Claims (15)

1. Filamentøs svampecelle som omfatter en komplet eller delvis inaktivering af den filamentøse svampecelles atg11-aktivitet, hvor cellen omfatter en ekspressionsvektor, som omfatter en sekvens af nukleotider, som koder for et heterologt enzym af interesse, og genetiske elementer, der er egnede til fremstilling af det heterologe enzym i den filamentøse svampecelle.
2. Filamentøs svampecelle ifølge krav 1, hvor det heterologe enzym af interesse er valgt fra gruppen bestående af en peptidase, (carboxypeptidase, aminopepetidase, protease), amylase (f.eks., alfa-amylase eller glucoamylase), carbohydrase, pullulanase, katalase, cellulase, chitinase, cutinase, cyclodextrin-glycosyltransferase, deoxyribonuclease, esterase, endo-glucosidase, alfa-galaktosidase, beta-galaktosidase, alfa-glucosidase, beta-glucosidase, invertase, laccase, lipase, phospholipase, mannosidase, mutanase, oxidase, pektinolytisk enzym, peroxidase, phytase, polyphenoloxidase, ribonuclease, transglutaminase og xylanase.
3. Filamentøs svampecelle ifølge krav 1, hvor det heterologe enzym af interesse er en glucoamylase.
4. Filamentøs svampecelle ifølge krav 1, hvor det heterologe enzym af interesse er en amylase.
5. Filamentøs svampecelle ifølge ethvert af de foregående krav, hvor den filamentøse svampecelle er valgt fra gruppen bestående af Acremonium, Aspergillus, Fusarium, Humicola, Mucor, Myceliophthora, Neurospora, Penicillium, Thielavia, Tolypocladium og Trichoderma.
6. Filamentøs svampecelle ifølge ethvert af de foregående krav, hvor den filamentøse svampecelle er valgt fra gruppen bestående af Aspergillus awamori, Aspergillus foetidus, Aspergillus japonicus, Aspergillus nidulans, Aspergillus niger, Aspergillus oryzae, Fusarium bactridioides, Fusarium cerealis, Fusarium crookweiiense, Fusarium culmorum, Fusarium gram-inearum, Fusarium graminum, Fusarium heterosporum, Fusarium negundi, Fusarium oxyspo-rum, Fusarium reticulatum, Fusarium roseum, Fusarium sambucinum, Fusarium sarcochroum, Fusarium sporotrichioides, Fusarium sulphureum, Fusarium torulosum, Fusarium trichothecioi-des, Fusarium venenatum, Humicola insolens, Humicola lanuginosa, Mucor miehei, Myceliophthora thermophila, Neurospora crassa, Penicillium purpurogenum, Thielavia terrestris, Trichoderma harzianum, Trichoderma koningii, Trichoderma longibrachiatum, Trichoderma ree-sei og Trichoderma viride-celle.
7. Filamentøs svampecelle ifølge ethvert af de foregående krav, hvor den filamentøse svampecelle er en Asperg/7/us-celle.
8. Filamentøs svampecelle ifølge ethvert af de foregående krav, hvor den filamentøse svampecelle er en Aspergillus niger-celle eller Aspergillus oryzae-celle.
9. Filamentøs svampecelle ifølge ethvert af de foregående krav, som omfatter en deletion eller disruption af de regulatoriske sekvenser, som ligger opstrøms eller nedstrøms for atg 11-genet.
10. Filamentøs svampecelle ifølge ethvert af kravene 1-9, som omfatter en deletion eller disruption af atg11 -genet.
11. Filamentøs svampecelle ifølge ethvert af de foregående krav, som omfatter en deletion af atg11-genet.
12. Filamentøs svampecelle ifølge ethvert af de foregående krav, hvor svampecellen yderligere omfatter en modificering for at forøge tilstedeværelsen af et had UPR moduleringsprotein i den filamentøse svampeværtscelle.
13. Filamentøs svampecelle ifølge ethvert af de foregående krav, hvor svampecellen yderligere omfatter en modificering for at forøge tilstedeværelsen af et ire1 UPR moduleringsprotein i den filamentøse svampeværtscelle.
14. Fremgangsmåde til fremstilling af et heterologt polypeptid af interesse omfattende dyrkning af en filamentøs svampecelle ifølge ethvert af de foregående krav under betingelser, der er fremmende for at fremkalde ekspression af et heterologt polypeptid af interesse.
15. Fremgangsmåde ifølge krav 14, som yderligere omfatter genindvinding af polypeptidet af interesse fra værtscelle-dyrkningsmediet.
