The same figure shows the ability of HFt-Glu-MP-PASE to encapsulate the free drugs mitoxantrone or Genz 644282. All of these drugs were encapsulated in the protein cavity of the fusion proteins using a protein uncoupling-coupling process as a function of pH according to the protocol disclosed in Falvo et al., 2016, Biomacromolecules. 17(2):514 22, with one modification. In reactions using molecules containing maleimide, they were added at pH 6.5 to 7.5 after an acidic pH step to avoid possible damage to the molecule due to low pH values. Moreover, in the reaction using maleimidopropionic acid linker and drug Genz 644282, the former was added before the drug at a linker/thiol ratio of 1.2:1.

Once the tumor size reached a volume of approximately 100 mm 3 , mice were randomized into groups of six animals and intravenously injected with 200 μl of saline, Genz-644282 (1.9 mg/kg) or HFt-Glu-MP-PASE-Genz (0 .95 or 1.9 mg/kg). Mice received intravenous injections twice a week for three weeks; tumor volume was measured twice a week using a digital caliper, and the weight of the mice was monitored. After the tumor size of the mice reached a volume of >1500 mm 3 , the animals were sacrificed. In fig. 15 shows tumor growth curves approximately two weeks after the last treatment. This figure clearly demonstrates the ability of HFt-Glu-MP-PASE-Genz to dramatically reduce cancer progression, with a 100% complete tumor response rate. In fig. Figure 16 shows the overall survival of the animal. All animals treated with HFt-Glu-MP-PASE-Genz were free of disease and alive 100 days after initiation of therapy, indicating the high efficacy of the test compound.
