Den foreliggende opfindelse angår hidtil ukendte antigeniske peptider indeholdende 12-16 aminosyrerester, hvilke peptider er ejendommelige ved, at de har en af de i krav 1 angivne formler.
Det har været kendt i mange år, at pattedyrspermatozoer er 05 antigeniske. Senere er det blevet vist, at pattedyrsperma indeholder et antigenisk enzym kendt som C^-isozymet af lactatdehydroge-nase (LDH-X, LDH-C^). LDH-C^ er blevet isoleret i ren krystallinsk form fra musetestis, Goldberg (1972), J. Biol. Chem., 247:2044-2048. Enzymet har en molekylvægt pi 140.000 og er sam-10 mensat af fire identiske C-underenheder. Aminosyresekvensen og den tredimensionelle struktur af LDH-C^ er blevet studeret og delvis bestemt af flere forskere, se Musick et al. (1976), J. Mol.
Biol., 104:659-668, og Wheat et al. (1977), Biochem. & Biophys.
Res. Comm., 74, Nr. 3:1066-1077. Wheat et al. bestemte sekvensen 15 af det essentielle thiolpeptid fra aminosyre 159 til 171 og fandt, at dette var næsten identisk med essentielle thiolpeptider fra andre hvirveldyr-LDH-isozymer.
I 1974 gav Dr. Erwin Goldberg en redegørelse for virkningerne af immunisering med LDH-X (LDH-C^) på fertilitet, og fremsatte 20 den mulighed, at det "ved anvendelse af en defineret makromoleky-lær sperma komponent bliver muligt at klarlægge dens primære struktur i form af aminosyresekvens, specifikt at kortlægge den eller de antigeniske determinanter, der er ansvarlige for fremkaldelse af infertilitet, og derefter konstruere syntetiske peptider indeholdende 25 disse determinanter. Når man kan syntetisere et molekyle med sådanne egenskaber, muliggøres fertilitetskontrol ad immunologisk vej", Karolinska Symposia on Research Methods in Reproductive Endocrinology, 7th Symposia: Immunological Approaches to Fertility Control, Geneve, 1974, 202-222. Sådanne syntetiske antigeniske 30 peptider vedblev imidlertid at være et mål og ikke et resultat, selv om deres teoretiske ønskelighed var erkendt. I 1979 summerede Dr.
Erwin Goldberg teknikkens standpunkt som følger: "Som konklusion kræver immunoterapi til fødselskontrol i praksis mere end effektivitet, specificitet, reversibilitet og 35 fravær af systemisk bireaktion. Temmelig store mængder af an tigenet må være tilgængelige i utvetydigt ren form. Denne betingelse kan muligvis ikke opfyldes med et naturproduktenzym- 2 150205 antigen fra sperma eller testis. Antikonceptionel teknologi kræver snarere et syntetiserbart peptidfragment, der bibeholder antigenicitet og fremkalder et respons, der svækker fertilitet. Afslutning af de strukturelle analyser af LDH-C^ skulle mulig-05 gøre kortlægning af antigeniske determinanter og syntese af sådanne peptider til anvendelse ved en ny antikonceptionel teknologi", "Recent advances in Reproduction and Regulation of Fertility", G.P. 10 Det har nu vist sig, at stærkt antigeniske peptider kan frem stilles ved syntetisering af lineære sekvenser af aminosyrer, hvilke sekvenser menes at svare til aminosyresekvenser, der forefindes i det naturlige enzym LDH-C^. Disse sekvenser indbefatter: glutamin-syre-glutamin-leucin-isoleucin-glutamin-asparagin-leucin-valin-prolin-15 glutaminsyre-asparaginsyre-lysin, der menes at svare til LDH-C^ aminosyrerne 5 til 16. Denne sekvens svarer dog ikke til den publicerede foreløbige kortlægning af LDH-C^, se Musick et al. (25. august 1979), J. Biol. Chem., 254, nr. 16:7621-7623.

I de foregående formler betegner bogstavet "N" N-terminal 35 aminosyren, mens bogstavet "C" betegner C-terminal aminosyren.
Glu, Gin, Leu, Ile, Asn, Val, Pro, Asp, Lys, Ser, Arg og Cys repræsenterer henholdsvis L-aminosyreformerne af glutaminsyre, glu- 3 150205 tarnin, leucin, isoleucin, asparagin, valin, prolin, asparaginsyre, lysin, serin, arginin og cystein. Som man vil bemærke, indbefatter forbindelserne (a) til (h) den sekvens af 12 aminosyrer, begyndende med glutaminsyre og sluttende med lysin, der menes at svare til 05 aminosyrerne 5 til 16 i LDH-C^. De yderligere aminosyrer i forbindelserne (b), (c) og (d) menes at svare til de næste aminosyrer i LDH-C^, dvs. nr. 17, 18 og 19. Forbindelse (d) vil derfor indeholde aminosyrerne i sekvensen 5 til 19. Forbindelserne (e) til (h) indbefatter de samme antigeniske sekvenser som henholdsvis forbin-10 delserne (a) til (d), men cystein er tilføjet ved N-terminalenden for at lette disse forbindelsers kobling til proteiner ved fremstilling af antigeniske vacciner.
Peptidforbindelserne ifølge opfindelsen kan syntetiseres ud fra de aminosyrer, hvoraf de er opbygget. Syntesen kan for eksempel 15 udføres ved Merrifield fastfase-metoden som beskrevet i J.A.C.S. 85:2149-2154 (1963). Denne fastfase-metode til syntetisering af ami-nosyresekvenser er også beskrevet i Stewart og Young, Solid Phase Peptide Synthesis (W.H. Freeman & Co., San Francisco, 1969), pp.
1-4. Ved denne fremgangsmåde fastgøres C-terminal-aminosyren, så-20 som lysin, ved fremstilling af forbindelserne (a) og (e), eller leucin ved fremstilling af forbindelserne (b) og (f), til chlormethylerede polystyren-divinylbenzencopolymer-perler. Derefter tilføjes hver efterfølgende aminosyre med passende beskyttelsesgrupper sekventielt til den voksende kæde. Den a-amino-beskyttende gruppe kan for 25 eksempel, som beskrevet i Merryfield-artiklen, være en carbobenz-oxygruppe. Ved hjælp af kobling, afbeskyttelse og kobling af den næste aminosyre kan den ønskede aminosyresekvens og kædelængde frembringes. Som et afsluttende trin fjernes den beskyttende gruppe fra N-terminal-aminosyren og eventuelle sidekædebeskyttelses-30 grupper, og C-terminal-aminosyren spaltes fra harpiksen under anvendelse af et passende reagens, såsom trifluoreddikesyre og hy-drogenbromid. De ovenfor beskrevne forbindelser (a) til (h) fremstilles ved denne syntesemetode til brug ved reduktion af pattedyrs fertilitet.
35 For at udnytte antigeniske peptider ifølge opfindelsen i fertili tetsreducerende vacciner, bringes peptidet i forbindelse med et bærermolekyle, som fortrinsvis er et protein, der selv fremkalder et 4 150205 antigenisk respons og som kan administreres sikkert. For eksempel kan peptidet kobles til tetanustoxoid til administrering ved intra-muskulær injektion. For eksempel giver en blanding af 1 μ moJ tetanustoxoid, 60 μ mol antigenisk peptid og 18 millimol 1-ethyl-3-(3-05 dimethylaminopropyl)carbodiimid,hydrochlorid omsat i vand (pH 6) i 12 timer ved stuetemperatur og 24 timer ved 4° et produkt, der indeholder 3,5 mol peptid/mol tetanustoxoid. Overskud af reaktanter kan fjernes ved dialyse eller gelfiltrering, se Pique et al., fmmuno-chemistry, 15:55-60 (1978). Alternativt kan peptidet kobles under 10 anvendelse af bisdiazoteret benzidin (Bassiri et al., Endocrinology, 90:722 (1972)) eller glutaraldehyd.
Til intramuskulær injektion kan det koblede peptid suspenderes i en steril isotonisk salineopløsning eller anden konventionel bærer og et adjuvans kan om ønsket tilsættes. En foretrukken anvendelse 15 af en sådan vaccine er til administrering til kvinder. Der vil dannes antistoffer, som vil vise sig i ovidukt-fluiderne, hvorved der opnås en signifikant fertilitetsreduktion. Til dette formål vil mængden, der skal administreres, variere fra ca. 1 til 10 mg af det antigeniske peptid.
20 Peptidforbindelserne ifølge opfindelsen, deres fremstilling og deres antigeniske virkning illustreres nærmere i de efterfølgende eksempler.
Eksempel 1 25 Fremstilling af lineært peptid
Cys-Glu-Gln-Leu-lle-GIn-Asn-Leu-Val-Pro-Glu-Asp-Lys
Syntese af ovenstående peptid, heri omtalt som forbindelse (e), kan udføres ved anvendelse af fastfase-teknikker, der nu er velkendte inden for området. Ved en foretrukken fremgangsmåde 30 kobles aminobeskyttet lysin, der repræsenterer -COOH terminalgruppen i ovenstående peptid, til en konventionel fastfase-peptid-synteseharpiks, såsom chlormethylpolystyren, der er tværbundet med 1 til 2% divinylbenzen. Den aminobeskyttende gruppe fjernes si selektivt under anvendelse af et passende reagens, hvis art vil af-35 hænge af den anvendte beskyttende gruppe. I den foretrukne udførelsesform anvendes t-butyioxycarbonylgruppen (Boc) til amino-gruppebeskyttelse, og 40% trifluoreddikesyre i methylenchlorid er 5 150205 det selektive, afbeskyttelsesmlddel.
Efter afbeskyttelse behandles lysin-harpiksen med beskyttet asparaginsyre, fortrinsvis N-Boc-O-benzylasparaginsyre, og dicyc-lohexylcarbodiimid pi i og for sig kendt måde til dannelse af en 05 peptidbinding mellem lysinrestens fri amino-gruppe og den beskyttede asparaginsyres carboxylgruppe.
Denne cyklus med afbeskyttelse og kobling med aminosyrederi-vater og dicyclohexylcarbodiimid gentages så med de resterende aminosyrer i sekvensrækkefølgen i ovennævnte peptid. Nogle af 10 aminosyrerne kræver sidekædebeskyttelsesgrupper ud over alpha- aminobeskyttelsen. Sådanne aminosyrer og blokeringsgrupperne er som følger:
Cys(MBzl), Glu(oBzl), Asp(oBzl), Lys(CI-z), hvori oBzl er benzyl, Cl-z er o-chlorbenzyloxycarbonyl og MBzl er 15 methoxybenzyl.
Fuldendelse af syntesen gav følgende peptid koblet til styrendi vinylbenzencopolymerharpiksen: TFA-Cys(MBzl)-Glu(oBzl)-Gln-Leu-lle-Gln-Asn-Leu-Val-Pro-
Glu(oBzl)-Asp(oBzl)-Lys(CI-z)-harpiks 20 Frakobling af peptidet fra harpiksen opnås ved behandling med flydende hydrogenfluorid under ledsagende fraspaltning af alle beskyttende grupper til frembringelse af det ønskede peptid.
Fastfase-syntese af N-Boc-Lys(CI-z)-harpiks 25 Knytning af N-Boc-Lys(CI-z) til chlormethylharpiks udførtes ved cæsiumsaltmetoden. En prøve af chlormethylharpiks (200 g) indeholdende 0,74 mmol chlorid pr gram behandles med cæsiumsaltet af Boc-Lys-(CI-z) hidrørende fra neutralisation af Boc-Lysin med cæ-siumcarbonat. Ca. 73,8 g Boc-Lysin opløses i 80% methanol og 20% 30 vand og indstilles til pH 7,0 med ca. 29 g cæsiumcarbonat. Den resulterende opløsning tørres i en rotationsinddamper, hvorefter den tørres yderligere tre gange efter tre tilsætninger af 100 ml xylen.
Til cæsiumsaltet af Boc-Lys(CI-z) sættes 200 g chlormethylharpiks som ovenfor og tilstrækkeligt 1-methyl-2-pyrrolidinon til at bringe 35 det samlede volumen på ca. 1,90 liter. Den resulterende blanding omrøres ved 55°C i 48 timer. Harpiksen vaskes så ekstensivt med methanol efterfulgt af vand og derefter igen med methanol. Harpik- 6 150205 sen lufttørres og tørres si under vakuum. Efter spaltning af lysln fra harpiks med HF gav aminosyreanalyse en top svarende til 0,36 mmol/g.
En prøve af den netop beskrevne harpiks (6,0 g) underkaste-05 des følgende synteseprogram: (1) vask med tre 100 ml portioner methylenchlorid, (2) fjernelse af Boc-gruppen med 40% TFA i methylenchlorid under et minuts vask og en 20 minutters reaktionstid, (3) vask med tre 100 ml portioner methylenchlorid, (4) vask med to 100 ml portioner isopropa-10 nol, (5) vask med tre 100 ml portioner methylenchlorid, (6) vask i et minut og neutralisation i 10 minutter med 100 ml portioner af 10% triethylamin i methylenchlorid, (7) vask med tre portioner af 100 ml methylenchlorid, (8) tilsætning af 2,5 ækvivalenter (7,2 mmol) Boc-aminosyre og 2,5 ækvivalenter (7,2 mmol) dicyclohexylcarbodiimid i 15 methylenchlorid og rystning i 2 timer, (9) vask med tre 100 ml portioner methylenchlorid, (10) vask med to 100 ml portioner isopropa-nol, (11) vask med tre 100 ml portioner methylenchlorid. Ovenstående cyklus gentoges for følgende N-beskyttede aminosyrer:

Boc-Asn Boc-Cys(MBzl) 25 Den beskyttede peptidharpiks underkastedes afbeskyttelse, hvilket gav TFA-saltet af den afbeskyttede peptidharpiks. Den tørrede harpiks (5,88 g) omrørtes i nærværelse af 6 ml anisol og 60 ml flydende HF ved 0°C i 1 time. HF fjernedes ved vakuum, og den olieagtige remanens vaskedes med to 50 ml portioner ethylether.
30 Peptidet ekstraheredes fra harpiksen med tre 50 ml portioner 1 molær eddikesyre, og de kombinerede filtrater lyofiliseredes, hvilket gav 2,27 g råt peptid. Dette rensedes ved 250 overførsler i et modstrømsfordelingsapparat. Opløsningsmiddelsystemet til ovennævnte fraktionering var butanol:eddikesyre:vand i forholdene 4:1:5.
35 Noget rensede fraktioner fra modstrømsfordelingsapparatet ren- sedes yderligere ved søjlechromatografi på diethylaminoethylcellulose med en linær gradient på 0,01 til 0,5 molær ammoniumbicarbonat, 150205 7 hvilket gav 356 mg rent peptid.
Aminosyreanalyse af det rene peptid efter sur hydrolyse gav:
Lys-,,02' ammoniak^, Asp^g, Glu^, PrOggg, Cysggg,
Valg 97, Neg 7g, Leu1 85· Dette peptid gav en enkelt plet med en 05 Rf p| 0,89 ved cellulosetyndtlagschromatografi med et opløsningsmiddelsystem af butanol:ethylacetat:eddikesyre:vand i forholdene 1:1:1:1.
Forbindelserne (a) til (d) og (f) til (h), der indeholder samme antigeniske sekvenser som forbindelse (e) fremstilles og karakteri-10 seres pi tilsvarende måde.
Forsøgsresultater
Rensede peptider indbefattende lineære aminosyresekvenser fra muse LDH-C^ fremstilledes pi følgende måde: 15 Ren muse LDH-C4 reduceres og carboxymethyleres med iod- eddikesyre. Nedbrydning med trypsin (4 vægt/vægtprocent) forløber i 4 timer i nærvær af 2M urinstof. Efter afsaltning på "Sepha-dex G-10" fraktioneres nedbrydningsproduktet på en "pBondapak C_jgu søjle (3,9 mm x 30 cm; Waters Associates) med en voksende 20 acetonitrilgradient. Trifluoreddikesyre (0,04%) forefindes under hele gradienten. Søjleeffluenten overvåges ved 214 nm, og fraktioner opsamles manuelt på grundlag af absorbanstoppe. Fraktioner tørres under en nitrogenstrøm og lyofiliseres for vand. Renhed vurderes isokratisk i samme chromatografiske system, og peptider renses igen 25 efter behov. Efter hydrolyse (6N HCI, 107°, 40 timer) bestemmes aminosyresammensætninger ved reversfasechromatografi af o-pthalal-dehydderivater. Se Hili et al. (1979), Anal. Chem. 41:1338-1341. Aminosyresekvenser blev bestemt ved manuel Edman-nedbrydning.

Et af de fraskilte rensede peptider betegnedes Peptid G-5 og havde en aminosyresekvens svarende til aminosyrerne 5-15 i nativ muse LDH-C^. Peptid G-5 var sammensat af følgende aminosyrer (fra N-terminal til C-terminal): Glu-GIn-Leu-lle-GIn-Asn-Leu-Val-35 Pro-Glu-Asp.
Antistofbinding med peptid G-5 blev bestemt ved en fast ma-trixradioimmunoanalyseprocedure som beskrevet af Pierre et al.
8 150205 (1976), J. Exp, Med. 144:1254-1262. Peptidet blev anbragt som belægning på væggene af en polyvinylchloridmikrotiterplade ved inkubation natten over ved 4°C af en opløsning indeholdende 5 nmol peptid i 100 μΙ 0,04M NaPO^, 0,14M NaCl (PBS) i hver fordybning.
05 Hver fordybning vaskedes med 200 μΙ/fordybning 10% hesteserum i PBS og inkuberedes i samme opløsning i 1 time. Efter vask inkuberedes pladen med 50 μΐ af gammaglobulinfraktionen af sammenblandede kaninantimuse LDH-C. sera. Efter 4 timers inkubation vaskeri 125 des pladen og inkuberedes derefter med 100 μΙ/fordybning I- ”*0 gedeantikaningammaglobulin ved 4°C i 16 timer. Efter omfattende vask bestemtes bindingsradioaktivitet under anvendelse af en gammatæller.
Det bestemte bindingsaktivitetsniveau for peptid G-5 er anført i nedenstående tabel A.

De foranstående data viser, at peptid G-5 har højt antigenici-tetsniveau, som svarer til tilsvarende antigeniske områder i nativ LDH-C4· Den immunogene evne af peptid G-5 påvistes også ved 20 konjugering af peptidet med okseserumalbumin som bærer og immunisering af kaniner med konjugaterne til frembringelse af et anti-LDH-C4 serum. Dette antiserum fandtes at være stærkt aktivt overfor nativ LDH-C4· Hver kanin injiceret med G-5-konjugatet frembragte antistoffer, som var specifikke for nativ LDH-C4· 35 '
