Claims (12)
1. Kulturmedium omfattende: a) noggin eller fragmenter deraf, der inhiberer BMP-signalvejen; og b) en inhibitor af Transforming Growth Factor (TGF)/aktivin/nodal-signalvejen valgt fra gruppen bestående af SB431542, Lefty-A og Cerberus og fragmenter af Lefty-A og Cerberus, der inhiberer TGF/aktivin/nodal-signalvejen.
2. Kulturmedium ifølge krav 1, hvor kulturmediet yderligere omfatter : a) en kulstofkilde; b) essentielle aminosyrer; c) vitaminer: d) en purin og en pyrimidin; e) uorganiske salte; f) selenium; g) en antioxidant; h) en phospholipid prækursor; i) en unik fedtsyre; og j) et bærerprotein.
3. In vitro-fremgangsmåde til fremstilling afen population af neurale prækursorer, hvor fremgangsmåden omfatter trinnet at dyrke pluripotente celler med kulturmediet som defineret i et hvilket som helst af kravene 1 til 2.
4. In v/fro-fremgangsmåde ifølge krav 3, hvor de pluripotente celler er humane pluripotente celler.
5. In v/fro-fremgangsmåde ifølge krav 3 eller 4, hvor de pluripotente celler er stamceller.
6. In v/'tro-fremgangsmåde ifølge krav 5, hvor stamcellerne er embryonale stamceller.
7. In v/fro-fremgangsmåde ifølge krav 3 eller 4, hvor de pluripotente celler er inducerede pluripotente celler (IPS).
8. Kit til dyrkning af celler omfattende: a) noggin eller fragmenter deraf, der inhiberer BMP-signalvejen og b) en inhibitor af Transforming Growth Factor (TGF)/aktivin/nodal-signalvejen valgt fra gruppen bestående af SB431542, Lefty-A og Cerberus og fragmenter af Lefty-A og Cerberus, der inhiberer TGF/aktivin/nodal-signalvejen.
9. Kit til dyrkning af celler ifølge krav 8, yderligere omfattende en mediumbase.
10. In v/'fro-fremgangsmåde ifølge et hvilket som helst af kravene 3-7, hvor de pluripotente celler dyrkes med mediet som defineret i krav 1 eller 2 i mindst 5 dage.
11. In Wfro-fremgangsmåde ifølge et hvilket som helst af kravene 3-7, hvor de pluripotente celler dyrkes med mediet som defineret i krav 1 eller 2 i mindst 7 dage.
12. In vitro-fremgangsmåde ifølge et hvilket som helst af kravene 3-7, hvor de pluripotente celler dyrkes med mediet som defineret i krav 1 eller 2 i mindst 10 dage.
