Abstract

On a isolé et séquencé des ADNc codant la myrcène synthase, (-)-limonène synthase et (-)-pinène synthase tirée du sapin grandissime (Abies grandis) et on a déterminé les séquences d'acides aminés correspondantes. Par conséquent, l'invention concerne les séquences d'ADN isolées (SEQ ID NO:1; SEQ ID NO:3 ET SEQ ID NO:5) codant pour l'expresssion de myrcène synthase (SEQ ID NO:2), (-)-pinène synthase (SEQ ID NO:4) et (-)-limonène synthase (SEQ ID NO:6), respectivement, tirées du sapin grandissime (Abies grandis). Dans d'autres aspects, l'invention concerne des véhicules de clonage recombinés réplicables codant pour la myrcène synthase, la (-)-limonène synthase et la (-)-pinène synthase, ou pour une séquence de base suffisamment complémentaire d'au moins une partie d'ADN ou d'ARN de myrcène synthase, de (-)-limonène synthase ou de (-)-pinène synthase afin de permettre une hybridation avec ceux-ci. Dans d'autres aspects, l'invention concerne des cellules hôtes modifiées ayant été transformées, transfectées, infectées et/ou injectées avec un véhicule de clonage recombiné et/ou une séquence d'ADN codant la myrcène synthase, la (-)-limonène synthase ou la (-)-pinène synthase. Ainsi, on obtient des systèmes et des procédés d'expression recombinée de monoterpènes synthases recombinées pré-citées pouvant être utilisées pour faciliter leur reproduction, isolement et purification en quantités significatives. On peut utiliser les myrcène synthase (-)-limonène synthase ou (-)-pinène synthase recombinées pour obtenir une expression ou une expression améliorée de myrcène synthase, (-)-limonène synthase ou (-)-pinène synthase dans des plantes afin d'améliorer la production de monoterpènoïdes, ou on peut également les utiliser à la régulation ou à l'expression de myrcène synthase, (-)-limonène synthase et (-)-pinène synthase ou dans la production de leurs produits.
