesser (siehe Handbuch zu Malvern Mastersizer S, Malvern Instruments Ltd. , UK) .
25 Die erfindungsgemäßen Suspensionen können zusätzlich Vitamin E, Vitamin E-Derivate oder Mischungen davon enthalten. Die Bezeichnung Vitamin E steht in diesem Zusammenhang für natürliches oder synthetisches α- , ß- , γ- oder δ-Tocopherol sowie für Tocotrienol . Vitamin E-Derivate sind z . B . Tocopheryl-Cι-C20-Alkansäur-eester wie
30 Tocopherylacetat oder Tocopherylpalmitat . Bevorzugt verwendet man natürliches oder synthetisches α-Tocopherol . Vitamin E wird in einer Menge von 1 bis 50 Gew. -% , bevorzugt von 2 bis 40 Gew. -%, besonders bevorzugt von 5 bis 30 Gew.-% , ganz besonders bevorzugt von 15 bis 25 Gew. -% eingesetzt (Gew.-% Angabe bezogen auf die
35 Gesamtmenge der Suspensionen) .
Es ist auch möglich, Vitamin E, Vitamin E-Derivate oder Mischungen davon als Dispersionsmittel für die erfindungsgemäßen Suspensionen zu verwenden . 40
In diesem Fall können die Suspensionen
A) 0 , 1 bis 40 Gew. -% einer oder mehrerer oxidationsempf ind- licher Substanzen, ausgewählt aus der Gruppe , bestehend 45 aus Carotinoiden, Retinoiden und ungesättigten Fettsäuren, B) 1 bis 50 Gew.-% eines oder mehrerer Salze der Ascorbinsäure und
C) 10 bis 98,9 Gew.-% Vitamin E, Vitamin E-Derivate oder Mischungen davon enthalten,
wobei sich die Gew.-% Angaben auf die Gesamtmenge der Suspensionen beziehen und die Summe der Gewichtsanteile A bis C 100 % ergibt.
In manchen Fällen kann es von Vorteil sein, wenn die erf indungs- cjemäßen Suspensionen zusätzlich mindestens ein Trockenmittel, mindestens einen Verdicker und/oder mindestens ein oberf lächen- a.ktives Mittel enthalten.
Als Verdicker kommen beispielsweise Polysaccharide, insbesondere Xanthan-Gum, Guar-Guar, Agar-Agar, Alginate und Tylose , Carboxy- methylcellulose und Hydroxyethylcellulose, ferner höhermolekulare Polyethylenglycolmono- und -diester von Fettsäuren, Polvacrylate (z.B. Carbopole® von Goodrich oder Synthalene® von Sigma), Poly- acrylamide , Polyvinylalkohol (PVA) und Polyvinylpyrroliόlon (PVP) sowie VA/VP-Copolymere (Luviskol®, Fa. BASF), Polyethylene, Tenside wie beispielsweise ethoxylierte Fettsäur eglyceride (u.a. Cremophor® C0, Fa. BASF), Ester von Fettsäuren mit Polyolen wie beispielsweise Pentaerythrit oder Trimethylolpropan, Fettalkoho- lethoxylate mit eingeengter Homologenverteilung oder Alkiyloligog- lucoside, Elektrolyte wie Kochsalz und Ammoniumchlorid sowie Silikate wie Magnesium-Aluminiumsilikate (Veegum® Ultra, Fa. R.T. Vanderbilt Comp.,Inc), Schichtsilikate wie Bentone® EW (Fa. Ele- xnentis Specialities) oder Tixogel® VP (Fa. Süd Chemie) . Weitere Verdicker sind hochdisperse Kieselsäure (Aerosil®, Fa. Degussa) , Polyethylenglykole (Lutrol®, Fa. BASF) , Stearinsäure, Trihydroxy- stearin, Polyoxyethylenglyceroltriricinoleat, Triglycer-ide von C12 /Cχ4-Fettsäuren .
Die Menge an Verdicker in den erfindungsgemäßen Suspensionen liegt im Bereich von 0,1 bis 10 Gew.-%, bevorzugt 0,5 bis 8 Gew.-%, besonders bevorzugt 0,7 bis 3 Gew.-%, ganz besonders bevorzugt 0,8 bis 1,5 Gew.-%, bezogen auf die Gesamtmenge der Suspensi onen .
Als oberflächenaktives Mittel kommen beispielsweise folgende Substanzen in Frage: ) Anlagerungsprodukte von 2 bis 30 Mol Ethylenoxid und/ oder 0 bis 5 Mol Propylenoxid an lineare Fettalkohole mit 8 bis 22 C-Atomen, an Fettsäuren mit 12 bis 22 C-Atomen und an Alkylphenole mit 8 bis 15 C-Atomen in der Alkylgruppe ;
) Cl2/18-Fettsäuremono- und -diester von Anlagerungsprodukten von 1 bis 30 Mol Ethylenoxid an Glyc rin;
) Glycerinmono- und -diester und Sorbi anmono- und -diester von gesättigten und ungesättigten Fettsäuren mit 6 bis 22 Kohlenstoff atomen und deren Ethylenoxiάanlagerungsprodukte ;
) Alkylmono- und -oligoglycoside mit 8 bis 22 Kohlenstoff atomen im Alkylrest und deren ethoxylierte Analoga;
) Anlagerungsprodukte von 15 bis 60 Mol Ethylenoxid an Ricinus- öl und/oder gehärtetes Ricinusöl;
) Polyol- und insbesondere Polyglycerinester, wie z . B . Poly- glycerinpolyricinoleat , Polyglycerinpoly-12-hydroxystearat oder Polyglycerindimerat . Ebenfalls geeignet sind Gemische von Verbindungen aus mehreren dieser Substanzklassen;
) Anlagerungsprodukte von 2 bis 15 Mol Ethylenoxid an Ricinusöl und/oder gehärtetes Ricinusöl ;
) Partialester auf Basis linearer, verzweigter, ungesättigter bzw. gesättigter C6/22-Fettsäuren, Ricinolsäure sowie 12-Hydroxystearinsäure und Glycerin , Polyglycerin, Penta- erythrit , Dipentaerythrit , Zuckeralkohole ( z . B . Sorbit ) ,
Alkylglucoside ( z . B . Methylglucosid , Butylglucosid, Lauryl- glucosid) sowie Polyglucoside (z .B. Cellulose) ;
) Mono- , Di- und Trialkylphosphate sowie Mono- , Di- und/ oder Tri-PEG-alkylphosphate und deren Sal ze;
0 ) Wollwachsalkohole ;
1 ) Polysiloxan-Polyalkyl-Polyether-Copolymere bzw. entsprechende Derivate ;
2 ) Mischester aus Pentaerythrit, Fettsäuren, Citronensäure und Fettalkohol gemäß DE-PS 1165574 uncl/ oder Mischester von ' Fettsäuren mit 6 bis 22 Kohlenstoff atomen , Methylglycose und Polyolen, vorzugsweise Glycerin oder Polyglycerin sowie
3 ) Polyalkylenglycole . Die Anlagerungsprodukte von Ethylenoxid und/ oder von Propylenoxid an Fettalkohole , Fettsäuren, Alkylphenole , Glycerinmono- und -diester sowie Sorbitanmono- und -diester von Fettsäuren oder an Ricinusöl stellen bekannte , im Handel erhältliche Produkte dar. Es handelt sich dabei um Homologengemische, deren mittlerer
Alkoxylierungsgrad dem Verhältnis der St off mengen von Ethylenoxid und/ oder Propylenoxid und Substrat, mit denen die Anlagerungsreaktion durchgeführt wird, entspricht . Ci2/Ci8-Fe tsäuremono- und -diester von Anlageruncjsprodukten von Ethylenoxid an Glycerin sind aus DE-PS 2024051 als Rückf ttungsmittel für kosmetische
Zubereitungen bekannt . C8/Ci8-Alkylmono- und -oligoglycoside , ihre Herstellung und ihre Verwendung sind aus dem Stand der Technik bekannt . Ihre Herstellung erfolgt insbesondere durch Umsetzung von Glucose oder Oligosacchariden mit primären Alkoholen mit 8 bis 18 C-Atomen. Bezüglich des Glycosidesters gilt , dass sowohl Monoglycoside, bei denen ein cyclischer Zuckerrest glycosidisch an den Fettalkohol gebunden ist, als auch oligomere Glycoside mit einem Oligomerisationsgjrad bis vorzugsweise etwa 8 geeignet sind. Der Oligomerisierungsgrad ist dabei ein statistischer Mittel- wert , dem eine für solche technischen Produkte übliche Homologenverteilung zugrunde liegt .
Weiterhin können als Emulgatoren zwitterionische Tenside verwendet werden. Als zwitt er ionische Tenside werden solche oberflä- chenaktiven Verbindungen bezeichnet, die im Molekül mindestens eine quartäre Ammoniumcjruppe und mindestens eine Carboxylat- und eine Sulfonatgruppe tragen. Besonders geeignete zwitterionische Tenside sind die sogenannten Betaine wie die N- Alkyl -N,N-di- methylammoniumglycinate , beispielsweise das Kokosalkyldimethyl- ammoniumglycinat , N-Acγ-laminopropyl-N,N-dimethylammoniumglyci- nate , beispielsweise das Kokosacylaminopropyldimet.hylammonium- glycinat , und 2 -Alkyl-3 -carboxymethyl-3 -hydroxyethylimidazoline mit jeweils 8 bis 18 C— Atomen in der Alkyl- oder Acylgruppe sowie das Kokosacyla inoethylhydroxyethylcarboxymethylglycinat .
Besonders bevorzugt ist das unter der CTFA-Bezeichnung Cocamido- propyl Betaine bekannte Fettsäureamid-Derivat . Ebenfalls geeignete Emulgatoren sind ampholy tische Tenside. Unter ampholy tischen Tensiden werden solche oberflächenaktiven Verbindungen ver- standen, die außer einer Cs/Ci8-Alkγl- oder -Acylgruppe im Molekül mindestens eine freie Aminogruppe und mindestens eine -COOH- oder -S03H-Gruppe enthalten und zur Ausbildung innerer Salze befähigt sind. Beispiele für geeignete ampholytische Tenside sind N-Älkyl- glycine , N-Alkylaminopropionsäuren, N-Alkylaminobuttersäuren, N-Alkyliminodipropionsäuren, N-Hydroxyethyl-N-alkylamidopropyl- glycine , N-Alkyltaurine , N-Alkylsarcosine , 2 -Alkyl aminoprop ionsäuren und Alkylaminoessigsäuren mit j eweils etwa 8 bis 18 C-Ato- men in der Alkylgruppe . Besonders bevorzugte ampholytische Tenside sind das N-Kokosalkylaminopropiona , das Kokosacylaminoethy- laminopropionat und das Ci2/Ci8-Acylsarcosin. Neben den ampholyti- schen kommen auch quartäre Emulgatoren in Betracht , wobei solche vom Typ der Esterquats , vorzugsweise methylquatemierte Difett- säuretriethanolaminester-Salze , besonders bevorzugt sind.
Die Menge an oberflächenaktiven Mitteln in den erfindungsgemäßen Suspensionen liegt im Bereich von 0 , 1 bis 10 Gew. -% , bevorzugt 0 , 5 bis 8 Gew. -% , besonders bevorzugt 1 bis 5 Gew. -% , bezogen auf die Gesamtmenge der Suspensionen .
Als Trockenmittel kommen beispielsweise Verbindungen, ausgewählt aus der Gruppe , bestehend aus Alkali- und Erdalkali sulfaten wie Natrium- , Calcium- und Magnesiumsulfat , Alkali- und Erdalkalichloriden wie Natrium- , Calcium- und Magnesiumchlorid und Kieselgel in Frage . Als ganz besonders bevorzugtes Trockenmittel ist CaCl2 zu nennen.
Die Menge an eingesetztem Trockenmittel liegt im allgemeinen zwischen 0 , 1 und 20 Gew. -% , bevorzugt zwischen 0 , 5 und 15 Gew. -% , besonders bevorzugt zwischen 1 , 0 und 10 Gew. -%, bezogen auf die Gesamtmenge der Suspension.
Gegenstand der Erfindung ist auch ein Verfahren zur Herstellung der eingangs beschriebenen Suspensionen , dadurch gekennzeichnet , dass man
a) feste Teilchen eines oder mehrerer Salze der Ascorbinsäure in einem Dispersionsmittel, in dem die Salze der Ascorbinsäure unlöslich sind, bis auf eine mittlere Partikelgröße von 0 , 01 bis 1000 Um mahlt , wobei man die oscidationsempf indliche (n) Substanz (en) , ausgewählt aus der Gruppe, bestehend aus Carotinoiden, Retinoiden und ungesättigten Fettsäuren vor, während oder nach der Mahlung zum Dispersionsmittel geben kann oder
b) dass man feste Teilchen eines oder mehrerer Salze der Ascorbinsäure ohne Verwendung einer kontinuierlichen Phase bis auf eine mittlere Partikelgröße von 0 , 01 bis 1000 um mahlt und die gemahlenen Partikel anschließend in einem Dispersionsmittel, in dem die Salze der Ascorbinsäure unlöslich sind suspendiert , wobei man clie oxidationsempfindli- che (n) Substanz (en) , ausgewählt aus der Gruppe , bestehend aus Carotinoiden, Retinoiden und ungesättigten Fettsäuren vor, während oder nach der Suspendierung der festen Ascorbat-Teil- chen zum Dispersionsmittel geben kann-
Die Mahlung gemäß a) kann in an sich bekannter Weise z . B . mit einer Kugelmühle erfolgen. Dabei wird je nach verwendetem Mühlentyp so lange gemahlen, bis die Teilchen die oben genannte, über Fraunhofer Beugung ermittelte mittlere Partikelgröße D [4 , 3 ] von 0 , 01 bis 1000 um bevorzugt von 0, 1 bis 500 um, besonders bevorzugt von 0, 2 bis 200 um, ganz besonders bevorzugt im Bereich von 0, 5 bis 50 um, insbesondere 0 , 5 bis 20 Um aufweisen.
Nähere Einzelheiten zur Mahlung unόi den dafür eingesetzten Apparaturen finden sich u. a . in üllmann's Encyclopedia of Industrial Chemistry, Sixth Edition, 1999 , Electronic Release, Size Reduction, Kapitel 3 . 6 . : Wet Grinding.
Eine bevorzugte Aus führungs form der unter a) beschriebenen Nassmahlung erfolgt in Gegenwart eines oder mehrerer Verdickungs- mittel, wobei man die oxidationsempf indliche (n) Substanz (en) nach der Mahlung zum Dispersionsmittel gibt .
Neben der Nassmahlung, lassen sich die erfindungsgemäßen Suspensionen auch gemäß b) durch Trockenmahlung der feste Teilchen eines oder mehrerer Salze der Ascorbinsäure und anschließendes Suspendieren der gemahlenen Partikel in einem Dispersionsmittel , in dem die Salze der Ascorbinsäure unlöslich sind herstellen. Als Trockenmahlung versteht man in diesem Zusammenhang eine Mahlung ohne Verwendung einer kontinuierlichen Phase. Die oxidations empfindliche (n) Substanz (en) , ausgewählt aus der Gruppe, bestehend aus Carotinoiden, Retinoiden und ungesättigten Fettsäuren kann man vor, während oder nach der Suspendierung der festen Ascorbat— Teilchen zum Dispersionsmittel geben .
Eine bevorzugte Aus führungs form der unter b) beschriebenen Herstellung der Suspensionen erfolgt derart , dass die oxidationsempf indliche (n) Substanz (en) bereits vor der Zugabe der Ascorbat— Teilchen im Dispersionsmittel gelöst bzw. dispergiert vorliegen.

Gegenstand der Erfindung ist auch die Verwendung von festen Teilchen eines oder mehrerer Salze der Ascorbinsäure als Antioxidantien für oxidationsempfindliche Substanzen, ausgewählt aus der Gruppe, bestehend aus Carotinoiden, Retinoiden und ungesättigten Fettsäuren in einem Dispersionsmittel , in dem die Salze der Ascorbinsäure unlöslich sind.
Die erfindungsgemäße Verwendung erfolgt vorteilhafterweise mit f sten Teilchen eines oder mehrerer Salze der Ascorbinsäure mit einer mittleren Partikelgröße im Bereich von 0 , 01 bis 1000 Um, bevorzugt von 0 , 1 bis 500 um, besonders bevorzugt von 0 , 2 bis 2 00 um, ganz besonders bevorzugt im Bereich von 0 , 5 bis 50 μm, insbesondere 0 , 5 bis 20 um .
Ferner ist die Verwendung bevorzugt , welche α-Tocopherol als zusätzliches , öllösliches Antioxidans mitumfasst .
Die erfindungsgemäßen Suspensionen zeichnen sich u . a. dadurch aus , dass bei deren Herstellung, Abfüllung und Lagerung auf die Verwendung von Schutzgas verzichtet werden kann bei gleichzeitiger Gewährleistung einer ausreichenden Stabilität .
Unter ausreichender Stabilität im Sinne der Erfindung wird ver- standen, dass das Retinoid in der Zubereitung nach mindestens 2 Wochen Lagerung bei 40°C zu mindestens 90 % wiedergefunden wird . Ferner kommt es bei der Lagerung der erfindungsgemäßen Zubereitungen zu keinen unerwünschten Farbveränderungen.
Aus der folgenden Tabelle ist ersichtlich, dass die erfindungsgemäßen Suspensionen unter Verwendung von festen Teilchen -von Salzen der Ascorbinsäure, insbesondere unter Verwendung von festen Natriumascorbat-Partikeln vorteilhafte Eigenschaften aufweisen. . Während die Retinol Zubereitungen unter Verwendung von festen Ascorbinsäure-PartiJ eln, gemäß EP-B-590 029 , eine s ignifikante Abnahme an Retinol bereits nach zweiwöchiger Lagerung zeigen, liegt die Wieder findungsrate von Retinol in Gegenwart von f sten Natriumascorbat-Partikeln - mit und ohne Schutzgas — über 9 0 % .
Die Suspensionen eignen sich u. a . als Zusatzstoff für Lebensmittel- und Tierfuttermittelzubereitungen bzw Mischfutter, als Mittel für die Herstellung pharmazeutischer und kosmetischer Zubereitungen sowie für die Herstellung von Nahrungsergänzungs- präparaten im Human- und Tierbereich.
Bevorzugt lassen sich die Suspensionen für die Herstellung kosmetischer Zubereitungen einsetzen..
Ebenfalls bevorzugt lassen sich die Suspensionen als Futtermittelzusatz in der Tierernährung einsetzen, insbesondere zum Auftragen bzw. Aufsprühen auf Futtermittelpellets .
Die Anwendung als Futtermittelzusatzstoff erfolgt insbesondere durch direktes Aufsprühen der erfindungsgemäßen Suspensionen, gegebenenfalls nach Verdünnung mit Ölen, beispielsweise auf Tierfutterpellets als sogenannte "post-pelleting-application" .
Eine bevorzugte Aus führungs form des Sprühverfahrens besteht darin, dass man die Futtermittelpellets unter vermindertem Druck mit der öligen Suspension belädt . Beispiele hierfür finden sich u.a. in GB-A-2 232 573 sowie in EP-A-0 556 883.
Typische Einsatzgebiete im Lebensmittelbereich sind beispiels- weise die Vitaminierung von Getränken, Milchprodukten wie
Joghurt, Milchmixgetränken oder Milchspeiseeis sowie von Puddingpulvern, Eiprodukten, Backmischungen und Süßwaren.
Im Kosmetikbereich können die öligen Suspensionen beispielsweise für Vitamin-haltige Kδrperpflegemittel beispielsweise in Form einer Creme, einer Lotion, als Lippenstifte oder Make-up verwendet werden.
Gegenstand der Erfindung sind ferner Nahrungsergänzungsmittel, Tierfuttermittel, Lebensmittel sowie pharmazeutische und kosmetische Zubereitungen, enthaltend die eingangs beschriebenen öligen Suspensionen wasserlöslicher Vitamine.
Unter Nahrungsergänzungspräparate sowie pharmazeutische Zubereitungen, die die erfindungsgemäße Suspension enthalten, sind u.a. Tabletten, Dragees sowie bevorzugt Hart- und Weichgelatinekapseln zu verstehen.
Kosmetische Zubereitungen, die die erfindungsgemäßen Suspensionen enthalten können, sind beispielsweise topisch anwendbare Zubereitungen, insbesondere dekorative Körperpflegemittel wie Lippenstifte, Gesichts-Make-up in Form einer Creme sowie Lotionen.
In den folgenden Beispielen wird die Herstellung der erfindungsgemäßen Suspensionen näher erläutert .
Beispiel 1
Eine Mischung aus 200 g Natrium-L-ascorbat, 100 g all-rac-α-Toco- pherol und 98 g all-trans Retinol in 550 g eines mittelkettigen Triglycerids (Delios® SK der Fa. Grünau, Deutschland) wurde mit einem Blattrührer solange gerührt, bis eine homogene Suspension vorlag. Danach wurde die Mischung in eine rührbare Vorlage umge- füllt, aus der die Suspension mittels Schlauchpumpe durch eine kontinuierlich betriebene Kugelmühle (Dyno Mill KDL Spezial) gefördert wurde. Der Mahlbehälter der Kugelmühle war mit 400 g Glaskugeln (Durchmesser 800 bis 1200 μm) gefüllt. Die aus der Mühle austretende feinteilige Suspension wurde aufgefangen und mittels eines Partikelgrößenmessgerätes (Malvern Mastersizer) vermessen. Der Mahlvorgang wurde so oft wiederholt bis 90 % der suspendierten Teilchen eine Teilchengröße kleiner 10 μm [D(0.9) < 10 μ ] hatten. Dies entsprach einer mittleren Partikelgröße D [ 4 3 ] von 5 , 2 μm.
Beispiel 2
Eine Mischung aus 400 g Natrium-L-ascorbat, 100 g all-rac-α-Toco- pherol , 50 g all-trans Retinol und 15 g Tixogel® VP (Bentonit der Fa . Süd Chemie) in 300 g eines mittelkettigen Triglycerids (Delios® SK der Fa. Grünau, Deutschland) wurde mit einem Blatt- rührer solange gerührt, bis eine homogene Suspension vorlag.
Danach wurde die Mischung in eine rührbare Vorlage umgefüllt , aus der die Suspension mittels Schlauchpumpe durch eine kontinuierlich betriebene Kugelmühle (Dyno Mill KDL Spezial ) gefördert wurde . Der Mahlbehälter der Kugelmühle war mit 400 g Glaskugeln (Durchmesser 800 bis 1200 um) gefüllt . Die aus der Mühle austretende f einteilige Suspension wurde aufge angen und mittels eines Partikelgrößenmessgerätes (Malvern Mastersizer) vermessen. Der Mahl Vorgang wurde so oft wiederholt bis 90 % der suspendierten Teilchen eine Teilchengröße kleiner 20 Um [D(0 . 9 ) < 20 μm] hatten . Dies entsprach einer mittleren Partikelgröße D [ 4 , 3 ] von 10 , 9 .
Nach der Abtrennung von den Mahlkörpern wurde ein Teil der Dispersion mit der 10 fachen Menge des verwendeten Öls verdünnt und über 12 h stehengelassen. Weder die unverdünnte noch die verdünnte Dispersion zeigten über diesen Zeitraum Sedimentationserscheinungen .
Beispiel 3
Eine Mischung aus 100 g Natrium-L-ascorbat , 100 g all-rac-α-Toco- pherol , 50 g all-trans Retinol und 15 g Tixogel® VP (Bentonit der Fa . Süd Chemie) in 300 g eines mittelkettigen Triglycerids (Delios® SK der Fa . Grünau, Deutschland) wurde mit einem Blattrührer solange gerührt , bis eine homogene Suspension vorlag . Danach wurde die Mischung in eine rührbare Vorlage umgefüllt, aus der die Suspension mittels Schlauchpumpe durch eine kontinuierlich betriebene Kugelmühle (Dyno Mill KDL Spezial ) gefördert wurde . Der Mahlbehälter der Kugelmühle war mit 400 g Glaskugeln (Durch- esser 800 bis 1200 μm) gefüllt . Die aus der Mühle austretende feinteilige Suspension wurde aufgefangen und mittels eines Partikelgrößenmessgerätes (Malvern Mastersi zer) vermessen. Der Mahlvorgang wurde so oft wiederholt bis 90 % der suspendierten Teilchen eine Teilchengröße kleiner 20 μm [D ( 0 . 9 ) < 20 μm] hatten. Dies entsprach einer mittleren Partikelgröße D [4 , 3 ] von 10 , 9 um. Beispiel 4
Eine Mischung aus 200 g Natrium-L-ascorbat , 200 g all-rac-CC-Toco- pherol und 10 g Cremophor® CO 40 (Fa . BASF) in 255 g eines it- telkettigen Triglycerids (Miglyol® der Fa . Hüls , Deutschland) wurde mit einem Blattrührer solange gerührt , bis eine homogene Suspension vorlag . Danach wurde die Mischung in eine rührbare Vorlage umgefüllt, aus der die Suspension mittels Schlauchpumpe durch eine kontinuierlich betriebene Kugelmühle (Dyno Mill KDL Spezial ) gefördert wurde . Der Mahlbehälter der Kugelmühle war mit 400 g Glaskugeln (Durchmesser 800 bis 1200 μm) gefüllt . Die aus der Mühle austretende feinteilige Suspension wurde aufgefangen und mittels eines Partikelgrößenmessgerätes (Malvern Mastersizer ) vermessen . Der Mahl vor gang wurde so oft wiederholt bis 90 % der suspendierten Teilchen eine Teilchengröße kleiner 20 μm [D ( 0.9 ) < 20 μ ] hatten . Dies entsprach einer mittleren Partikelgröße D[4 , 3 ] von 10 , 9 μ . Anschließend wurde die Suspension mit einer Lösung von 50 g all-trans Retinol in 285 g Miglyol versetzt und erneut mit einem Blattrührer solange gerührt , bis eine homogene Suspension vorlag .
Beispiel 5
200 g -Natriumascorbat , das durch Trockenmahlung auf eine Parti- kelgröße von < 10 μm zerkleinert wurde , wurde zu einer Mischung aus 20 g all-rac-α-Tocopherol , 50 g all-trans Retinol und 10 g Cremop hor® CO 40 (Fa . BASF) in 540 - g eines mittelkettigen Triglycerids (Miglyol® der Fa. Hüls , Deutschland) gegeben. Diese Mischung wurde anschließend mit einem Blattrührer solange gerührt , bis eine homogene Suspension vorlag.

Claims

Patentansprüche
1. Suspensionen enthaltend
5
A) mindestens eine oxidationsempf indliche Substanz , ausgewählt aus der Gruppe, bestehend aus Carotinoiden, Retinoiden und ungesättigten Fettsäuren und
10 B) feste Teilchen eines oder mehrerer Salze der Ascorbinsäure
in einem Dispersionsmittel , in dem die Salze der Ascorbinsäure unlöslich sind. 15
2. Suspensionen nach Anspruch 1, enthaltend
A) 0 , 1 bis 40 Gew. -% einer oder mehrerer oxidationsempf indlicher Substanzen, ausgewählt aus der Gruppe,
20 bestehend aus Carotinoiden, Retinoiden und ungesättigten
Fettsäuren und
B) 1 bis 50 Gew. -% eines oder mehrerer Salze der Ascorbinsäure ,

-wobei sich die Gew. -% Angaben auf die Gesamtmenge der Suspensionen beziehen.
3. Suspensionen nach einem der Ansprüche 1 oder 2 , enthaltend 30
A) mindestens ein Retinoid und
B) feste Teilchen eines oder mehrerer Alkali- und/ oder Erd-alkalimetallsalze der Ascorbinsäure .

4. Suspensionen nach einem der Ansprüche 1 bis 3 , enthaltend als Komponente B) feste Teilchen eines oder mehrerer Salze der Ascorbinsäure , deren mittlere Partikelgröße im Bereich von 0 , 01 bis 1000 μ liegt .

5. Suspensionen nach einem der Ansprüche 1 bis 4, enthaltend Retinol und mindestens ein Alkali- und/oder Erdalkalimetallsalz der Ascorbinsäure.
456. Suspensionen nach Anspruch 5, enthaltend Natriumascorbat und Retinol .
7 . Suspensionen nach einem der Ansprüche 1 bis 6 , enthaltend zus ä t z 1 i ch α-Toc ophero 1.
8 . Suspensionen nach einem der Ansprüche 1 bis 7 , dadurch
5 gekennzeichnet, dass das Dispersionsmittel, in dem die Salze der Ascorbinsäure unlöslich sind, ein für die Anwendung in der Kosmetik sowie in der Human- oder Tierernährung geeignetes Öl ist .
10 9 . Suspensionen nach einem der Ansprüche 1 bis 8 , enthaltend zusätzlich mindestens ein Trockenmittel , mindestens einen Verdicker und/oder mindestens ein oberflächenaktives Mittel .
10. Verfahren zur Herstellung von Suspensionen, definiert gemäß 15 Anspruch 1 , dadurch gekennzeichnet , dass man
a) feste Teilchen eines oder mehrerer Salze der Ascorbinsäure in einem. Dispersionsmittel, in dem die Salze der Ascorbinsäure unlöslich sind, bis auf eine mittlere
20 Partikelgröße von 0 , 01 bis 1000 μm mahlt , wobei man die oxidationsempf indliche (n) Substanz (en) , ausgewählt aus der Gruppe, bestehend aus Carotinoiden, Retinoiden und ungesättigten Fettsäuren vor, während oder nach der Mahlung- zum Dispersionsmittel geben kann oder
25 b) dass man feste Teilchen eines oder mehrerer Salze der Ascorbinsäure ohne Verwendung einer kontinuierlichen Phase bis auf eine mittlere Partikelgröße von 0 , 01 bis 1000 μ mahlt und die gemahlenen Partikel anschließend
30 in einem Dispersionsmittel, in dem die Salze der
Ascorbinsäure unlöslich sind suspendiert , wobei man die oxidationsempf indliche (n) Substanz (en) , ausgewählt aus der Gruppe , bestehend aus Carotinoiden, Retinoiden und ungesättigten Fettsäuren vor, während oder nach
35 der Suspendierung der festen Ascorbat -Teilchen zum
Dispers ionsmittel geben kann .
11. Verwendung von festen Teilchen eines oder mehrerer Sal ze der Ascorbinsäure als Antioxidantien für oxidations-
40 empfindliche Substanzen., ausgewählt aus der Gruppe, bestehend aus Carotinoiden, Retinoiden und ungesättigten Fettsäuren in einem Dispersionsmittel , in dem die Salze der Ascorbinsäure unlöslich sind.
45 12 . Verwendung nach Anspruch 11 , worin die mittlere Partikielgröße der festen Teilchen im Bereich von 0 , 01 bis 1000 μm liegt .
13. Verwendung nach einem der Ansprüche 11 oder 12 , welche α-Tocopherol als zusätzliches , öllösliches Antioxidans mitumfasst .
5 14. Verwendung von Suspensionen, definiert gemäß einem der Ansprüche 1 bis 9 als Zusatz zu Lebensmitteln und Tier— futtermitteln, Pharmazeutika und kosmetischen Zubereitungen.
15. Verwendung nach Anspruch 14 als Futtermittelzusatz in der 10 Tierernährung.
16. Verwendung nach Anspruch 15 zum Auftragen auf Futtermittelpellets .
15 17. Verwendung nach Anspruch 16, dadurch gekennzeichnet, dass man die Futtermittelpellets unter vermindertem Druck mit der öligen Suspension belädt.
