Abstract

A pharmaceutical preparation for treating peptic ulcer comprises pepstatin and a histamine H2 receptor antagonist of the formula <IMAGE> in which R1 and R2, which can be identical or different, denote hydrogen or (lower)alkyl, and where R2 can also, when R1 denotes hydrogen, be allyl, propargyl, cyclo(lower)alkyl(lower)alkyl, cyclo(lower)alkyl, cyano(lower)alkyl, 2-fluoromethyl, 2,2,2-trifluoroethyl, hydroxyl, 2,3-dihydroxypropyl, <IMAGE> in which p is an integer from 1 to 6 inclusive, q is an integer from 1 to 6 inclusive, R3 and R4, which can be identical or different, denote hydrogen, (lower)alkyl, hydroxyl, (lower)alkoxy or halogen, and where R4 can also, when R3 denotes hydrogen, be trifluoromethyl, or R3 and R4 together denote methylenedioxy, R5 denotes hydrogen, m is an integer from 0 to 2 inclusive, n is an integer from 2 to 5 inclusive, Z is sulphur, oxygen or methylene, and A denotes <IMAGE> in which R6 denotes hydrogen, (lower)alkyl, (lower)alkoxy or halogen, r is an integer from 1 to 4 inclusive, and R8 and R9, which can be identical or different, denote hydrogen, (lower)alkyl, allyl, propargyl, (lower)alkoxy(lower)alkyl, in which the (lower)alkoxy moiety is at least two carbon atoms away from the nitrogen atom, cyclo(lower)alkyl or phenyl(lower)alkyl, with the proviso that R8 and R9 cannot both be cyclo(lower)alkyl or that R8 and R9, together with the nitrogen atom to which they are bonded, can be pyrrolidino, methylpyrrolidino, dimethylpyrrolidino, morpholino, thiomorpholino, piperidino, methylpiperidino, dimethylpiperidino, hydroxypiperidino, N- methylpiperazino, 1,2,3,6-tetrahydropyridyl, 3-pyrrolino, homopiperidino, heptamethyleneimino, octamethyleneimino or 3-azabicyclo[3.2.2]nonane, or a non-toxic, pharmaceutically acceptable acid addition salt, hydrate or solvate thereof, and also a pharmaceutically acceptable excipient. The simultaneous administration of the two constituents reduces the quantity of histamine H2 receptor antagonist which is required for treatment, with the tendency for side effects to occur being decreased.
