B. durch Extraktion, abgetrennt werden können. Dieses kann leicht mit wasserfreier Oxalsäure zu i4-Pregnen-3, 2i-diol-20-on-21-acetat dehydratisiert werden. Durch anschließende Oxydation mit Chromsäure in Essigsäure erhält man hieraus das bekannte i4-Pregnen-2i-ol-3, 20-dion-"21-acetat. Die Verbindung kann zur Herstellung von Absorptionsgrundlagen verwendet werden, die verbesserte Wasserabsorptions- und Weichmachungseigenschaften haben und in der Pharmazie und Kosmetik allein oder als Träger für bekannte Medikamente von Nutzen sind. Eine geeignete Absorptionsgrundlage kann man herstellen, indem man ein Gemisch aus 85°/0 farblosem Petroleum, Stearylalkohol und 5% I4a-Oxyprogesteron zusammenschmilzt und das Gemisch unter Rühren abkühlt, bis es erstarrt. ■ Die erhaltene Absorptionsgrundlage kann - leicht mit wäßrigem Material bei Raumtemperatur verrieben oder bei erhöhter Temperatur emulgiert werden und ergibt eine geschmeidige und beständige Wasser-in-Öl-Emulsion. Eng. Chem., Bd. 36, 1944, S. 504) die Sauerstoffaufnahme 6,3 bis 7 Millimol Na2SO3 je Stunde und Liter betrug. In diesem Nährboden, der eine 24 Stunden alte Kultur von Mucor griseo cyanus enthielt, wurden 5 g Progesteron in 250 ecm Aceton suspendiert. Nach einer unter den gleichen Temperatur- und Belüftungsbedingungen vorgenommenen weiteren Bebrütung von 24 Stunden wurden die Brühe und das Mycel extrahiert. Das Mycel wurde abfiltriert, zweimal mit je einem etwa dem Mycelvolumen entsprechenden Volumen Aceton gewaschen und zweimal mit etwa der gleichen Menge Methylenchlorid extrahiert. Die Lösungsmittel enthaltenden Aceton- und Methylenchloridextrakte wurden zu dem Filtrat der Brühe zugegeben. Das Gemisch aus den Extrakten und dem Filtrat wurde nacheinander zweimal mit je einem halben Volumen Methylenchlorid und zweimal mit je einem viertel Volumen Methylenchlorid extrahiert. Die vereinigten Methylenchloridextrakte wurden zweimal mit je einem zehntel Volumen einer 2°/oigen wäßrigen Natriumbicarbonatlösung und dann zweimal mit je einem zehntel Volumen Wasser gewaschen. Nach Trocknen der Methylenchloridextrakte mit etwa 3 bis 5 g wasserfreiem Natriumsulfat je Liter Lösungsmittel und Filtrieren wurde das Lösungsmittel durch Destillation entfernt, und es ergab sich ein kristalliner Rückstand von 4,815 g. Der kristalline Rückstand wurde mit einer Mischung aus Hexanen und Petroläther, bekannt unter dem Handelsnamen »Skellysolve-B-Petroläther«, verrieben. 14 a-Oxyprogesteron14 α-oxyprogesterone
Aus 20 g enzymatischem Lactalbuminextrakt, bekannt unter der Handelsbezeichnung »Edamin«·, 3 g Maiswasser und 50 g technisch reiner Dextrose, die mit Leitungswasser auf 11 verdünnt wurde, wurde ein Nährboden hergestellt und auf einen pH-Wert zwischen 4,3 und 4,5 eingestellt. 12 1 dieses sterilisierten Nährbodens wurden mit Helicostylum piriforme geimpft und 24 Stunden bei einer Temperatur von 280 bebrütet, wobei Belüftung und Rührung so vorgenommen wurden, daß nach dem Verfahren von Cooper, Fernstrom und Miller (Ind. Eng. Chim., Bd. 36, 1944, S. 504) die Sauerstoffaufnahme 6,3 bis 7 Millimol Na2SO3 je Stunde und Liter betrug. In diesem Nährboden, der eine 24 Stunden alte Kultur von Helicostylum piriforme enthielt, wurden 6 g Progesteron in 275 ecm Aceton suspendiert. Nach einer unter den gleichen Temperatur- und Belüftungsbedingungen vorgenommenen weiteren Bebrütung von 27 Stunden wurden die Brühe und das Mycel gemäß dem im Beispiel 1 beschriebenen Verfahren extrahiert. Es wurde ein kristalliner Rückstand von 8,313 g gewonnen, der in 600 ecm Benzol mit 10% Äther gelöst und über 300 g Tonerde (mit Salzsäure gewaschen und durch Erhitzung bei 1200 aktiviert) chromatographiert wurde. Das Sublimat wurde aus 3 ecm Aceton und Hexan umkristallisiert und ergab I4a-Oxyprogesteron mit einem Schmelzpunkt von 195 bis 200,5° und einer optischen Drehung [a]f? = + i88° (1,036 in Chloroform).Fraction 13 was dissolved in 20 ecm acetone and separated from an insoluble suspension by filtration. The clear solution was concentrated and diluted dropwise with ether. 585.5 mg of crystals having a melting point 182-194 0 were obtained. mg of this product were sublimed at 165 ° and a pressure of 0.05 mm.
