Claims (16)
- FILAMENTØSE SVAMPE MED REDUCERET INDHOLD AF UDP-GALACTOFURANOSE PATENTKRAV1. Rekombinant filamentøs svampecelle omfattende et nukleært genom omfattende et inaktiveret UDP-galactopyranosemutase- (UDP-gal/vmutase) gen, hvor svampecellen endvidere omfatter en rekombinant nukleinsyre til ekspression af et sekreteret protein i cellen.
- 2. Svampecelle ifølge krav 1, hvor UDP-galP-mutasegenet indeholder en deletion, en insertion eller en omfordeling.
- 3. Svampecelle ifølge krav 1, hvor svampecellen er en Aspergillus-celle.
- 4. Svampecelle ifølge krav 1, hvor svampecellen er en Aspergillus m'ger-celle eller en Aspergillus oryzae-celle.
- 5. Svampecelle ifølge krav 1, hvor det sekreterede protein er glycosyleret protein.
- 6. Svampecelle ifølge krav 1, hvor det sekreterede protein er heterologt med cellen.
- 7. Svampecelle ifølge krav 1, hvor det sekreterede protein er nativt i forhold til cellen.
- 8. Svampecelle ifølge krav 1, hvor det sekreterede protein er et terapeutisk protein, et antistofprotein eller et enzym.
- 9. Svampecelle ifølge krav 8, hvor enzymet er en amylase, protease, cellulase, xylanase eller phytase.
- 10. Svampecelle ifølge krav 1, hvor det nukleære genom omfatter mindst to inaktiverede UDP-galactopyranosemutasegener.
- 11. Svampecelle ifølge krav 1, hvor svampecellen endvidere omfatter en rekombinant nukleinsyre til ekspression af en mannosidase.
- 12. Fremgangsmåde til produktion af sekreret protein kodet for af den rekombinante nukleinsyre, hvilken fremgangsmåde omfatter dyrkning af cellen ifølge et hvilket som helst af kravene 1 til 11 i et dyrkningsmedium under forhold, der er egnede til at producere det sekreterede protein.
- 13. Fremgangsmåde ifølge krav 12, hvilken fremgangsmåde endvidere omfatter genvinding af det sekreterede protein fra dyrkningsmediet.
- 14. Fremgangsmåde til frembringelse af cellen ifølge 1, hvilken fremgangsmåde omfatter a) indføring af en rekombinant nukleinsyre i en filamentøs svampecelle, således at den rekombinante nukleinsyre rekombineres med UDP-galactopyranosemutasegenet af cellens genom, hvorved genet inaktiveres; og b) indføring af en anden rekombinant nukleinsyre i cellen, der tilvejebringer ekspression af det sekreterede protein.
- 15. Fremgangsmåde ifølge krav 14, hvor nukleinsyren indføres i UDP-galactopyranosemutasegenet.
- 16. Fremgangsmåde ifølge krav 14, hvor nukleinsyren deleterer mindst en del af UDP-galactopyranosemutasegenet.
