Abstract
Die vorliegende Erfindung betrifft Uracil-Thioether, Verfahren zur ihrer Herstellung, sie umfassen pharmazeutische Zusammensetzungen sowie ihre Verwendung bei der Behandlung von Erkrankungen bei Menschen oder Tieren.

Die vorliegende Erfindung betrifft Uracil-Thioether, Verfahren zur ihrer Herstellung, sie umfassen pharmazeutische Zusammensetzungen sowie ihre Verwendung bei der Behandlung von Erkrankungen bei Menschen oder Tieren.
Gram-positive Eubakterien enthalten drei unterschiedliche DNA-Polymerase-Exo- Nucleasen, die als Pol 1, Pol 2 und Pol 3 bezeichnet werden. Pol 3 ist ein Enzym, welches für die replikative Synthese der DNA notwendig ist.
Die Eignung von Uracilderivaten für die Behandlung bakterieller . Infektionen ist schon sein einiger Zeit bekannt. So beschreibt WO 01/29010 3-Aminocarbonyl-sub- stituierte Phenylaminouracile, WO 96/06614 beschreibt 3-Alkyliden-substituierte Uracile, WO 00/71523 beschreibt 3-Alkanoyloxyalkyluracile und WO 00/20556 beschreibt Uracile mit Zink-Finger-aktiver Einheit als antibakterielle Verbindungen.
In J Med. Chem., 1999, 42, 2035, Antimicro. Agents and Chemotherapy, 1999, 43, 1982 und Antimicro. Agents and Chemotherapy, 2000, 44, 2217 sind Phenylaminouracile als antibakterielle Verbindungen beschrieben.
Auf dem Markt sind zwar weitere, strukturell andersartige antibakteriell wirkende
Mittel vorhanden, es kann aber regelmäßig zu einer Resistenzentwicklung kommen. Neue Mittel für eine bessere und wirksame Therapie sind daher wünschenswert.
Eine Aufgabe der vorliegenden Erfindung ist es daher, neue Verbindungen mit gleicher oder verbesserter antibakterieller Wirkung zur Behandlung von bakteriellen
Erkrankungen bei Menschen und Tieren zur Verfügung zu stellen.
Überraschenderweise wurde gefunden, dass Derivate dieser Verbindungsklasse, worin das Uracil durch einen Thioether substituiert ist, antibakteriell hochwirksam sind.
Gegenstand der vorliegenden Erfindung sind daher Verbindungen der allgemeinen Formel (I)
in welcher
R1 für Hydroxy, Alkoxy, Alkenyl, Cycloalkyl, Aryl, Heterocyclyl oder Heteroaryl steht,
wobei R1 gleich Aryl gegebenenfalls substituiert sein kann mit 1 bis 3 Substituenten unabhängig ausgewählt aus der Gruppe Halogen, Cyano, Nitro, Alkyl, Alkoxy, Alkanoyl, Alkoxycarbonyl, Amino, Alkylamino, Alkyl- sulfonyl, Aminocarbonyl, Alkylaminocarbonyl, Aminosulfonyl und Alkyl- aminosulfonyl,
und
wobei R1 gleich Heterocyclyl gegebenenfalls substituiert sein kann mit 1 bis 3 Substituenten unabhängig ausgewählt aus der Gruppe Oxo, Alkyl, Alkoxy, Aryl, Heteroaryl, Alkanoyl und Alkylsulfonyl,

A ist eine C3-C6-Alkandiylkette, in der ein Kohlenstoffatom durch ein
Schwefelatom ersetzt ist, wobei sich zwischen dem Schwefelatom in A und dem Stickstoffatom im Uracilring mindestens 2 Kohlenstoffatome befinden müssen, und wobei sich für den Fall, dass R1 gleich Hydroxy oder Alkoxy zwischen den Schwefelatom in A und dem Sauerstoffatom in R1 mindestens 2 Kohlenstoffatome befinden müssen,
und die gegebenenfalls mit bis zu 2 Substituenten, ausgewählt aus der Gruppe Hydroxy, Alkoxy, Oxo oder Amino substituiert ist.
Die erfindungsgemäßen Verbindungen können auch in Form ihrer Salze, Solvate oder Solvate der Salze vorhegen.
Die erfindungsgemäßen Verbindungen können in Abhängigkeit von ihrer Struktur in stereoisomeren Formen (Enantiomere, Diastereomere) existieren. Die Erfindung betrifft deshalb die Enantiomeren oder Diastereomeren und ihre jeweiligen Mischungen. Aus solchen Mischungen von Enantiomeren und/oder Diastereomeren lassen sich die stereoisomer einheitlichen Bestandteile in bekannter Weise isolieren.
Die Erfindung betrifft in Abhängigkeit von der Struktur der Verbindungen auch
Tautomere der Verbindungen.
Als Salze sind im Rahmen der Erfindung physiologisch unbedenkliche Salze der erfindungsgemäßen Verbindungen bevorzugt.
Physiologisch unbedenkliche Salze der Verbindungen (I) umfassen Säureadditionssalze von Mineralsäuren, Carbonsäuren und Sulfonsäuren,. z.B. Salze der Chlorwasserstoff- säure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Ehansulfonsäure, Toluolsulfonsäure, Benzolsulfonsäure, Naphthalindisulfonsäure, Essigsäure, Propionsäure, Milchsäure, Weinsäure, Äpfelsäure, Zitronensäure, Fumar- säure, Maleinsäure und Benzoesäure.
Physiologisch unbedenkliche Salze der Verbindungen (I) umfassen auch Salze üblicher Basen, wie beispielhaft und vorzugsweise Alkalimetallsalze (z.B. Natrium- und Kaliumsalze), Erdalkalisalze (z.B. Calcium- und Magnesiumsalze) und Ammoniumsalze, abgeleitet von Ammoniak oder organischen Aminen mit 1 bis 16 C- Atomen, wie beispielhaft und vorzugsweise Ethylamin, Die ylamin, Triemylamin, Ethyldiiso- propylamin, Monoe anolarnin, Diethanolarnin, Triethanolamin, Dicyclo-hexylamin, Dimemylaminoethanol, Prokain, Dibenzylamin, N-Methylmorpholin, Dihydro- abie ylarnin, Ajginin, Lysin, E ylendiamin und Methylpiperidin.
Als Solvate werden im Rahmen der Erfindung solche Formen der Verbindungen bezeichnet, welche in festem oder flüssigem Zustand durch Koordination mit Lösungsmittelmolekülen einen Komplex bilden. Hydrate sind eine spezielle Form der Solvate, bei denen die Koordination mit Wasser erfolgt.
Im Rahmen der vorliegenden Erfindung haben die Substituenten, soweit nicht anders spezifiziert, die folgende Bedeutung:
Alkyl per se und "Alk" und "Alkyl" in Alkoxy, Alkylthio, Alkylamino, Alkanoyl, Alkanoylamino, Alkylaminocarbonyl, Alkylamiriόsulfonyl, Alkoxycarbonyl, und
Alkylsulfonyl stehen für einen linearen oder verzweigten Alkylrest mit in der Regel 1 bis 6, vorzugsweise 1 bis 4, besonders bevorzugt 1 bis 3 Kohlenstoffatomen, beispielhaft und vorzugsweise für Methyl, Ethyl, n-Propyl, Isopropyl, Butyl, Isobutyl, tert- Butyl, n-Pentyl und n-Hexyl.
Cycloalkyl umfasst polycyclische gesättigte Kohlenwasserstoffreste mit bis zu 14 C- Atomen, nämlich monocyclisches C3-C12-, vorzugsweise C3-C8-Alkyl, wie z.B. Cyclopropyl, Cyclobutyl, Cyclopentyl, Cyclohexyl, Cycloheptyl, Cyclooctyl, Cyclo- nonyl, und polycyclisches Alkyl, d.h. vorzugsweise bicyclisches und tricyclisches, gegebenenfalls spirocyclisches C7- 4- Alkyl, wie z.B. Bicyclo[2.2.1]-hept-l-yl,
Bicyclo[2.2.1]-hept-2-yl, Bicyclo[2.2.1]-hept-7-yl, Bicyclo[2.2.2]-oct-2-yl, Bicyclo- [3.2.1]-oct-2-yl5 Bicyclo[3.2.2 ]-non-2-yl und Adamantyl. .
Alkenyl steht für einen geradkettigen oder verzweigten Alkenylrest mit 2 bis 6 Kohlenstoffatomen. Bevorzugt ist ein geradkettiger oder verzweigter Alkenyhest mit 2 bis 4, besonders bevorzugt mit 2 bis 3 Kohlenstoffatomen. Beispielsweise und vorzugsweise seien genannt: Vinyl, Allyl, n-Prop-1-en-l-yl und n-But-2-en-l-yl.
Alkinyl steht für einen geradkettigen oder verzweigten Alkenyhest mit 2 bis 6 Kohlenstoffatomen. Bevorzugt ist ein geradkettiger oder verzweigter Alkenyhest mit
2 bis 4, besonders bevorzugt mit 2 bis 3 Kohlenstoffatomen. Beispielsweise und vorzugsweise seien genannt: n-Prop-1-in-l-yl und n-But-2-in-l-yl.
Alkandiyl (Alkyliden) steht für eine Kohlenstoffkette, die an beiden Termini end- ständig substituiert ist. Bevorzugt sind gesättigte Ketten mit 1 bis 6 Kohlenstoffatomen, insbesondere 2 bis 4 Kohlenstoffatomen.
Alkoxy steht beispielhaft und vorzugsweise für Methoxy, Ethoxy, n-Propoxy, Iso- propoxy, tert.-Butoxy, n-Pentoxy und n-Hexoxy.
Alkylthio steht beispielhaft und vorzugsweise für Methylthio, Ethylthio, n-Propylthio, Isopropylthio, tert.-Butylthio, n-Pentylthio und n-Hexylthio.
Alkylamino steht für einen AJ cylarninorest mit einem oder zwei (unabhängig von- einander gewählten) Alkylsubstituenten, beispielhaft und vorzugsweise für Methyl- amino, Ethylamino, n-Propylarnino, Isopropylamino, tert.-Butylamino, n-Pentylarnino, n-Hexylamino, N,N-Dimethylamino, ΛζN-Diethylamino, N-E yl-N-memylamino, N- Methyl-N-n-propylamino, N-Isopropyl-N-n-propylamino, N-t-Butyl-N-methylamino, N-Ethyl-N-n-pentylamino und N-n-Hexyl-7V-methylamino.
Alkanoyl steht beispielhaft und vorzugsweise für Acetyl und Propanoyl.
Alkanoylamino steht beispielhaft und vorzugsweise für Acetylamino und Ethyl- carbonylamino.
Alkylaminocarbonyl steht für einen Alkylarninocarbonylrest mit einem oder zwei (unabhängig voneinander gewählten) Alkylsubstituenten, beispielhaft und vorzugsweise für Me ylaminocarbonyl, EthylarninocarbonyL n-Propylaminocarbonyl, Iso- propylarninocarbonyl, tert.-Butylarninocarbonyl, n-Pentylaminocarbonyl, n-Hexyl- aminocarbonyl, N,N-Dimethylaminocarbonyl, N,N-Die ylaminocarbonyl, N-Ethyl-N- methylaminocarbonyl, N-Memyl-N-n-propylaminocarbonyl, N-Isopropyl-N-n-propyl-
aminocarbonyl, N-t-Butyl-N-methylaminocarbonyl, N-Emyl-N-n-pentylamino-carb- onyl und N-n-Hexyl-N-methylaminocarbonyl.
Alkylaminosulfonyl steht für einen Alkylaminosulfonylrest mit einem oder zwei (un- abhängig voneinander gewählten) Alkylsubstituenten, beispielhaft und vorzugsweise für Methyla inosulfonyl, Ethylaminosulfonyl, n-Propylaminosulfonyl, Isopropyl- aminosulfonyl, tert.-Butylaminosulfonyl, n-Pentylaminosulfonyl, n-Hexyl-amino- sulfonyl, N,N-Dimethylaminosulfonyl, N-Diethylaminosulfonyl, N-Ethyl-N- methylarninosulfonyl, N-Memyl-N-n-propylaminosulfonyl, N-Isopropyl-JV-n-propyl- aminosulfonyl, N-t-Butyl-N-memylaminosulfonyl, Λ Ethyl-N-n-pen1ylaminosulfonyl und N-n-Hexyl-N-methylaminosulfonyl.
Alkoxycarbonyl steht beispielhaft und vorzugsweise für Methoxycarbonyl, Ethoxy- carbonyl, n-Propoxycarbonyl, Isopropoxycarbonyl, tert.-Butoxycarbonyl, n-Pentoxy- carbonyl und n-Hexoxycarbonyl.
Alkylsulfonyl steht beispielhaft und vorzugsweise für Methylsulfonyl, Ethylsulfonyl, n-Propylsulfonyl, Isopropylsulfonyl, tert.-Butyl-sulfonyl, n-Pentylsulfonyl und n- Hexylsulfonyl.
Aryl steht für einen mono- bis tricyclischen aromatischen, carbocychschen Rest mit in der Regel 6 bis 14 Kohlenstoffatomen; beispielhaft und vorzugsweise für Phenyl, Naphthyl und Phenanthrenyl.
Heteroaryl steht für einen 5- bis 10-gliedrigen, insbesondere für einen 5- bis 6- gliedrigen aromatischen, gegebenenfalls über ein Stickstoffatom gebundenen mono- oder polycyclischen Heterocyclus mit bis zu 3 Heteroatomen aus der Reihe S, O und/oder N, beispielsweise für Pyridyl, Pyrimidyl, Thienyl, Furyl, Pyrrolyl, Thiazolyl, N-Triazolyl, Oxazolyl oder Imidazolyl. Bevorzugt sind Pyridyl, Furyl, Thiazolyl und N-Triazolyl.
Heterocyclyl (Heterocyclus) steht für einen gegebenenfalls über ein Stickstoffatom gebundenen mono- oder polycyclischen, heterocyclischen Rest mit 3 bis 11 Ringatomen und bis zu 3, vorzugsweise 1 Heteroatomen bzw. Heterogruppen aus der Reihe N, O, S, SO, SO2. Bei polycyclischen Resten können die Ringe kondensiert sein (z.B. mit einer [0]-Brücke) oder spiro-verknüpft sein. 4- bis 8-gliedriges, insbesondere 5- und 6-gliedriges Heterocyclyl ist bevorzugt. Mono- oder bicyclisches Heterocyclyl ist bevorzugt. Besonders bevorzugt ist monocyclisches Heterocyclyl. Als Heteroatome sind N und O bevorzugt. Die Heterocyclyl-Reste können gesättigt oder teilweise ungesättigt sein. Die ungesättigten Vertreter können eine oder mehrere Doppelbindungen im Ring enthalten oder, bei polycyclischen Systemen, in einem
Ring aromatisch sein, wie z.B Benzoxazin. Gesättigte Heterocyclyl-Reste sind bevorzugt. Die Heterocyclyl-Reste können über ein Kohlenstoffatom oder ein Heteroatom gebunden sein. Er kann aus zwei Substituentengruppen zusammen mit dem Stick- stoffatom, an das sie gebunden sind, gebildet werden. Besonders bevorzugt sind 5- bis 7-gliedrige, monocyclische gesättigte Heterocyclylreste mit bis zu zwei Heteroatomen aus der Reihe O, N und S. Beispielsweise und vorzugsweise seien genannt: Oxetanyl, Pyrrolidinyl, Pyrrolidinyl, Pyrrolinyl, Tetrahydrofuranyl, Tetrahydro- thienyl, Pyranyl, Piperidinyl, Thiopyranyl, Morpholinyl, Perhydroazepinyl, Thio- morpholinyl, Piperazinyl, Bicyclo[2.2.1]diazaheptyl.
Halogen steht für Fluor, Chlor, Brom und Jod.
Ein Symbol * an einer Bindung bedeutet die Verknüpfungsstelle im Molekül.
Die oben aufgeführten allgemeinen oder in Vorzugsbereichen angegebenen Restedefinitionen gelten sowohl für die Endprodukte der Formel (I) als auch entsprechend für die jeweils zur Herstellung benötigten Ausgangsstoffe bzw. Zwischenprodukte.
Die in den jeweiligen Kombinationen bzw. bevorzugten Kombinationen von Resten im einzelnen angegebenen Restedefinitionen werden unabhängig von den jeweilig
angegebenen Kombinationen der Reste beliebig auch durch Restedefinitionen anderer Kombinationen ersetzt.

wobei das Heteroaryl gegebenenfalls substituiert sein kann mit 1 bis 2 Substituenten unabhängig ausgewählt aus der Gruppe Halogen, Alkyl, Alkoxy, Alkylthio, Cycloalkyl, Aryl, Oxo, Alkanoyl, Alkanoylamino, Alkoxy- carbonyl, Amino, Alkylamino, Aminocarbonyl und Alkylaminocarbonyl,
und R und A die oben angegebene Bedeutung aufweisen.
Bevorzugt im Rahmen der vorliegenden Erfindungen sind auch Verbindungen der allgemeinen Formel (I), in welcher
R1 für Thiadiazol oder Isoxazol steht,
wobei das Thiadiazol oder Isoxazol gegebenenfalls substituiert sein kann mit 1 bis 2 Substituenten unabhängig ausgewählt aus der Gruppe Halogen, Alkyl, Alkoxy, Alkylthio, Cycloalkyl, Aryl, Oxo, Alkanoyl, Alkanoylamino,
Alkoxycarbonyl, Amino, Alkylamino, Aminocarbonyl und Alkylaminocarbonyl,
und R2 und A die oben angegebene Bedeutung aufweisen.

Der Wirkstoff kann systemisch und/oder lokal wirken. Zu diesem Zweck kann er auf geeignete Weise appliziert werden, wie z.B. oral, parenteral, pulmonal, nasal, sublingual, lingual, buccal, rectal, transdermal, conjunctival, otisch oder als Implantat.

Für diese Applikationswege kann der Wirkstoff in geeigneten Applikationsformen verabreicht werden.
Für die orale Applikation eignen sich bekannte, den Wirkstoff schnell und/oder modifiziert abgebende Applikationsformen, wie z.B. Tabletten (nichtüberzogene sowie überzogene Tabletten, z.B. mit magensaftresistenten Überzüge versehene Tabletten oder Filmtabletten), Kapseln, Dragees, Granulate, Pellets, Pulver, Emulsionen, Suspensionen und Lösungen.
Die parenterale Applikation kann unter Umgehung eines Resorptionsschrittes geschehen (intravenös,, intraarteriell, intrakardial, intraspinal oder intralumbal) oder unter Einschaltung einer Resorption (intramuskulär, subcutan, intracutan, percutan, oder intraperitoneal). Für die parenterale Applikation eignen sich als Applikationsformen u.a. Injektions- und Infusionszubereitungen in Form von Lösungen, Sus- pensionen, Emulsionen, Lyophilisaten und sterilen Pulvern.
Für die sonstigen Applikations wege eignen sich z.B. Inhalationsarzneiformen (u.a. Pulverinhalatoren, Nebulizer), Nasentropfen / -lösungen, Sprays; lingual, sublingual oder buccal zu applizierende Tabletten oder Kapseln, Suppositorien, Ohren- und Augen-präparationen, Vaginalkapseln, wässrige Suspensionen (Lotionen, Schüttelmixturen), lipophile Suspensionen, Salben, Cremes, Milch, Pasten, Streupuder oder Implantate.
Die Wirkstoffe können in an sich bekannter Weise in die angeführten Applikations- formen überführt werden. Dies geschieht unter Verwendung inerter nichttoxischer, pharmazeutisch geeigneter Hilfsstoffe. Hierzu zählen u.a. Trägerstoffe (z.B. mikrokristalline Cellulose), Lösungsmittel (z.B. flüssige Polyethylenglycole), Emulgatoren (z.B. Natriumdodecylsulfat), Dispergiermittel (z.B. Polyvinylpyrrolidon), synthetische und natürliche Biopolymere (z.B. Albumin), Stabilisatoren (z.B.Antioxi- dantien wie Ascorbinsäure), Farbstoffe (z.B. anorganische Pigmente wie Eisenoxide) oder Geschmacks- und/oder Geruchskorrigentien.
Im allgemeinen hat es sich als vorteilhaft erwiesen, bei parenteraler Applikation Mengen von etwa 0,001 bis 10 mg/kg, vorzugsweise etwa 0,01 bis 1 mg/kg Körpergewicht zur Erzielung wirksamer Ergebnisse zu verabreichen. Bei oraler Applikation beträgt die Menge etwa 0,01 bis 500 mg/kg, vorzugsweise etwa 1 bis 10 mg/kg Körpergewicht.
Trotzdem kann es gegebenenfalls erforderlich sein, von den genannten Mengen abzuweichen, und zwar in Abhängigkeit von Körpergewicht, Applikationsweg, indivi- duellem Verhalten gegenüber dem Wirkstoff, Art der Zubereitung und Zeitpunkt bzw. Intervall, zu welchem die Applikation erfolgt.
Besonders bevorzugt ist die parenterale, insbesondere die intravenöse Applikation, z.B. als iv Bolus Injektion (d.h. als Einzelgabe, z.B. per Spritze), Kurzzeitinfusion (d.h. Infusion über einen Zeitraum von bis zu einer Stunde) oder Langzeitinfusion
(d.h. Infusion über einen Zeitraum von mehr als einer Stunde). Das applizierte Volumen kann dabei in Abhängigkeit von den speziellen Bedingungen zwischen 0,5 bis 30, insbesondere 1 bis 20 ml bei der iv Bolus Injektion, zwischen 25 bis 500, insbesondere 50 bis 250 ml bei der Kurzzeitinfusion und zwischen 50 bis 1000, insbe- sondere 100 bis 500 ml bei der Langzeitinfusion betragen. Hierzu kann es vorteilhaft sein, den Wirkstoff in fester Form bereitzustellen (z.B. als Lyophilisat) und erst unmittelbar vor der Applikation im Lösungsmedium zu lösen.
Hierbei (müssen die Applikationsformen steril und pyrogenfrei sein. Sie können auf wässrigen oder Mischungen aus wässrigen und organischen Lösungsmitteln basieren.
Dazu zählen z.B. wässrige Lösungen, Mischungen aus wässrigen und organischen Lösungsmitteln (insbesondere Ethanol, Polyethylenglykol (PEG) 300 oder 400), wässrige Lösungen enthaltend Cyclodextrine oder wässrige Lösungen enthaltend Emulgatoren (grenzflächenaktive Lösungsvermittler, z.B. Lecithin oder Pluronic
F 68, Solutol HS 15, Cremophor). Bevorzugt sind hierbei wässrige Lösungen.
Für die parenterale Applikation eignen sich weitgehend isotone und euhydrische Formulierungen, z.B. solche mit einem pH- Wert zwischen 3 und 11, besonders 6 und 8, insbesondere um 7,4.
Die Verpackung der Injektionslösungen erfolgt in geeigneten Behältnissen aus Glas oder Kunststoff, z.B. in Durchstichflaschen (Vials). Diese können ein Volumen von 1 bis 1000, insbesondere 5 bis 50 ml aufweisen. Aus diesen kann die Lösung direkt entnommen und appliziert werden. Im Falle eines Lyophilisats wird sie in dem Vial aufgelöst durch Zuspritzen eines geeigneten Lösungsmittels und dann entnommen.
Die Verpackung der Infusionslösungen erfolgt in geeigneten Behältnissen aus Glas oder Kunststoff, z.B. in Flaschen oder kollabierenden Kunststoffbeuteln. Diese können ein Volumen von 1 bis 1000, insbesondere 50 bis 500 ml aufweisen.
Die vorliegende Erfindung betrifft weiterhin ein Verfahren zur Herstellung der Verbindungen der allgemeinen Formel (I) dadurch gekennzeichnet, dass Verbindungen der allgemeinen Formel (II)

ethylamin, oder andere Basen, wie z.B. N-Methylmorpholin, N-Methylpiperidin, 4- Dimethylaminopyridin oder l,8-Diazabicyclo(5.4.0)undec-7-en, bevorzugt ist 1,8- Diazabicyclo(5.4.0)undec-7-en.
Die Verbindungen der allgemeinen Formel (III) sind bekannt oder lassen sich nach bekannten Verfahren aus den entsprechenden Edukten synthetisieren.

Die Verbindungen der allgemeinen Formel (V) sind bekannt oder lassen sich nach bekannten Verfahren aus den entsprechenden Edukten synthetisieren.
Zur Herstellung der Verbindungen der allgemeinen Formel (IV), setzt man Verbindungen der allgemeinen Formel (VI),

Die Verbindungen der allgemeinen Formel (VII) sind bekannt oder lassen sich nach bekannten Verfahren aus den entsprechenden Edukten synthetisieren.
Ein Herstellungsweg soll durch folgendes Schema 1 beispielhaft erläutert werden:

Die erfindungsgemäßen Verbindungen zeigen ein nicht vorhersehbares, wertvolles pharmakologisches und pharmakokinetisches Wirkspektrum. Sie eignen sich daher zur Verwendung als Arzneimittel zur Behandlung und/oder Prophylaxe von Krankheiten bei Menschen und Tieren.
Besonders wirksam sind die erfindungsgemäßen Verbindungen gegen Bakterien und bakterienähnliche Mikroorganismen, insbesondere Gram-positive Bakterien. Sie sind daher besonders gut zur Prophylaxe und Chemotherapie von lokalen und gegebenenfalls systemischen Infektionen in der Human- und Tiermedizin geeignet, die durch diese Erreger hervorgerufen werden.
Die Verbindungen der vorliegenden Erfindung zeichnen sich durch ein breites Wirkspektrum gegenüber Gram-positiven Bakterien aus, wobei auch multiresistente Keime erfasst werden können, insbesondere Staphylokokken, Pneumokokken und Enterokokken einschließlich Vancomycin-resistenter Stämme.
Beispielsweise können lokale und/oder systemische Erkrankungen behandelt und/oder verhindert werden, die durch die folgenden Erreger oder durch Mischungen der folgenden Erreger verursacht werden:
Gram-positive Kokken, z.B. Staphylokokken (Staph. aureus, Staph. epidermidis) und Streptokokken (Strept. agalactiae, Strept. faecalis, Strept. pneumoniae, Strept. pyogenes) sowie strikt anaerobe Bakterien wie z.B. Clostridium, ferner Myko- plasmen (M. pneumoniae, M. hominis, M. urealyticum).
Die obige Aufzählung von Erregern ist lediglich beispielhaft und keineswegs beschränkend aufzufassen. Als Krankheiten, die durch die genannten Erreger oder Mischinfektionen verursacht und durch die erfindungsgemäßen Verbindungen verhindert, gebessert oder geheilt werden können, seien beispielsweise genannt:
Infektionskrankheiten beim Menschen wie z.B. septische Infektionen, Knochen- und Gelenkinfektionen, Hautinfektionen, postoperative Wundinfektionen, Abszesse, Phlegmone, Wundinfektionen, infizierte Verbrennungen, Brandwunden, Infektionen im Mundbereich, Infektionen nach Zahnoperationen, septische Arthritis, Mastitis, Tonsillitis, Genital-Infektionen und Augeninfektionen.
Außer beim Menschen können bakterielle Infektionen auch bei anderen Spezies behandelt werden. Beispielhaft seien genannt:
Schwein: Sepsis, Metritis-Mastitis-Agalaktiae-Syndrom, Mastitis;
Wiederkäuer (Rind, Schaf, Ziege): Sepsis, Bronchopneumonie, Mykoplasmose, Genitalinfektionen;
Pferd: Bronchopneumonien, puerperale und postpuerperale Infektionen;
Hund und Katze: Bronchopneumonie, Dermatitis, Otitis, Harnwegsinfekte, Prostatitis;
Geflügel (Huhn, Pute, Wachtel, Taube, Ziervögel und andere): Mycoplasmose, chronische Luftwegserkrankungen, Psittakose.
Ebenso können bakterielle Erkrankungen bei der Aufzucht und Haltung von Nutz- und Zierfischen behandelt werden, wobei sich das antibakterielle Spektrum über die vorher genannten Erreger hinaus auf weitere Erreger wie z.B. Brucella, Campylo- bacter, Listeria, Erysipelothris, Nocardia erweitert.
A. Bewertung der pharmakologischen Wirksamkeit
in vitro-Wirkuns.
Die in vzYro-Wirkung der erfindungsgemäßen Verbindungen kann in folgenden Assays gezeigt werden:
Klonierung, Expression und Reinigung von Pol III aus S. aureus Zur Klonierung von polC mit einem N-terminalem His-Tag wird das Strukturgen polC mit Hilfe der PCR aus genomischer DNA von S. aureus amphfiziert. Mit Hilfe der Primer SAPoM 5'-GCGCCATATGGACAGAGCAACAAAAATTTAA-3' und SAPolrev 5'-GCGCGGATCCTTACATATCAAATATCGAAA-3' werden die Restriktionsschnittstellen Ndel und BamHI vor bzw. hinter dem amplifizierten Gen ein- geführt. Nachdem das 4300 bp große PCR-Produkt mit Ndel und BamHI verdaut worden ist, wird es in den ebenfalls mit Ndel und BamHI verdauten Vektor pET15b (Novagen, USA) ligiert und in E. coli XL-1 Blue transformiert.
Nach Transformation in E. coli BL21(DE3) werden die Zellen zur Expression von PolC bei 30°C in LB-Medium mit 100 μg/ml Ampicillin bis zu einer OD595nm von
0,5 kultiviert, auf 18°C abgekühlt und nach Zusatz von 1 mM IPTG 20 Stunden weiter inkubiert. Die Zellen werden durch Zentrifugation geerntet, einmal in PBS mit
1 mM PMSF gewaschen und in 50 mM NaH2PO4 pH 8,0, 10 mM Imidazol, 2 mM ß-
Mercaptoethanol, 1 mM PMSF, 20 % Glycerin aufgenommen. Die Zellen werden mit Hilfe einer French Press bei 12.000 psi aufgeschlossen, die Zelltrümmer abzentri- fugiert (27.000xg, 120 min, 4°C) und der Überstand mit einer entsprechenden
Menge Ni-NTÄ-Agarose (Fa. Quiagen, Deutschland) 1 Stunde bei 4°C gerührt. Nach dem Einfüllen in eine Säule wird die Gelmatrix mit 50 mM NaH PO4 pH 8,0, 2 mM ß-Mercaptoethanol, 20 mM Imidazol, 10 % Glycerin gewaschen und das gereinigte Protein wird anschließend mit demselbem Puffer, welcher 100 mM Imidazol enthält,
eluiert. Das gereinigte Protein wird mit 50 % Glycerin versetzt und bei -20°C gelagert.
Bestimmung der ICgn gegen Polymerase III Die Aktivität von PolC wird in einer enzymatisch gekoppelten Reaktion gemessen, wobei das während der Polymerisation gebildete Pyrophosphat mit Hilfe der ATP- Sulfurylsae in ATP umgewandelt wird, welches mit Hilfe der Firefly-Luciferase de- tektiert wird. Der Reaktionsansatz enthält in einem Endvolumen von 50 μl 50 mM Tris/Cl pH 7,5; 5 mM DTT, 10 mM MgCl2, 30 mM NaCl, 0,1 mg/ml BSA, 10 % Glycerol, je 20 μM dATP, dTTP, dCTP, 2U/ml aktivierte Kalbsthymus-DNA (Fa.
Worthington, USA), 20 μM APS und 0,06 mM Luciferin. Die Reaktion wird durch Zusatz von gereinigter PolC in einer Endkonzentration von ~2 nM, gestartet und 30 min bei 30°C inkubiert. Die Menge an gebildetem Pyrophosphat wird anschließend durch Zugabe von ATP-Sulfurylase (Sigma, USA) in einer Endkonzen- tration von 5 nM und Inkubation bei 30°C für 15 min in ATP umgewandelt. Nach
Zugabe von 0,2 nM Firefly-Luciferase wird die Lumineszenz in einem Luminometer für 60 s gemessen. Als IC50 wird die Konzentration eines Inhibitors angegeben, die zu einer 50 %-igen Inhibition der Enzymaktivität von PolC führt.

Bestimmung der minimalen Hemmkonzentrationen (MHK) gegenüber einer Reihe von Keimen
Die MHK- Werte gegen verschiedene Bakterienstämme (S. aureus, S. pneumoniae, E. faecalis) werden unter Verwendung der Mikrodilutionsmethode in BHI-Bouillon durchgeführt. Die Bakterienstämme werden über Nacht in BHI-Bouillon (Staphylokokken) bzw. BHI-Bouillon + 10 % Rinderserum (Streptokokken, Enterokokken) angezüchtet. Die Testsubstanzen werden in einem Konzentrationbereich von 0,5 bis 256 μg/ml geprüft. Nach serieller Verdünnung der Testsubstanzen werden die Microtiterplatten mit den Testkeimen beimpft. Die Keimkonzentration liegt bei ca. lxlO6 Keimen/ml Suspension. Die Platten werden bei 37°C unter 8 % CO2 (für Streptokokken, Enterokokken) für 20 h inkubiert. Als MHK- Wert wird die niedrigste Konzentration protokolliert, bei der das sichtbare Wachstum der Bakterien vollständig inhibiert wird.

Die Eignung der erfindungsgemäßen Verbindungen zur Behandlung bakterieller Infektionen kann in folgenden Tiermodellen gezeigt werden:
Systemische Infektion mit S.aureus 133
S. aureus 133 Zellen werden über Nacht in BH-Bouillon angezüchtet. Die Übernachtkultur wird 1:100 in frische BH-Bouillon verdünnt und für 3 Stunden hochgedreht. Die in der logarithmischen Wachstumsphase befindlichen Bakterien werden abzentri- fugiert und 2 x mit gepufferter, physiologischer Kochsalzlösung (303) gewaschen. Danach wird am Photometer (Modell LP 2W, Fa. Dr. Lange, Deutschland) eine Zellsuspension mit einer Extinktion von 50 Einheiten in 303 eingestellt. Nach einem Verdünnungsschritt (1:15) wird diese Suspension 1:1 mit einer 10 %-igen Mucinsus- pension gemischt. Von dieser Infektionslösung werden 0,25 ml/20 g Maus ip appli- ziert. Dies entspricht einer Zellzahl von etwa 1 x 10E6 Keimen/Maus. Die ip-
Therapie erfolgt 30 Minuten nach der Infektion. Für den Infektionsversuch werden weibliche CFWl -Mäuse verwendet. Das Überleben der Tiere wird über 6 Tage protokolliert.

Ausgangsverbindungen
Beispiel I
7-Amino-2,3 -dihydro-5H-[ 1 ,3]thiazolo [3 ,2-a]pyrimidin-5-on
27,22 g (144,9 mmol) 1,2-Dibromethan und 85,84 g (263,46 mmol) Cäsiumcarbonat werden in 125 ml Dimethylformamid unter Argon suspendiert. Dann werden innerhalb von 30 Minuten 18,86 g (131,73 mmol) 6-Amino-4-hydroxy-2-mercaptopyrimi- din zugegeben. Man rührt 4 Stunden bei Raumtemperatur und erwärmt dann über Nacht auf 60°C. Die Reaktionslösung wird abgekühlt und im Vakuum eingeengt. Der
Rückstand wird mit 100 ml Wasser versetzt. Man rührt 10 Minuten nach und lässt dann noch 10 Minuten stehen. Der Niederschlag wird abgesaugt, zweimal mit 25 ml
Wasser nachgewaschen und im Hochvakuum getrocknet. Man erhält 15,8 g (71 % d.Th.) Produkt.
LC-MS (Methode 2): Rt = 0,53 min MS (ESIpos): m/z = 170 (M+H)+
Beispiel II
8-Amino-3 ,4-dihydro-2H,6H-pyrimido [2, 1 -b] [ 1 ,3]thiazin-6-on
Die Darstellung erfolgt wie für Beispiel I aus 11,26 g (69,85 mmol) 6-Amino-4- hydroxy-2-mercaptopyrimidin, 16,22 g (80,32 mmol) 1,3-Dibrompropan und 45,52 g (136,69 mmol) Cäsiumcarbonat. Man erhält 12 g (93 % d. Th.) Produkt. LC-MS (Methode 1): Rt = 0,64 min MS (ESIpos): m z = 184 (M+H)+
Beispiel III
6-Amino-3-(2-sulfanylethyl)-2,4(lH,3H)-pyrimidindion
15,5 g (91,6 mmol) der Verbindung aus Beispiel I und 14,66 g (366,42 mmol)
Natriumhydroxid werden zusammen in 100 ml Wasser suspendiert und auf Rückfluss
2 Stunden erhitzt. Es entsteht eine klare Lösung. Man kühlt die Reaktionslösung auf 0°C ab und neutralisiert mit konzentrierter Salzsäure. Es wird eine Stunde bei 0°C
nachgerührt. Dann filtriert man den Niederschlag ab, wäscht zweimal mit je 20 ml
Wasser und Diethylether nach und trocknet über Nacht im Vakuumtrockenschrank bei 60°C. Man erhält 13,93 g (81 % d. Th.) Produkt.
1H-NMR (200MHz, DMSO-d6) δ = 3.79 (t, 2H), 4.54 (d, 1H), 6.23 (s, 2H), 10.42 (s,

Beispiel IV
6-Amino-3-(3-sulfanylpropyl)-2,4(lH,3H)-pyrimidindion
Die Darstellung erfolgt wie für Beispiel III aus 4,3 g (23,47 mmol) der Verbindung aus Beispiel II und 3,75 g (93,87 mmol) Natriumhydroxid. Man erhält 1,52 g (32 % d. Th.) Produkt.
LC-MS (Methode 1): Rt = 0,94 min MS (ESIpos): m/z = 218 (M+H)+
Beispiel V
6-(2,3-Dmydro-lH-inden-5-ylamino)-3-(2-sulfanylethyl)-2,4(lH,3H)-pyrimidindion
3 g (16,02 mmol) der Verbindung aus Beispiel HI, 3,2 g (24,04 mmol) 5-Aminoindan und 4,08 g (24,04 mmol) 5-Aminoindan Hydrochlorid werden zusammen 6 Stunden auf 160°C erhitzt. Nach dem Abkühlen auf Raumtemperatur wird die Reaktionslösung mit 50 %igem Ethanol verrührt und der Niederschlag abgesaugt. Man wäscht noch einmal mit 20 ml 50 %igem Ethanol und zweimal mit je 20 ml Diethylether nach und trocknet im Hochvakuum. Man erhält 4,71 g (97 % d. Th.) Produkt. 1H-NMR (300MHz, DMSO-d6) δ = 2.02 (quintett, 2H), 2.59 (q, 2H), 2.84 (q, 4H), 3.83 (t, 2H), 4.71 (s, 1H), 6.95 (d, 1H), 7.06 (s, 1H), 7.22 (d, 1H), 8.17 (s, 1H), 10.43 (s, 1H)
LC-MS (Methode 2): Rt = 3,53 min MS (ESIpos): m/z = 304 (M+H)+
Beispiel VI
6-[(3-Ethyl-4-methylphenyl)amino]-3-(2-sulfanylethyl)-2,4(lH,3H)-pyrimidindion
Die Darstellung erfolgt wie für Beispiel V aus 3 g (16,02 mmol) der Verbindung aus Beispiel III, 5,78 g (33,65 mmol) 3-Ethyl-4-methylanilin Hydrochlorid und 2,93 ml (16,82mmol) N,N-Diisopropylethylamin. Man erhält 4,83 g (98 % d. Th.) Produkt. 1H-NMR (300MHz, DMSO-d6) δ = 1.14 (t, 3H), 2.24 (s, 3H), 2.53-2.63 (m, 4H), 3.83 (t, 2H), 4.71 (s, 1H), 6.91-6.98 (m, 2H), 7.14 (d, 1H), 8.1 (s, 1H), 10.41 (s, 1H) LC-MS (Methode 3): Rt = 2,39 min MS (ESIpos): m/z = 306 (M+H)+
Beispiel VII
6-[(3-Ethyl-4-methylphenyl)amino]-3-(3-sulfanylpropyl)-2,4(lH,3H -pyrimidindion
Die Darstellung erfolgt wie für Beispiel V aus 2,98 g (14,81 mmol) der Verbindung aus Beispiel IV, 7,63 g (44,42 mmol) 3-Ethyl-4-methylanilin Hydrochlorid und 3,87 ml (22,21 mmol) N,N-Diisopropylethylamin. Man erhält 4,25 g (90 % d. Th.) Produkt.
1H-NMR (200MHz, DMSO-d6) δ = 1.13 (t, 3H), 1.7-1.83 (m, 2H), 2.23 (s, 3H), 3.77 (t, 2H), 4.72 (s, IH), 6.97 (s, IH), 7.05 (dd, 2H), 8.15 (s, IH), 10.48 (s, IH), 4H liegen noch unter dem DMSO-Peak LC-MS (Methode 1): Rt = 3,7 min MS (ESIpos): m/z = 320 (M+H)+
Herstellungsbeispiele
Beispiel 1
6-(2,3-Dihydro-lH-inden-5-ylamino)-3-(2-{[(5-phenyl-l,3,4-oxadiazol-2-yl)methyl] sulfanyl} ethyl)-2,4(lH,3H)-pyrimidindion
40 mg (0,13 mmol) der Verbindung aus Beispiel V und 28,23 mg (0,15 mmol) 2- (Chlormethyl)-5-phenyl-l,3,4-oxadiazol werden in 3 ml Dimethylformamid vorgelegt. Dann gibt man 40,14 mg (0,26 mmol) l,8-Diazabicyclo(5.4.0)undec-7.-en hinzu und rührt 3 Stunden bei Raumtemperatur. Die Reaktionslösung wird im Vakuum eingeengt. Der Rückstand wird in Dichlormethan aufgenommen und einmal mit IN Salzsäure und einmal mit gesättigter Natriumchloridlösung gewaschen. Die orga- nische Phase trocknet man über Natriumsulfat und engt im Vakuum ein. Nach dem
Trocknen im Hochvakuum erhält man 5.6,8 mg (84 % d. Th.) Produkt. LC-MS (Methode 2) : Rt = 3,97 min MS (ESIpos): m/z *= 462 (M+H)+
Die in der folgenden Tabelle aufgeführten Beispiele können analog den oben beschriebenen Vorschriften aus den entsprechenden Ausgangsverbindungen hergestellt werden.
C. Ausftthrungsbeispiele für pharmazeutische Zusammensetzungen
Die erfindungsgemäßen Substanzen können folgendermaßen in pharmazeutische Zubereitungen überführt werden:
Tablette:
Zusammensetzung:
100 mg der Verbindung von Beispiel 1, 50 mg Lactose (Monohydrat), 50 mg Mais- stärke (nativ), 10 mg Polyvinylpyrolidon (PVP 25) (Fa. BASF, Ludwigshafen,
Deutschland) und 2 mg Magnesiumstearat.
Tablettengewicht 212 mg, Durchmesser 8 mm, Wölbungsradius 12 mm.
Herstellung:
Die Mischung aus Wirkstoff, Lactose und Stärke wird mit einer 5 %-igen Lösung (m/m) des PVPs in Wasser granuliert. Das Granulat wird nach dem Trocknen mit dem Magnesiumstearat für 5 min gemischt. Diese Mischung wird mit einer üblichen Tablettenpresse verpresst (Format der Tablette siehe oben). Als Richtwert für die Verpressung wird eine Presskraft von 15 kN verwendet.
Oral applizierbare Suspension:
Zusammensetzung: 1000 mg der Verbindung von Beispiel 1, 1000 mg Ethanol (96 %), 400 mg Rhodigel
(Xanthan gu der Fa. FMC, Pennsylvania, USA) und 99 g Wasser.
Einer Einzeldosis von 100 mg der erfindungsgemäßen Verbindung entsprechen 10 ml orale Suspension.
Herstellung:
Das Rhodigel wird in Ethanol suspendiert, der Wirkstoff wird der Suspension zugefügt. Unter Rühren erfolgt die Zugabe des Wassers. Bis zum Abschluss der Quellung des Rhodigels wird ca. 6h gerührt.
Intravenös applizierbare Lösung:
Zusammensetzung:
1 mg der Verbindung von Beispiel 1, 15 g Polyethylenglykol 400 und 250 g Wasser für Inj ektionszwecke.
Herstellung:
Die Verbindung von Beispiel 1 wird zusammen mit Polyethylenglykol 400 in dem Wasser unter Rühren gelöst. Die Lösung wird sterilfiltriert (Porendurchmesser 0,22 μm) und unter aseptischen Bedingungen in hitzesterilisierte Infusionsflaschen abgefüllt. Diese werden mit Infusionsstopfen und Bördelkappen verschlossen.

Patentansprüche
1. Verbindungen der Formel
in welcher
R für Hydroxy, Alkoxy, Alkenyl, Cycloalkyl, Aryl, Heterocyclyl oder Heteroaryl steht,
wobei R1 gleich Aryl gegebenenfalls substituiert sein kann mit 1 bis 3 Substituenten unabhängig ausgewählt aus der Gruppe Halogen, Cyano, Nitro, Alkyl, Alkoxy, Alkanoyl, Alkoxycarbonyl, Amino, Alkylamino, Alkylsulfonyl, Aminocarbonyl, Alkylaminocarbonyl, Aminosulfonyl und Alkylaminosulfonyl,
und
wobei R1 gleich Heterocyclyl gegebenenfalls substituiert sein kann mit 1 bis 3 Substituenten unabhängig ausgewählt aus der Gruppe Oxo, Alkyl, Alkoxy, Aryl, Heteroaryl, Alkanoyl und Alkylsulfonyl,
worin Aryl gegebenenfalls substituiert sein kann mit 1 bis 3 Substituenten unabhängig ausgewählt aus der Gruppe Halogen, Nitro, Alkyl und Alkoxy,
und
wobei R1 gleich Heteroaryl gegebenenfalls substituiert sein kann mit 1 bis 3 Substituenten unabhängig ausgewählt aus der Gruppe Halogen, Alkyl, Alkoxy, Alkylthio, Cycloalkyl, Aryl, Oxo, Alkanoyl, Alkanoylamino, Alkoxycarbonyl, Amino, Alkylamino, Aminocarbonyl und Alkylaminocarbonyl,
R2 ein Substituent der folgenden Formel
worin
R^-l und R2-2 unabhängig voneinander ausgewählt werden aus der Gruppe -Ce-Alkyl, C2-C6-Alkenyl, C2-C6-Alkinyl, C3-C6- Cycloalkyl und Halogen, oder
R^-l und R2-2 bilden zusammen mit dem Kohlenstoffatom, an das sie gebunden sind, einen C3-C6-Cycloalkyl- oder Heterocyclyl- Ring, der gegebenenfalls mit bis zu 3 Halogen substituiert sein kann,
und .
A ist eine C3-C6-Alkandiylkette, in der ein Kohlenstoffatom durch ein Schwefelatom ersetzt ist, wobei sich zwischen dem Schwefelatom in A und dem Stickstoffatom im Uracilring mindestens 2 Kohlenstoffatome befinden müssen, und wobei sich für den Fall, dass R1 gleich Hydroxy oder Alkoxy zwischen den Schwefelatom in A und dem
Sauerstoffatom in R1 mindestens 2 Kohlenstoffatome befinden müssen, und die gegebenenfalls mit bis zu 2 Substituenten, ausgewählt aus der Gruppe Hydroxy, Alkoxy, Oxo oder Amino substituiert ist.
Verbindungen der Formel (I) nach Anspruch 1, in welcher
R1 für Aryl, Heterocyclyl oder Heteroaryl steht,
wobei R1 gleich Aryl gegebenenfalls substituiert sein kann mit 1 bis 2 Substituenten unabhängig ausgewählt aus der Gruppe Halogen,
Cyano, Nitro, Alkyl, Alkoxy, Alkanoyl und Amino,
und
wobei R1 gleich Heterocyclyl gegebenenfalls substituiert sein kann mit 1 bis 3 Substituenten unabhängig ausgewählt aus der Gruppe Oxo, Alkyl, Alkoxy, Aryl, Heteroaryl, Alkanoyl und Alkylsulfonyl,
worin Aryl gegebenenfalls substituiert sein kann mit 1 bis 2 Sub- stituenten unabhängig ausgewählt aus der Gruppe Halogen, Nitro,
- Alkyl und Alkoxy,
und
wobei R1 gleich Heteroaryl gegebenenfalls substituiert sein kann mit 1 bis 2 Substituenten unabhängig ausgewählt aus der Gruppe Halogen, Alkyl, Alkoxy, Alkylthio, Cycloalkyl, Aryl, Oxo, Alkanoyl, Alkanoylamino, Alkoxycarbonyl, Amino, Alkylamino, Aminocarbonyl und Alkylaminocarbonyl,
R2 . ein Substituent der folgenden Formel
worin
R2-1 und R2-2 unabhängig voneinander ausgewählt werden aus der Gruppe CrC6-Alkyl, C2-C6-Alkenyl, C2-C6-Alkinyl, C3-C6- Cycloalkyl und Halogen, oder
R -1 und R -2 bilden zusammen mit dem Kohlenstoffatom, an das sie gebunden sind, einen C3-C6-Cycloalkyl- oder Heterocyclyl- Ring, der gegebenenfalls mit bis zu 3 Halogen substituiert sein kann,
und
A ist eine Cs-Cg-Alkandiylkette, in der ein Kohlenstoffatom durch ein Schwefelatom ersetzt ist, wobei sich zwischen dem Schwefelatom in A und dem Stickstoffatom im Uracilring mindestens 2 Kohlenstoffatome befinden müssen, und wobei sich für den Fall, dass R1 gleich Hydroxy oder Alkoxy zwischen den Schwefelatom in A und dem Sauerstoffatom in R1 mindestens 2 Kohlenstoffatome befinden müssen, und die gegebenenfalls mit bis zu 2 Substituenten, ausgewählt aus der Gruppe Hydroxy oder Oxo substituiert ist.
Verbindungen der Formel (I) nach Anspruch 1 oder 2, in welcher
R1 für Heteroaryl steht,
wobei R1 gleich Heteroaryl gegebenenfalls substituiert sein kann mit 1 bis 2 Substituenten unabhängig ausgewählt aus der Gruppe Halogen,
Alkyl, Alkoxy, Alkylthio, Cycloalkyl, Aryl, Oxo, Alkanoyl, Alkanoylamino, Alkoxycarbonyl, Amino, Alkylamino, Aminocarbonyl und Alkylaminocarbonyl.
Verbindungen der Formel (I) nach einem der Ansprüche 1 bis 3, in welcher
R2 ausgewählt wird aus der Gruppe

Verfahren zur Herstellung der Verbindungen der Formel (I) durch Umsetzung von Verbindungen der Formel
in welcher
R die in Anspruch 1 angegebene Bedeutung hat, und
A1 für den Teil von A steht, der sich zwischen dem Schwefelatom und dem Uracilring befindet,
mit Verbindungen der allgemeinen Formel (HI)
in welcher
R1 die in Anspruch 1 angegebene Bedeutung hat,
A für den Teil von A steht, der sich zwischen dem Schwefelatom und dem Rest R1 befindet,
und
X1 für Halogen, bevorzugt Chlor, Brom oder Iod, steht.
Verbindungen nach einem der Ansprüche 1 bis 6 zur Behandlung und/oder Prophylaxe von Krankheiten.
Arzneimittel enthaltend mindestens eine Verbindung nach einem der Ansprüche 1 bis 6 in Kombination mit mindestens einem pharmazeutisch verträglichen, pharmazeutisch unbedenklichen Träger oder Exzipienten.
10. Verwendung von Verbindungen nach einem der Ansprüche 1 bis 6 zur Herstellung eines Arzneimittels zur Behandlung und oder Prophylaxe bakterieller Infektionen.
11. Arzneimittel nach Anspruch 9 zur Behandlung und/oder Prophylaxe bakte- rieller Infektionen.
12. Verfahren zur Bekämpfung bakterieller Infektionen in Menschen und Tieren durch Verabreichung einer antibakteriell wirksamen Menge mindestens einer Verbindung nach einem der Ansprüche 1 bis 6.
